Plasmopara viticola effector PvRxLR24 (a potential phosphatase) interferes grapevine stomatal immunity by targeting VvOST1.

Numerous pathogens invade plants through stomata, and plants, on the other hands, could close the stomata after sensing the pathogens PAMPs. Subsequently, pathogens could overcome this plant immune response with their phytotoxins and effectors. A previous study showed that Plasmopara viticola, a strictly obligate biotrophic oomycete, could inhibit the Vitis vinifera stomatal closure to promote infection. However, the underlying mechanism stimulating the stomata movement is still poorly understood. Here we demonstrate that PvRxLR24, an effector from P. viticola, functions analogously to a phosp

Actin depolymerization is able to increase plant resistance against pathogens via activation of salicylic acid signalling pathway.

The integrity of the actin cytoskeleton is essential for plant immune signalling. Consequently, it is generally assumed that actin disruption reduces plant resistance to pathogen attack. Here, we demonstrate that actin depolymerization induced a dramatic increase in salicylic acid (SA) levels in Arabidopsis thaliana. Transcriptomic analysis showed that the SA pathway was activated due to the action of isochorismate synthase (ICS). The effect was also confirmed in Brassica napus. This raises the question of whether actin depolymerization could, under particular conditions, lead to increased res

[Cowpea mosaic virus chimeric particles bearing ectodomain of matrix protein 2 (M2E) of influenza A virus: production and characteristics].

The epitope presentation system for ectodomain of M2-protein of influenza A virus (M2e) based on Cowpea Mosaic Virus (CPMV) was constructed for expression in plants Vigna unguiculata. CPMV is widely used as a vector for production of immunogenic chimeric virus particles (CVPs) bearing epitopes of different infectious human and animal pathogens. To produce chimeric CPMV virus particles in plants, two binary vectors were constructed bearing modified gene coding for S-coat protein of CPMV with insertions of M2e epitopes of human influenza and bird influenza viruses. Antigenic and immunogenic prop

Subversion of plant endomembrane trafficking by a Plasmodiophora brassicae effector PbALE that recruits membrane components at the host-pathogen interface to aid in colonization.

Plasmodiophora brassicae, an intracellular biotrophic protist that causes clubroot in crucifers, extensively alters the host endomembrane system for colonization. However, the molecular mechanisms and functions of effectors to achieve this remain poorly understood. Using fluorescent and transmission electron microscopy of P. brassicae-infected Arabidopsis, we show that host endomembranes and organelles become closely associated with P. brassicae secondary plasmodial structures and are subsequently phagocytosed by the pathogen. PbALE is a secreted endomembrane-targeting effector that binds a ra

Plant-made polio type 3 stabilized VLPs-a candidate synthetic polio vaccine.

Poliovirus (PV) is the causative agent of poliomyelitis, a crippling human disease known since antiquity. PV occurs in two distinct antigenic forms, D and C, of which only the D form elicits a robust neutralizing response. Developing a synthetically produced stabilized virus-like particle (sVLP)-based vaccine with D antigenicity, without the drawbacks of current vaccines, will be a major step towards the final eradication of poliovirus. Such a sVLP would retain the native antigenic conformation and the repetitive structure of the original virus particle, but lack infectious genomic material. I

An RLP23/Cf-9TM-IC Chimeric Receptor Enhances nlp24-Triggered Immunity and Resistance to Phytophthora nicotianae in Nicotiana benthamiana.

Receptor-like protein AtRLP23 from Arabidopsis thaliana perceives the conserved microbial peptide nlp24 derived from necrosis- and ethylene-inducing peptide 1-like proteins (NLPs), but immune outputs are often weak in heterologous systems. Here, we engineered RLP23/Cf-9 chimeric receptors by fusing the extracellular domain of AtRLP23 to either the Cf-9 intracellular region (IC) or the Cf-9 transmembrane-intracellular region (TM-IC), and assessed their activity in Nicotiana benthamiana. In transient expression assays, AtRLP23 alone did not trigger hypersensitive cell death after infiltration wi

The tomato cytoplasmic receptor-like kinase TPK09 mediates resistance to Botrytis cinerea and tolerance to light-induced stress.

Plant responses to pathogens often rely on receptor-like cytoplasmic kinases (RLCKs) that mediate signaling through interactions with receptor kinases and downstream components. Here, we studied the tomato RLCK, TPK1b Related Protein Kinase (TPK09), and demonstrate its function in integrating defense with light stress responses. Tomato tpk09 mutants exhibited increased susceptibility to Botrytis cinerea and the vascular pathogen Fusarium oxysporum, whereas transgenic expression of TPK09 enhanced resistance to Botrytis. Disease severity in tpk09 mutants was elevated under light-emitting diode (

Immune Priming Promotes Thermotolerance, Whereas Thermopriming Suppresses Systemic Acquired Resistance in Arabidopsis.

Heat stress and pathogens are two serious yield-limiting factors of crop plants. Plants that previously experienced high but sub-lethal temperatures become subsequently tolerant to higher temperatures through the development of acquired thermotolerance (ATT). ATT activation is associated with the elevated expression of heat shock (HS)-related genes such as HSFA2, HSFA3, and HSP101. Similarly, through the development of systemic acquired resistance (SAR), previously experienced plants achieve a higher resistance than naïve plants. SAR activation requires mobile signals and primarily depends on

The novel elicitor AsES triggers a defense response against Botrytis cinerea in Arabidopsis thaliana.

AsES (Acremonium strictum Elicitor and Subtilisin) is a novel extracellular elicitor protein produced by the avirulent isolate SS71 of the opportunist strawberry fungal pathogen A. strictum. Here we describe the activity of AsES in the plant-pathogen system Arabidopsis thaliana-Botrytis cinerea. We show that AsES renders A. thaliana plants resistant to the necrotrophic pathogen B. cinerea, both locally and systemically and the defense response observed is dose-dependent. Systemic, but not local resistance is dependent on the length of exposure to AsES. The germination of the spores in vitro wa

CASAR82A, a pathogen-induced pepper SAR8.2, exhibits an antifungal activity and its overexpression enhances disease resistance and stress tolerance.

Pepper SAR8.2 gene (CASAR82A) was previously reported to be locally or systemically induced in pepper plants by biotic and abiotic stresses. In this study, the physiological and molecular functions of the pepper SAR8.2 protein in the plant defense responses were investigated by generating Arabidopsis transgenic lines overexpressing the CASAR82A gene. The transgenic Arabidopsis plants grew faster than the wild-type plants, indicating that the CASAR82A gene was involved in plant development. The ectopic expression of CASAR82A in Arabidopsis was accompanied by the expression of the Arabidopsis pa

The Circadian Clock Regulates Receptor-Mediated Immune Responses to a Herbivore-Associated Molecular Pattern.

Plants activate induced defences through the recognition of molecular patterns. Like pathogen-associated molecular patterns, herbivore-associated molecular patterns (HAMPs) can be recognised by cell surface pattern recognition receptors, leading to defensive transcriptional changes in host plants. Herbivore-induced defensive outputs are regulated by the circadian clock, but the underlying molecular mechanisms remain unknown. To investigate how the plant circadian clock regulates transcriptional reprogramming of a specific HAMP-induced pathway, we characterised the daytime and nighttime transcr

iTRAQ analysis of the tobacco leaf proteome reveals that RNA-directed DNA methylation (RdDM) has important roles in defense against geminivirus-betasatellite infection.

UNLABELLED: Geminiviruses have caused serious losses in crop production. To investigate the mechanisms underlying host defenses against geminiviruses, an isobaric tags for relative and absolute quantification (iTRAQ)-based quantitative proteomic approach was used to explore the expression profiles of proteins in Nicotiana benthamiana (N. benthamiana) leaves in response to tomato yellow leaf curl China virus (TYLCCNV) with its betasatellite (TYLCCNB) at an early phase. In total, 4155 proteins were identified and 272 proteins were changed differentially in response to TYLCCNV/TYLCCNB infection.

A salicylic acid-induced lectin-like protein plays a positive role in the effector-triggered immunity response of Arabidopsis thaliana to Pseudomonas syringae Avr-Rpm1.

Salicylic acid (SA) is one of the key hormones that orchestrate the pathogen-induced immune response in plants. This response is often characterized by the activation of a local hypersensitive reaction involving programmed cell death, which constrains proliferation of biotrophic pathogens. Here, we report the identification and functional characterization of an SA-induced legume lectin-like protein 1 (SAI-LLP1), which is coded by a gene that belongs to the group of early SA-activated Arabidopsis genes. SAI-LLP1 expression is induced upon inoculation with avirulent strains of Pseudomonas syring

Non-host defense response in a novel Arabidopsis-Xanthomonas citri subsp. citri pathosystem.

Citrus canker, caused by Xanthomonas citri subsp. citri (Xcc), is one of the most destructive diseases of citrus. Progress of breeding citrus canker-resistant varieties is modest due to limited resistant germplasm resources and lack of candidate genes for genetic manipulation. The objective of this study is to establish a novel heterologous pathosystem between Xcc and the well-established model plant Arabidopsis thaliana for defense mechanism dissection and resistance gene identification. Our results indicate that Xcc bacteria neither grow nor decline in Arabidopsis, but induce multiple defens

Systemic acquired resistance in Arabidopsis requires salicylic acid but not ethylene.

Systemic acquired resistance (SAR) is an inducible plant response to infection by a necrotizing pathogen. In the induced plant, SAR provides broad-spectrum protection against not only the inducing pathogen, but also against other, unrelated pathogens. Both salicylic acid (SA) and SAR-gene expression have been implicated as playing important roles in the initiation and maintenance of SAR. Here, we describe the characterization of transgenic Arabidopsis plants that express the bacterial nahG gene encoding salicylate hydroxylase, an enzyme that can metabolize SA. Strong, constitutive expression o

Mechanistic Aspects of Medicinal Plants and Secondary Metabolites against Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2).

BACKGROUND AND OBJECTIVE: Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2), a highly pathogenic virus, is responsible for a respiratory disease termed coronavirus disease 2019 (COVID-19). SARS-CoV-2 genome encodes various structural and non-structural proteins, which are necessary for viral entry and replication. Among these proteins, papain-like protease (PLpro), 3C-like protease (3CLpro), RNAdependent RNA polymerase (RdRp), helicase, serine protease, and spike protein are potential targets of herbal remedies and phytocompounds for inhibition of viral infection and replication. Gut microbiota-derived butyrate mediates the anticolitic effect of indigo supplementation through regulating CD4+ T cell differentiation.

This study explored the effect of plant-derived indigo supplementation on intestinal inflammation using in vivo, in vitro, and clinical sample analyses. Our results showed that indigo decreased mucosal inflammation by regulating CD4+ T cell differentiation in a gut microbiota-dependent manner. Microbes transferred from indigo-treated mice, indigo-induced enrichment of Roseburia intestinalis, and its metabolite butyrate played a role in Th17/Treg immunity similar to that of indigo in intestinal inflammation, which was involved in mTORC1/HIF-1α signal-mediated reprogrammed glucose metabolism. cGAS-STING pathway as a promising target for digestive diseases: insights from natural plant products.

Digestive diseases remain a major challenge to public health systems globally. Cyclic GMP-AMP synthase (cGAS) and stimulator of interferon gene (STING) play important roles in innate immunity as well as inflammatory responses. Dysregulation of the cGAS-STING pathway has been demonstrated to be an important pathogenetic mechanism in diverse gastrointestinal diseases. Therefore, targeting the cGAS-STING pathway is a potential therapeutic strategy for digestive diseases. Encouragingly, increasing studies have revealed that natural plant products are promising candidates for the treatment of diges
