Acidic stress induces the formation of P-bodies, but not stress granules, with mild attenuation of bulk translation in Saccharomyces cerevisiae.

The stress response of eukaryotic cells often causes an attenuation of bulk translation activity and the accumulation of non-translating mRNAs into cytoplasmic mRNP (messenger ribonucleoprotein) granules termed cytoplasmic P-bodies (processing bodies) and SGs (stress granules). We examined effects of acidic stress on the formation of mRNP granules compared with other forms of stress such as glucose deprivation and a high Ca²⁺ level in Saccharomyces cerevisiae. Treatment with lactic acid clearly caused the formation of P-bodies, but not SGs, and also caused an attenuation of translation initiat…

Novel insights into iron metabolism by integrating deletome and transcriptome analysis in an iron deficiency model of the yeast Saccharomyces cerevisiae.

BACKGROUND: Iron-deficiency anemia is the most prevalent form of anemia world-wide. The yeast Saccharomyces cerevisiae has been used as a model of cellular iron deficiency, in part because many of its cellular pathways are conserved. To better understand how cells respond to changes in iron availability, we profiled the yeast genome with a parallel analysis of homozygous deletion mutants to identify essential components and cellular processes required for optimal growth under iron-limited conditions. To complement this analysis, we compared those genes identified as important for fitness to th…

Polymerase eta is a short-lived, proteasomally degraded protein that is temporarily stabilized following UV irradiation in Saccharomyces cerevisiae.

Saccharomyces cerevisiae Rad30 is the homolog of human DNA polymerase eta whose inactivation leads to the cancer-prone syndrome xeroderma pigmentosum variant. Both human and yeast polymerase eta are responsible for error-free bypass of UV-induced cis-syn pyrimidine dimers and several other DNA lesions. Here we show, using yeast strains expressing TAP-tagged Rad30, that the level of this protein is post-translationally regulated via ubiquitination and proteasome-mediated degradation. The half-life of Rad30 is 20 min and it increases due to proteasomal defects. Mutations inactivating components …

Anti-Saccharomyces cerevisiae antibody titers are stable over time in Crohn's patients and are not inducible in murine models of colitis.

AIM: To investigate ASCA production over time in CD and murine colitis in order to further our understanding of their etiology. MATERIALS AND METHODS: Sixty-six CD patients were compared to ulcerative colitis (UC) and irritable bowel syndrome patients with respect to ASCA production as measured by ELISA. ASCA IgG or IgA positivity as well as change in titers over a period of up to 3 years (Delta IgG/A) was correlated with clinical parameters such as CD activity index (CDAI) and C-reactive protein levels (CRP). Moreover, two murine models of colitis (DSS and IL-10 knock out) were compared to c

[Assessment of usefulness of anti-Saccharomyces cerevisiae and anti-neutrophil cytoplasmic antibodies in patients with unspecific inflammatory bowel diseases].

UNLABELLED: Ulcerative colitis (UC) and Crohn's disease (CD) are the nonspecific inflammatory bowel diseases with unknown etiology. Existing diagnostic methods are in many cases insufficient for proper diagnosis and choice of treatment method. In 10% cases of inflammatory bowel disease indeterminate colitis (IC) is described. It includes cases with diverse clinical manifestations, range and histopathological picture. New methods to distinguish inflammatory bowel diseases, chose proper treatment and monitor their activity are searched. Anti-Saccharomyces cerevisiae antibodies (ASCA) and antineu…

Increased genome instability and telomere length in the elg1-deficient Saccharomyces cerevisiae mutant are regulated by S-phase checkpoints.

Gross chromosomal rearrangements (GCRs) are frequently observed in cancer cells. Abnormalities in different DNA metabolism including DNA replication, cell cycle checkpoints, chromatin remodeling, telomere maintenance, and DNA recombination and repair cause GCRs in Saccharomyces cerevisiae. Recently, we used genome-wide screening to identify several genes the deletion of which increases GCRs in S. cerevisiae. Elg1, which was discovered during this screening, functions in DNA replication by participating in an alternative replication factor complex. Here we further characterize the GCR suppressi…

Comparison of the supplemental effects of Saccharomyces cerevisiae and antibiotics in low-protein and high-fibre diets fed to broiler chickens.

Two experiments were completed to compare the supplemental effects of yeast Saccharomyces cerevisiae and sub-therapeutic antibiotics in high-fibre and low-protein diets for broiler chicks. In experiment 1, yeast was added at 1.5, 3.0 and 6.0 g kg-1 while penicillin, tylosin or neoterramycin were added at 150 mg kg-1 into different batches of a high-fibre diet containing 250 g kg-1 palm kernel meal. Saccharomyces cerevisiae and antibiotics increased BW, BWG, FCR, feed intake, carcass and breast weights above the control. Small intestine weight was reduced by antibiotics, while yeast caused a lo…

Evidence that the MIF2 gene of Saccharomyces cerevisiae encodes a centromere protein with homology to the mammalian centromere protein CENP-C.

The MIF2 gene of Saccharomyces cerevisiae has been implicated in mitosis. Here we provide genetic evidence that MIF2 encodes a centromere protein. Specifically, we found that mutations in MIF2 stabilize dicentric minichromosomes and confer high instability (i.e., a synthetic acentric phenotype) to chromosomes that bear a cis-acting mutation in element I of the yeast centromeric DNA (CDEI). Similarly, we observed synthetic phenotypes between mutations in MIF2 and trans-acting mutations in three known yeast centromere protein genes-CEP1/CBF1/CPF1, NDC10/CBF2, and CEP3/CBF3B. In addition, the mif…

The production of extracellular and intracellular free amino acids during aerated fermentation of glucose by baker's yeast (Saccharomyces cerevisiae).

During a study of the effects of a high level of NaCl on the content of free intracellular amino acids in baker's yeast grown in aerated fermentation of glucose it was found (Malaney et al. 1988, 1989; Malaney and Tanner 1988) that 0.6 mol/L exogenous NaCl significantly increased the content of free intracellular citrulline, glutamine, ornithine, arginine and lysine (all basic amino acids) over that observed at zero mol/L exogenous NaCl. (Exogenous is defined as salt added beyond that present in the mineral salts in the culture medium.) This paper describes the production and relative relation

Multistress resistance of Saccharomyces cerevisiae is generated by insertion of retrotransposon Ty into the 5' coding region of the adenylate cyclase gene.

Heat shock-resistant mutants, which were isolated by their ability to withstand lethal heat treatment, were characterized. Resistance was demonstrated to be a consequence of insertion of retrotransposon Ty into either the 5' coding or noncoding region, close to the putative initiation codon of the adenylate cyclase gene CYR1 (or CDC35). These heat shock-resistant mutants contained about threefold lower adenylate cyclase activity than wild-type strains. The mutants were also observed to be resistant to other stresses such as UV light and ethanol. These results demonstrate that multistress resis

Mixed-fermentation effects of co-fermentation with Hanseniaspora uvarum and Saccharomyces cerevisiae on the flavor quality of Ziyan tea wine.

Ziyan tea wine is an anthocyanin-rich fermented beverage with distinctive colour and flavor. In this study, we compared single and mixed yeast fermentation. Co-fermentation with S. cerevisiae and Hanseniaspora uvarum at a ratio of 1:1 significantly increased the sensory score compared with S. cerevisiae monoculture. Enartis Ferm Aroma White and H. uvarum at a ratio of 1:2 also produced a high sensory score, but the difference from Enartis Ferm Aroma White monoculture wasn't significant. HS-SPME-GC-MS and spectrophotometric analyses showed that co-fermentation changed the volatile profile of Zi

Effects of Sequential Fermentation with Saccharomyces bayanus and Lactobacillus brevis on the Metabolite Composition and Antioxidant Activity of Chinese Yam Juice.

Chinese yam (Dioscorea opposita Thunb.) is a nutrient-rich tuber with recognized health benefits, yet its application in beverage products remains limited due to processing and formulation challenges. In this study, a sequential fermentation strategy was adopted, using Saccharomyces bayanus followed by Lactobacillus brevis to enhance microbial viability and metabolic activity in Chinese yam juice. Samples were collected as an unfermented control (CY), yeast-fermented juice (SP), and sequentially fermented juice (LB). Microbial analysis showed that sequential fermentation supported high LAB via…

Co-Fermentation of Wickerhamomyces anomalus and Saccharomyces cerevisiae in Bread Dough: Effects on Ethyl Acetate Production and Flavor Enhancement.

Fermentation affects the quality of bakery products. Saccharomyces cerevisiae is the traditional baking yeast, but the desire for alternative flavor profiles has driven interest in non-Saccharomyces yeasts. Wickerhamomyces anomalus, a non-Saccharomyces yeast with high ester-producing ability, has attracted attention as a promising co-fermentation partner. Although previous studies have explored its co-fermentation potential in alcoholic beverage production, the effects of its co-fermentation with S. cerevisiae on dough fermentation characteristics, bread quality, and flavor remain largely unex…

Salutary Effects of Overexpression of Rsm22, an Assembly Factor for the Mitochondrial Ribosome, on Frataxin/Yfh1 Depletion Phenotypes in Saccharomyces cerevisiae.

Frataxin is a component of the iron-sulfur (Fe-S) cluster assembly complex in mitochondria, and deficiency is associated with Friedreich ataxia (FA). The yeast homolog Yfh1 resembles and cross-complements with its human equivalent, and frataxin bypass scenarios are of particular interest because they may point to strategies for treating FA. Here, we describe frataxin/Yfh1 bypass by overexpression of Rsm22, an assembly factor for the mitochondrial ribosome. Rsm22 overexpression in Yfh1-depleted yeast cells restored critical processes in mitochondria, including Fe-S cluster assembly, lipoic acid
