Simulation-based Training in Ectopic Pregnancy and Salpingostomy.

Objective Ectopic pregnancy leads to approximately 3% of deaths in pregnancy. Surgical management is indicated when patients are hemodynamically unstable or have signs of a ruptured ectopic pregnancy. Salpingectomy is more commonly performed, but salpingostomy is preferred in a patient with prior salpingectomy with a desire for future pregnancy. Due to the lack of exposure, salpingostomy is not frequently performed and most residents do not feel adequately trained. Our goal was to provide a hands-on simulation about ectopic pregnancy and salpingostomy in hopes that the simulation will improve

Attitudes Toward Hysterectomy in Saudi Arabian Women Undergoing Evaluation for Uterovaginal Prolapse: A Cross-Sectional Study.

BACKGROUND: Hysterectomy is an effective management approach for uterovaginal prolapse. However, the decision to undergo hysterectomy is a complex matter, influenced not only by medical factors but also cultural beliefs, societal norms, and individual attitudes. In Saudi Arabia, a nation with its distinctive cultural and social norms, the understanding of women's attitudes toward hysterectomy is of utmost importance. Unfortunately, such related attitudes have not been explored. OBJECTIVE: This first-ever study aimed to investigate the attitudes toward hysterectomy among Saudi Arabian women un

Vaginal Pessaries for Pelvic Organ Prolapse or Stress Urinary Incontinence: A Health Technology Assessment.

BACKGROUND: Pelvic organ prolapse (POP) is the downward descent of the female pelvic organs into or through the vagina. The symptom that most strongly correlates with and is most specific for POP is a feeling of vaginal bulging. Stress urinary incontinence (SUI) is an involuntary loss of urine upon physical exertion or sneezing or coughing. Conservative (non-surgical) treatment options for both conditions include vaginal pessaries. We conducted a health technology assessment of vaginal pessaries for the treatment of POP and SUI, which included an evaluation of effectiveness, safety, cost-effec

Molecular Insights into the Protective Activity of Bioactive Purpurin against UV-Induced DNA Damage: Elucidation of the Binding Mechanism Using Spectroscopic and Computational Studies.

DNA, the fundamental genetic material in living organisms, is of great scientific interest due to its biological role and interactions with small molecules. Ultraviolet (UV) radiation poses a substantial environmental risk by inducing DNA damage, potentially leading to mutagenesis and skin cancers. The study investigated purpurin interaction with ct-DNA and its protective effect against UV-induced DNA damage. UV exposure caused significant DNA damage, indicated by 36% reduced absorbance; however, purpurin significantly lessened this damage to only 19%. In addition, Raman spectra showed no sign

Dual Regulatory Functions of Classical Zinc Finger Clusters from Myeloid Zinc Finger-1.

Zinc finger (ZF) proteins constitute a large family of transcriptional regulators that control gene expression through sequence-specific DNA recognition stabilized by Zn2+-coordination. Myeloid zinc finger-1 (MZF1) is a classical ZF protein associated with diverse gene regulation, although the biochemical basis of its promoter recognition remains unclear. MZF1 contains 13 CX2CX12HX3H-type ZF domains organized into two discrete clusters, MZF1(ZF1-4) and MZF1(ZF5-13), suggesting the modular regulation of distinct binding partners. In this study, we investigate the metal-binding properties and pr

Molecular characterization and differential regulation of sex steroids on the gonadotropin ligand-receptor system in mandarin fish (Siniperca chuatsi).

The gonadotropin system plays a fundamental role in regulating teleost reproduction. This study characterizes the molecular properties and physiological functions of the Gth system in the mandarin fish (Siniperca chuatsi). Sequence analysis reveals that both fshb and lhb subunits possess a single N-glycosylation site. Phylogenetic evidence indicates that the lhcgr duplication occurred prior to the teleost specific whole genome duplication event. Tissue distribution analysis demonstrates that lhcgr2 is strictly gonad-specific, while lhcgr1 exhibits a broad distribution across multiple organs, i

Derivation and characterization of an embryonic‑derived muscle progenitor cell line from Atlantic salmon (Salmo salar).

Unlike mammals, teleost fish exhibit lifelong skeletal muscle growth, characterized by continued fiber hypertrophy and the formation of new muscle fibers maintained by a persistent progenitor cell population. However, the limited availability of stable muscle progenitor cell lines from commercially important species such as Atlantic salmon (Salmo salar) constrains mechanistic studies and emerging applications in cellular aquaculture. Here, we report the establishment and characterization of a novel embryonic-derived salmon muscle progenitor cell line, termed SsEC. These cells were derived from

Pathogenicity and host range of avian influenza viruses: molecular determinants and virological perspectives.

The pathogenicity of avian influenza virus (AIV) is governed primarily by the hemagglutinin (HA) glycoprotein. Highly pathogenic avian influenza viruses contain a highly cleavable HA molecule susceptible to ubiquitous host proteases, such as furin, resulting in severe systemic infections. In contrast, low-pathogenic avian influenza viruses have HAs that require activation by trypsin-like proteases, restricted to specific organs, causing localized and mild infections. We generated highly pathogenic virus variants by serially passaging low-pathogenic waterfowl isolates that originally replicated

Alcohol-based solvents as mobile phases for LC-MS characterization of therapeutic proteins.

Acetonitrile (ACN) is currently the preferred solvent in reversed phase (RP) chromatography for protein characterization through peptide mapping. Despite its effective performance, ACN poses toxicity risks to humans and has adverse effects on environmental sustainability. In the current work, we developed a novel alcohol-based solvent system for peptide mapping by systematic evaluation of parameters such as organic eluent composition, solvent gradient, flow rate, and column temperature. We compared the chromatographic performance and MS response of peptides between the standard (ACN based) and
