Tissue response in sporotrichosis: light and electron microscopy studies.

On the basis of electron microscopy examination of human tissue affected by sporotrichosis, we have proposed a classification, at the light microscopy level, of fungal cells in tissue. Light and electron microscopy observations clarified the following five points: (1) in sporotrichosis lesions in man, the fungal cells are frequently phagocytized by polymorphonuclear leucocytes (PMNs); (2) phagocytosis of phagocytes by other phagocytes was common, and giant cells phagocytized both fungal cells and PMNs; (3) fungal cells in the tissues of lesions were thought to be highly viable, and their buddi

Ultrastructure of asteroid bodies in sporotrichosis.

Ultrathin sections were prepared from a paraffin-embedded tissue specimen which had been obtained from a sporotrichosis patient and in which large numbers of asteroid bodies had been observed. Electronmicroscopic examination of the ultrastructure of the asteroid bodies revealed a central fungal cell with peripheral rays. These cells were almost structurally identical with fungal elements found elsewhere than in asteroid bodies; and in the rays, electron-dense granular substance was arranged in layers marked by fissures and material resembling remnants of destroyed host cells. The outermost lay

Cutaneous sporotrichosis in forestry workers. Epidemic due to contaminated Sphagnum moss.

In December 1975 and January and February 1976, an epidemic of cutaneous sporotrichosis occurred in Mississippi among forestry workers and other persons exposed to sphagnum moss used in packing pine seedlings. Seventeen cases were identified, 15 of which were from patients who had been exposed to sphagnum moss from a single source. Attack rates were significantly higher among workers exposed to this moss than among those not exposed. Sporothrix schenckii was cultured from the implicated batch of sphagnum moss but not from other batches. The source of contamination of the sphagnum moss that cau…

A Diabetic Elderly Man with Finger Ulcer.

Fixed cutaneous sporotrichosis is a differential diagnosis that can be considered in diabetic patients who present with a poorly healing ulcer. Although its prevalence is low, it can occur in patients with immunocompromised status. Here we report a case of a 70-year-old man with diabetes mellitus who presented with a 1-month history of an unhealed ulcer over the tip of his left middle finger. He experienced a cat bite over his left middle finger 1 month prior to the appearance of the lesion. A skin biopsy revealed the presence of Sporothrix schenckii. Oral itraconazole 200 mg twice daily was s…

A case series and review of sporotrichosis in Sikkim.

Sporotrichosis caused by the fungus Sporothrix schenckii has been widely reported from the northern Himalayan belt and the north eastern region of India. Three autochthonous cases of lymphocutaneous sporotrichosis from east and south districts of Sikkim are reported. Fluid aspirate from the nodulo-ulcerative lesions were sent for cytology and fungal culture. S. schenckii was isolated on culture and cytological examination in all three cases showed granulomatous reaction. Thermal dimorphism was demonstrated and animal pathogenicity testing was performed. Saturated solution of potassium iodide w…

Familial occurrence of zoonotic sporotrichosis.

Sporotrichosis is a subacute or chronic mycosis caused by the dimorphic fungus Sporothrix schenckii which is endemic in Brazil and is transmitted primarily through traumatic inoculation of its causative agent into the skin. The zoonotic transmission, especially from infected cats, has been demonstrated in several reports and case series. We present simultaneous occurrence of the disease in three members of the same family by scratches from an infected domestic cat. Two patients developed the lymphocutaneous form and one only developed the fixed cutaneous form. Two patients were successfully tr…

Zoonotic transmission of sporotrichosis: case report and review.

We report a case of sporotrichosis in a veterinarian who acquired the infection from a cat. Transmission was confirmed at the genetic level by demonstration that the two clinical isolates of Sporothrix schenckii had identical restriction-fragment-length profiles of whole-cell DNA. Review of the literature indicates that zoonotic transmission of sporotrichosis is rare and is virtually always associated with direct contact with an infected cat. Exposure to the large number of fungal organisms present in skin lesions of cats with sporotrichosis can result in transmission of infection to humans ev…

[Lymphocutaneous sporotrichosis in a pediatric patient, a case report].

Sporotrichosis is a subacute-chronic fungal infection caused by dimorphic fungi of the Sporothrix schenckii Complex. It is more common in tropical areas. The prevalence in Argentina is estimated between 0.01 and 0.02%. In half of the patients it manifests as lymphocutaneous sporotrichosis, which occurs after inoculation of the fungus into the skin after minor trauma. The initial lesion is a papule or nodule that occurs with the appearance of an ascending chain of mobile, painless and erythematous subcutaneous nodules. The diagnosis is made from mycological culture. The antifungal of choice is …

[Autochthonous cutaneous sporotrichosis in France].

BACKGROUND: Sporotrichosis is rare in Europe. We report a case of fixed cutaneous sporotrichosis in an immunocompetent French patient. PATIENT AND METHODS: A 35-year-old French man presented with a cutaneous nodule on the right forefinger that had been present for 6 months. The patient's work involved contact with soil. He was diagnosed with fixed cutaneous sporotrichosis following isolation of Sporothrix schenckii in a skin biopsy culture in Sabouraud medium. The culture was negative for atypical mycobacterioses. The patient recovered on oral itraconazole (Sporanox) treatment. DISCUSSION: W…

Epidemiology of human sporotrichosis investigated by amplified fragment length polymorphism.

Amplified fragment length polymorphism (AFLP) was used to analyze the genetic diversity of Peruvian strains of Sporothrix schenckii and to compare them to a panel of non-Peruvian strains. AFLP analysis suggests that the Peruvian strains can be divided into two homogeneous clusters with no reference to geographical origin or the clinical form of sporotrichosis. The strains from abroad present heterogeneous profiles, with the Bolivian strain and the Colombian strains related to one of the Peruvian population. Sequencing of internal transcribed spacer 2, used to examine the relationships over a l…

Effect of immunization and potassium iodide on polymorphonuclear leukocyte chemiluminescence in experimental murine sporotrichosis.

This study was undertaken to examine the effects of immunization with Sporothrix schenckii and oral potassium iodide (KJ) administration on the chemiluminescence (CL) response of mouse polymorphonuclear leukocytes (PMNs) in experimental murine sporotrichosis. When N-formyl-L-methionyl-L-leucyl-L-phenylalanine (FMLP) particles were used as foreign bodies to be phagocytosed, the time to the peak CL response of the PMNs in an immunized group was shortened in comparison with a non-immunized control group, and the CL intensity was found to be prolonged. Whereas administration of KJ resulted in a re…

Accuracy of diagnostic tests for feline sporotrichosis: A systematic review and meta-analysis.

BACKGROUND: Feline sporotrichosis is a subcutaneous infection caused by fungi of the Sporothrix genus, particularly concerning in endemic regions of South America, where cats are important vectors. Traditional diagnosis through fungal culture is time-consuming, which can delay the start of treatment. OBJECTIVE: Our goal was to synthesize the evidence on the accuracy of alternative diagnostic methods for feline sporotrichosis. METHODS: A systematic review was conducted following the Cochrane Collaboration recommendations and the PRISMA-DTA checklist (CRD42023456520). Searches for observationa…

Epidemiological and geographical distribution profile of urban sporotrichosis in the city of São Paulo.

BACKGROUND: Sporotrichosis is the most frequent subcutaneous mycosis in Latin America, where it is considered endemic. At the end of the 20th century, the first cases of zoonotic transmission were described in Rio de Janeiro, triggering an epidemic outbreak that spread to other regions of Brazil. The lack of disease notification omits its real occurrence in the country, which happens in its most populous city, São Paulo. OBJECTIVE: To evaluate the epidemiological aspects of the patients seen at a hospital in São Paulo aiming at establishing the geographic distribution of this disease. METHOD

Characterization of animal sporotrichosis in a highly urbanized area.

An epidemiological characterization of animal sporotrichosis was carried out between 2017 and 2018, in a highly urbanized area in Brazil, including outcomes and the spatial distribution of the cases according to the health vulnerability index (HVI) of the study territory. One hundred and sixty-five cats and four dogs suspected of sporotrichosis were identified by a surveillance and control program previously implemented in the study area. One hundred and five of these animals (62.1 %) were considered positive for Sporothrix spp., of which 103 were cats and two were dogs. Cases predominated amo…
