Negative modulation of NO for diaphragmatic contractile reduction induced by sepsis and restraint position.

In practice of forensic medicine, potential disease can be associated with fatal asphyxia in restraint position. Research has demonstrated that nitric oxide (NO) and nitric oxide synthase (NOS) are plentifully distributed in skeletal muscle, contributing to the regulation of contractile and relaxation. In the current study, respiratory functions, indices of diaphragmatic biomechanical functions ex vivo, as well as NO levels in serum, the expressions of diaphragmatic inducible NOS (iNOS) mRNA, and the effects of L-NNA on contractility of the diaphragm were observed in sepsis induced by cecal li

Changes in activity of stress-realizing body systems as a result of terahertz irradiation at nitric oxide frequencies in albino rats under conditions of acute stress.

We studied the effects of irradiation with teraherz electromagnetic waves at frequencies corresponding to nitric oxide emission and absorption molecular spectrum (150,176-150,664 GHz) on activity of the sympathoadrenal and pituitary-adrenal axes of the stress response in albino male rats under conditions of acute immobilization stress. It was shown that terahertz waves exert stress-limiting effect and are capable of reducing the levels of catecholamines, corticotropin, and corticosterone in male rats exposed to acute stress.

Intravital microscopy to image membrane trafficking in live rats.

Intravital microscopy is a powerful tool that enables imaging various biological processes in live animals. Here, we describe a series of procedures designed to image subcellular structures, such as endosomes and secretory vesicles in the salivary glands (SGs) of live rats. To this aim, we used fluorescently labeled molecules and/or fluorescently tagged proteins that were transiently transfected in the live animal.

Assessment of routine procedure effect on breathing parameters in mice by using whole-body plethysmography.

We used whole-body plethysmography to investigate the effect of restraint, ear marking, tail vein and retroorbital blood sampling, and tail clipping on respiration in Balb/c × TCR-HA +/- F1 hybrid mice (F1h). Baseline values of breathing parameters were determined. During the experiment, mice experienced a procedure and then plethysmographic recordings were obtained immediately and at 4, 24, and 48 h afterward. Baseline breathing parameters showed significant differences between sexes. Restraint affected minute volume differently than did handling in male mice and to a lesser extent in female

Modified constraint induced movement therapy enhanced by a neuro-development treatment-based therapeutic handling protocol: two case studies.

Modified Constraint Induced Movement Therapy (CIMT) and Neuro-Developmental Treatment (NDT) are both intervention strategies that focus on active practice to optimize function. CIMT involves constraint of the less involved upper extremity during function and NDT includes facilitation of optimal postural control and symmetry to enhance the ability to complete a given motor function. The purpose of this article is to describe an intervention protocol for children with hemiplegia that integrates key NDT and CIMT principles. Two children participated in a modified CIMT (mCIMT)/NDT program 2 hours

Intramuscular olanzapine in patients with schizophrenia: an observational study in an emergency room.

BACKGROUND: Between 2001 and 2005 important changes were observed in expert opinion about the clinical management of patients with schizophrenia (SCZ) and agitation requiring an IM psychotropic, with a growing interest for IM atypical antipsychotics. STUDY OBJECTIVES: In an effort to acquire a typical, medically and psychiatrically unselected population of severely agitated patients with SCZ, we conducted a naturalistic study in an emergency setting, consecutively enrolling agitated patients who refused oral treatment. METHOD: Measures were collected prospectively for patients with acute agi

Effect of maternal restraint stress on fetal development of ICR mice.

The present study was conducted to elucidate the susceptibility of embryos and fetuses at different gestational stages to the maternal stress in mice. Groups of pregnant ICR mice were subjected to daily 12-h restraint stress, taped in the supine position on a plastic board, on gestational days (GD) 1-4, 5-8, 9-12 and 13-16, respectively. Caesarean sections were performed on gestational day 18, and the fetuses were weighed and examined for morphological defects. During the daily restraint for 4 days, the maternal body weights markedly decreased. Although the body weights recovered gradually aft

Adaptive plasticity of head movement propensity.

Individual humans exhibit differing propensity to move the head in association with saccadic shifts in gaze. We assessed whether this tendency can be modified in normal subjects by either reducing neck mobility with a cervical collar or restricting the field of view using aperture spectacles. We quantified head movement propensity in terms of the range of orbital eccentricity within which the eyes are customarily maintained (customary ocular motor range), and the range of final eye-in-head eccentricity for which a planned saccade is likely to be executed without a concomitant head movement (ey

Stress induced alterations in striatal GABAA receptor complex.

The effect of cold-immobilized stress on the gamma-aminobutylic acid (GABA)/benzodiazepine (BZP) receptor complex in the rat striatum was examined. The stressful manipulation induced a significant decrease in the amount of [3H]muscimol binding sites in the striatal particulate fraction. On the other hand, [3H]flunitrazepam (FLN) binding and the enhancing effect of FLN or secobarbital on the [3H]muscimol binding to the striatal particulate fraction were not influenced by the stress treatment. These results suggest that cold-immobilized stress may selectively change GABAA receptor binding withou

[Disorders of cardiac contractile function and myocardial adrenoreactivity during stress depending on the thyroid hormone level].

An effect of the administration of thyroid hormones at small doses (on the one hand) and hypothyroidism (on the other hand) on stress-induced changes of myocardial contractility and adrenoreactivity was studied in experiments on 86 random bred male rats. The use of thyroid hormones at small doses irrespective of a drug form prevents stress-induced decrease of cardiac contractility and preserves a sufficiently high level of myocardial adrenoreactivity. The suppression of thyroid function of the thyroid gland led to a decrease in adrenoreactivity and caused more profound disorder of contractilit

Intrahypothalamic microinjection of calcitonin prevents stress-induced gastric lesions in rats.

Injection of salmon calcitonin into the lateral ventricle or the cisterna magna was reported to potently inhibit gastric lesions induced by cold restraint stress. The forebrain sites of action were investigated using unilateral microinjection of salmon calcitonin prior to exposing conscious pylorus-ligated rats to cold restraint stress for 2 hr. Calcitonin (100 ng), microinjected in 100 nl volume by pressure ejection from glass micropipette positioned into the lateral or ventromedial hypothalamus or the paraventricular nucleus, prevented the development of gastric lesions whereas microinjectio

Interactions amongst factors which influence severity of gastric ulceration in rats.

A number of factors have previously been demonstrated to influence the severity of stress-induced gastric ulceration in rats, including prior exposure to pre-shock and allowing animals a post-stress rest or recovery period. The primary purpose of this experiment was to investigate how one of these factors might modulate the expression of the other. Animals were pre-exposed to either signalled or unsignalled shock (using "learned helplessness" parameters), or no shock. They were later subjected to 2 hours restraint-in-water (immersion) stress, or appropriate handling control procedures. Half of

Stress-induced elevation of testosterone concentration in high ranking baboons: role of catecholamines.

In an ongoing study of wild olive baboons living in a protected reserve in Kenya, the response of the testicular axis to the acute stress of rapid capture and immobilization was studied. The stress suppressed LH and testosterone (T) concentrations; previous work showed this to be due to stress-induced release of opiates and glucocorticoids, acting to inhibit LH release and testicular sensitivity to LH, respectively. There was considerable individual variation in this phenomenon, which was related to the social status of individual males. High ranking males (by reproductive criteria) were less

Effects of vasopressin deficiency, age, and stress on stomach ulcer induction in rats.

Susceptibility to ulceration induced by restraint, restraint plus intermittent shock, and activity stress was studied in 6 week and 18 week old Brattleboro and Long-Evans rats. Older animals developed more glandular ulcers than younger animals with Brattleboro rats having significantly greater ulceration than Long-Evans rats in both conditions. With activity stress, younger subjects developed significantly more glandular ulcers than older subjects; whereas, older subjects developed significantly more nonglandular ulcers than younger subjects. In both instances, the ulceration was significantly

Opioid effects on hepatic disposition of dyes in mice.

Morphine administration acutely reduced plasma clearance of sulfobromophthalein (BSP) in mice and increased hepatic retention of this dye. Increasing morphine doses from 5 to 40 mg/kg s.c. progressively raised plasma and liver BSP levels. Morphine-treated mice, warmed to reverse hypothermia, still had higher plasma and liver BSP levels. The narcotic also raised plasma levels of two dyes which are not conjugated, indocyanine green and dibromosulfophthalein. Naloxone reversed morphine-induced elevation of plasma BSP levels. In bile duct-ligated mice, plasma BSP levels were very high but hepatic

Associations between adverse childhood experiences and clinical characteristics of eating disorders.

Patients with eating disorders (EDs) frequently report a history of childhood trauma (CT). We investigated whether certain subtypes of CT are associated with more severe features of EDs, independently of psychiatric comorbidity, and whether they act additively. One hundred and ninety-two patients with DSM-V-defined EDs were consecutively recruited. Five clinical characteristics were assessed: restraint, eating, shape and weight concerns on the EDE-Q, and daily functioning. CT was assessed by the childhood traumatism questionnaire. The clinical features were associated with at least one CT subt

Behavioral interventions to reduce maladaptive responding in youth with dual sensory impairment. An analysis of direct and concurrent effects.

Many persons who have multiple sensory, physical, and developmental disabilities display behaviors that interfere significantly with adaptive functioning and social acceptance. In this investigation, the efficacy of multiple component behavioral interventions for reducing maladaptive responding exhibited by two individuals with dual sensory impairment and profound mental retardation was evaluated. Results indicated that differential reinforcement of other behavior (DRO) approaches were not effective in either case. However, when deceleration procedures (overcorrection or brief physical restrai
