Tolerance of liver fluke infection varies between breeds and producers in beef cattle.

Liver flukes (Fasciola spp.) are important helminth parasites of livestock globally and cause substantial reductions in health and productivity of beef cattle. Attempts to control fluke have been thwarted by the difficulty of vaccine design, the evolution of flukicide resistance and the need to control the intermediate snail host. Mechanisms to reduce the impact of parasites on animal performance have typically focused on promoting host resistance - defined as the ability of the host to kill and remove the parasite from its system - and such strategies include improving protein nutrition or se

KNOX1 Transcription Factors in Plants with a Special Focus on Horticultural Crops: A Review.

Class I KNOX1 (KNOTTED1-like homeobox 1) transcription factors integrate gibberellin (GA), cytokinin (CK), and auxin (IAA) signaling to maintain shoot apical meristem identity and coordinate plant organogenesis. This review examines the structural conservation, evolutionary dynamics, and regulatory architecture of KNOX1 genes across horticultural crops, drawing essential mechanistic context from model species. We synthesize KNOX1 functions in six agronomic domains, including plant architecture and branching, leaf morphogenesis and ornamental traits, floral development and sex determination, fr

Evaluating Causal Claims in Plant Developmental Metabolism: A When-Where-How Framework.

Metabolic changes are often described as drivers of plant development even when the evidence establishes only association or a permissive requirement. This review separates biological role from causal status and evaluates claims across seven dimensions: association, necessity, localization, transport, rescue and mediation, sufficiency, and quantitative-threshold testing. We apply a when-where-how framework to four cases. Perturbations of T6P, SnRK1, and TOR support T6P-dependent kinase regulation as a mediator of lateral-root development and are consistent with a context-dependent candidate ga

Cloning and functional analysis of downy mildew resistance gene RPF1 in spinach (Spinacia oleracea L.).

Downy mildew, caused by the obligate pathogen Peronospora effusa, is a devastating disease in spinach (Spinacia oleracea L.). Resistance to P. effusa is largely controlled by a dominant resistance locus RPF1, for which two candidate genes have been previously proposed but lacked functional validation. In this study, we employed virus-induced gene silencing (VIGS) to demonstrate that silencing of Spo12903 significantly compromised resistance to P. effusa, thereby confirming Spo12903 as the functional gene underlying RPF1. Sequence characterization and comparative analyses revealed that RPF1 pos

Disease-induced changes in Panax ginseng phyllosphere fungal community assembly and functional adaptation.

INTRODUCTION: Phyllosphere microorganisms play essential roles in plant health and disease resistance, yet their responses to pathogen infections remain poorly understood. Panax ginseng is susceptible to multiple fungal diseases, which threaten its quality and yield. This study aimed to clarify the underlying disease resistance mechanisms of Panax ginseng by analyzing the phyllosphere fungal communities associated with fungal infections. METHODS: Phyllosphere fungal communities of healthy Panax ginseng plants and those with three fungal infections (gray mold, damping-off and root rot) were co

The PtARF6/PtARF8-PtGH3.1 transcriptional module regulates auxin homeostasis and dwarfism in citrus.

Dwarfing rootstocks are crucial for high-density and labor-efficient citrus cultivation, yet the molecular mechanisms underlying plant height regulation remain poorly understood. Using an integrated multiomics analysis of dwarf and normal-height red tangerine (Citrus reticulata) × trifoliate orange (Poncirus trifoliata) hybrid seedlings, we identified the Gretchen Hagen 3.1 gene PtGH3.1, an auxin-conjugating enzyme, as an important regulator of dwarfism. Compared with normal-height hybrid seedlings, PtGH3.1 was significantly upregulated in the shoot tips of dwarf hybrid seedlings, correlating

Loss-of-Function of a Tomato Receptor-Like Kinase Impairs Male Fertility and Induces Parthenocarpic Fruit Set.

Parthenocarpy arises when an ovary develops into fruit without pollination/fertilization. The mechanisms involved in genetic parthenocarpy have attracted attention because of their potential application in plant breeding and also for their elucidation of the mechanisms involved in early fruit development. We have isolated and characterized a novel small parthenocarpic fruit and flower (spff) mutant in the tomato (Solanum lycopersicum) cultivar Micro-Tom. This plant showed both vegetative and reproductive phenotypes including dwarfism of floral organs, male sterility, delayed flowering, altered…

Genetic Analyses of Mungbean [Vigna radiata (L.) Wilczek] Breeding Traits for Selecting Superior Genotype(s) Using Multivariate and Multi-Traits Indexing Approaches.

Mungbean [Vigna radiata (L.) Wilczek] is an important food, feed, and cash crop in rice-based agricultural ecosystems in Southeast Asia and other continents. It has the potential to enhance livelihoods due to its palatability, nutritional content, and digestibility. We evaluated 166 diverse mungbean genotypes in two seasons using multivariate and multi-traits index approaches to identify superior genotypes. The total Shannon diversity index (SDI) for qualitative traits ranged from moderate for terminal leaflet shape (0.592) to high for seed colour (1.279). The analysis of variances (ANOVA) ind

Genome-wide identification of the WRKY gene family in Codonopsis pilosula and functional analysis of CpWRKY8 under drought stress.

Codonopsis pilosula (Franch.) Nannf., a perennial herbaceous species in the family Campanulaceae, is widely used in traditional medicine for its dried roots. Drought stress severely constrains both the yield and the accumulation of bioactive compounds in this species. In this study, we performed a genome-wide identification and phylogenetic analysis of the WRKY TF (TF) family in C. pilosula, identifying 49 members that were classified into seven subfamilies. Among them, CpWRKY8, a member of the Group IIa subfamily, localized to the nucleus and showed transcriptional activation activity in yeas

Natural Variation of COLD and CATECHINS REGULATOR 1 Coordinately Fine-Tunes Cold Tolerance and Tea Quality in Tea Plants.

Cold tolerance and tea quality are essential for profitable tea cultivation, yet the genetic interplay between them remains elusive. Here, we systematically evaluated cold tolerance and quantified 12 key tea quality-related metabolites over two consecutive years of 108 geographically diverse tea accessions in China. Multi-trait genome-wide association mapping identified a central hub regulator, COLD AND CATECHINS REGULATOR 1 (CCR1), significantly associated with cold tolerance and catechins biosynthesis in tea plants. Functional analysis demonstrated that CsCCR1 positively regulates cold toler

Mapping of quantitative trait loci (QTL) in Brassica napus L. for tolerance to water stress.

Brassica napus L. plants are sensitive to water stress conditions throughout their life cycle from seed germination to seed setting. This study aims at identifying quantitative trait loci (QTL) linked to B. napus tolerance to water stress mimicked by applications of 10% polyethylene glycol-6000 (PEG-6000). Two doubled haploid populations, each consisting of 150 genotypes, were used for this research. Plants at the two true leaf stage of development were grown in the absence (control) or presence (stress) of PEG-6000 under controlled environmental conditions for 48 h, and the drought stress ind

A Genome-Wide Analysis of the CEP Gene Family in Cotton and a Functional Study of GhCEP46-D05 in Plant Development.

C-TERMINALLY ENCODED PEPTIDEs (CEPs) are a class of peptide hormones that have been shown in previous studies to play an important role in regulating the development and response to abiotic stress in model plants. However, their role in cotton is not well understood. In this study, we identified 54, 59, 34, and 35 CEP genes from Gossypium hirsutum (2n = 4x = 52, AD1), G. barbadense (AD2), G. arboreum (2n = 2X = 26, A2), and G. raimondii (2n = 2X = 26, D5), respectively. Sequence alignment and phylogenetic analyses indicate that cotton CEP proteins can be categorized into two subgroups based on

Physiological and Full-Length Transcriptome Analyses Reveal the Dwarfing Regulation in Trifoliate Orange (Poncirus trifoliata L.).

Dwarfing rootstocks are capable of high-density planting and are therefore urgently needed in the modern citrus cultivation system. However, little is known about the physiological relevance and molecular basis underlying citrus height. This study aimed to comprehensively analyze phytohormone, carbohydrate, and associated transcriptome changes in the stem of two weak growth rootstocks ('TO' and 'FD') relative to the vigorous 'CC' rootstock. The phenotypic observation revealed that the plant height, plant weight, and internode length were reduced in dwarfing rootstocks. Moreover, the contents o

A Lactuca universal hybridizer, and its use in creation of fertile interspecific somatic hybrids.

A Lactuca sativa cv. Ardente line heterozygous for a gene encoding resistance to kanamycin, a positive and dominant trait, was crossed with cv. Girelle, which is heterozygous for a recessive albinism marker. The resulting seeds yielded 25% albino seedlings, of which 50% were also resistant to kanamycin. Such plantlets (KR, a) grown in vitro were used for preparation of universal hybridizer protoplasts, since green buds that can develop on kanamycin containing-medium should result from fusion with any wild-type protoplast. To test the practicability of this selection scheme, we fused L. sativa

Traditional South African medicinal plants and their role in improving goat reproduction: a review.

INTRODUCTION: Goats are an integral part of the livelihoods of South Africans, particularly in the rural communities, yet reproductive inefficiencies (low libido, poor semen quality, uterine infections, postpartum disorders, parasitism, and nutrition gaps) limit their productivity. Most resource challenged farmers resort to the use of ethnoveterinary plants for reproductive health of their goats due to their safety and ease of access; however, there is still a lack of standardization on their safety, dosing and efficacy. AIM: This study aimed to review and document the South African tradition

Utilizing RNA-Seq-Derived SNPs for KASP Marker Development in Maize.

Maize is the third most significant cereal crop, and most of its agronomic traits are quantitative in nature. The fine mapping of genes or quantitative trait loci (QTLs) associated with these traits is essential for marker-assisted selection (MAS) in maize breeding. Although significant progress has been made, there remains a lack of high-density, highly polymorphic single-nucleotide polymorphism (SNP) markers, especially competitive allele-specific PCR (KASP) SNP markers suitable for automated genotyping. The development of next-generation sequencing (NGS) platforms has generated vast amounts

Marker-assisted pyramiding of lycopene-ε-cyclase, β-carotene hydroxylase1 and opaque2 genes for development of biofortified maize hybrids.

Malnutrition affects growth and development in humans and causes socio-economic losses. Normal maize is deficient in essential amino acids, lysine and tryptophan; and vitamin-A. Crop biofortification is a sustainable and economical approach to alleviate micronutrient malnutrition. We combined favorable alleles of crtRB1 and lcyE genes into opaque2 (o2)-based four inbreds viz. QLM11, QLM12, QLM13, and QLM14 using marker-assisted backcross breeding.
