Association between semaglutide 2.4 mg and risk of cardiovascular events in people with overweight or obesity without atherosclerotic cardiovascular disease in the real-world: The SCORE - primary prevention study.

BACKGROUND: Semaglutide 2.4 mg is approved for the secondary prevention of major adverse cardiovascular events (MACE) among individuals with overweight/obesity. We investigated its association with MACE among individuals with overweight/obesity without diabetes who had risk factors for atherosclerotic cardiovascular disease (ASCVD) in the real-world setting. METHODS: Adults (≥45 years) with overweight/obesity and ≥3 risk factors for ASCVD but without diabetes were identified from a U.S. database (01/01/2016-12/31/2024). Patients initiating semaglutide 2.4 mg were propensity-score matched 1:2 …

Case Report: Oral semaglutide-associated depression in a patient with type 2 diabetes mellitus: a dose-dependent recurrence confirmed on rechallenge and managed with vortioxetine-bupropion combination therapy.

BACKGROUND: Oral semaglutide carries a neuropsychiatric safety profile that continues to evolve. Large clinical trials have not, at the group level, demonstrated excess psychiatric adverse events-but pharmacovigilance databases and case reports have documented a signal for depression and suicidal ideation in individual patients. A dose-dependent onset with rechallenge has not, to our knowledge, been previously described for the oral formulation, though such an association cannot be established from a single observation. CASE PRESENTATION: We describe a 54-year-old man with type 2 diabetes and…

Efficacy and safety of once-weekly subcutaneous semaglutide 2.4 mg for the management of overweight or obesity in Asian populations: A systematic review, meta-analysis and meta-regression of randomised trials.

AIMS: The rising prevalence of overweight and obesity among Asian populations poses a critical public health concern. Semaglutide 2.4 mg, a glucagon-like peptide-1 receptor agonist, has demonstrated promising weight-reduction effects in global populations, but its efficacy and safety profile in Asians remain less comprehensively examined. This meta-analysis aimed to evaluate the effectiveness and tolerability of semaglutide 2.4 mg compared to placebo in Asian individuals with overweight or obesity. MATERIALS AND METHODS: A systematic literature search was conducted in Medline, ScienceDirect, …

As Few as Three Months of Preoperative Semaglutide Exposure Prior to Total Knee Arthroplasty Is Associated With Reduced Postoperative Adverse Events in Patients Who Have Type II Diabetes.

BACKGROUND: Semaglutide, a glucagon-like peptide-1 receptor agonist, has been increasingly used for the management of type II diabetes mellitus (T2DM). Although recent literature suggests semaglutide may reduce postoperative total knee arthroplasty (TKA) complications, to our knowledge, the minimum duration of semaglutide exposure associated with superior outcomes has never been studied. METHODS: A retrospective cohort of patients with T2DM was identified from a national database and stratified into one of five mutually exclusive groups of semaglutide exposure before TKA: six to 12 months, th…

Efficacy and safety of semaglutide versus placebo for people with schizophrenia on clozapine with obesity (COaST): a phase 2, multi-centre, participant and investigator- blinded, randomised controlled trial in Australia.

BACKGROUND: People with schizophrenia have a 16-20-year reduction in life expectancy, primarily due to cardiometabolic disease. Clozapine, the most efficacious antipsychotic for treatment-resistant schizophrenia, is associated with weight gain and metabolic dysfunction. Glucagon-like peptide-1 receptor agonists, including semaglutide, contribute to substantial weight loss in the general population, but their effect and safety profile in people with schizophrenia remain unknown. We evaluated the efficacy and safety of semaglutide for weight reduction in individuals with schizophrenia who were p…

Efficacy and safety of once-weekly subcutaneous semaglutide on weight loss in patients with overweight or obesity without diabetes mellitus-A systematic review and meta-analysis of randomized controlled trials.

Semaglutide is found to be efficient for weight loss in patients with overweight or obesity with diabetes mellitus (DM). With a wide range of adverse events reported, the efficacy and safety of once-weekly subcutaneous semaglutide in individuals without DM, with overweight or obesity, is unclear. We conducted a comprehensive meta-analysis of randomized studies on once-weekly semaglutide in this patient population. We identified nine studies with 11,641 patients in the semaglutide group and 10,479 in the placebo group. We observed that semaglutide resulted in significant benefits, including cha…
