Aniracetam augments, and midazolam inhibits, the long-term potentiation in guinea-pig hippocampal slices.

The effects of aniracetam, a nootropic drug, and midazolam, which produces amnesia, on the long-term potentiation (LTP) of population spikes was investigated using hippocampal slices (CA3 area) from the guinea pig. Aniracetam at concentrations of 10(-7) and 10(-8) M, but not at 10(-6) M, significantly augmented LTP. On the other hand, midazolam (10(-9) M) significantly suppressed LTP. The suppressive effect was antagonized by Ro 15-1788 (10(-8) M). Both drugs did not affect the population spikes in the absence of tetanic stimulation at those concentrations. It was suggested that in vitro appli

[Algoneurodystrophy].

The authors consider the extensive literature concerning a quite frequently observed clinical picture, that has been named in different way by each researcher. Algoneurodystrophy is the name used in this analysis. They make a review of the most important works appeared in literature. It points the contrasting aspects of the research made till now. Therefore, in authors' opinion, it's necessary for them to investigate with further studies various aspects of the disease (etiological, pathogenetic, clinical, therapeutic ones, etc.) associated with a prevention of the secondary and tertiary damage

The effect of dopamine on glomus cell membranes in the rabbit.

Rabbit carotid bodies were excised and the membrane potentials (MPs) of the glomus cells were recorded in vitro. Upstream injections of dopamine (DA; 25-250 micrograms) elicited a slow, long lasting depolarization in 76% of the cells studied. The remaining 24% were hyperpolarized by DA. Regardless of polarity, the DA-induced potential was associated with a significant increase in input resistance (Ro). On the other hand, changes in MP which were produced artificially by passing direct current through the recording electrode had little effect on Ro. It is concluded that DA alters the MP and Ro

Effects of temperature on denervated carotid body (glomus) cells.

Cat carotid bodies were denervated for 3-31 days by sectioning the carotid nerve. The normal response of glomus cells to cooling by about 6 degrees C, namely, membrane depolarization and reduced input resistance (Ro) was altered. At 3 days, cooling induced normal depolarization although Ro decreased more markedly. At 7 days, cooling depolarized 53% of the cells which was accompanied by variable Ro changes. Forty-seven percent of the cells were hyperpolarized and their Ro increased. At 16 days, a fall in temperature depolarized 25% of the cells which showed variable changes in Ro, whereas the r

Postfacial palsy phenomena: faulty nerve regeneration or ephaptic transmission?

Certain postfacial palsy phenomena--'facial contracture', 'crocodile tears', etc.--have been examined in relation to faulty misrouting of the nerve fibers, which thus far has been offered as an explanation for such phenomena. Analogy is made with the problem of hemifacial spasm, occurring without antecedant facial palsy. It is seen that the two conditions have many things in common and are amenable to similar methods of therapy--such as surgical decompression of the facial nerve without interrupting its fibers or by pharmacological blocking of the facial nerve. Evidence is presented to support

Preferred centripetal conduction of dendritic spikes in alligator Purkinje cells.

Dendritic action potentials in alligator Purkinje cells tend to have a unidirectional preference which favors centripetal over centrifugal propagation. This unidirectional tendency funnels the peripherally evoked dendritic spikes into the lower dendrites and soma of these cells, and it allows the peripheral dendritic branches to operate to a certain extent as partially independent functional units.

Non-Synaptic Mechanism of Ocular Dominance Plasticity.

Classic experiments showing that monocular visual disruption alters synaptic connections to binocular neurons established the fundamental concept of synaptic plasticity. Synaptic inputs that are activated coincidently with postsynaptic action potential firing are strengthened, and inputs from cells firing before or after the postsynaptic action potential are weakened. An implicit assumption, however, is that the speed of impulse transmission is not altered by visual deprivation. If so, spike time arrival at binocular neurons would be affected, thereby inducing synaptic plasticity. This possibi

Hippocampal Commissural Circuitry Shows Asymmetric cAMP-Dependent Synaptic Plasticity.

Hemispheric asymmetries in NMDAR-dependent synaptic plasticity have been described in hippocampal area CA1, but it remains unclear whether similar lateralized mechanisms exist for cyclic adenosine monophosphate (cAMP)-dependent plasticity. Here, we investigated whether cAMP-mediated potentiation of synaptic transmission in mouse CA1 exhibits hemisphere-specific properties. In recordings with electrical stimulation of CA1 inputs, a subset of recordings in the left, but not in the right hemisphere CA1, exhibited a pronounced cAMP-induced potentiation of field excitatory postsynaptic potentials (…

Correlation of Presynaptic and Postsynaptic Proteins with Pathology in Alzheimer's Disease.

Synaptic transmission is essential for nervous system function and the loss of synapses is a known major contributor to dementia. Alzheimer's disease dementia (ADD) is characterized by synaptic loss in the mesial temporal lobe and cerebral neocortex, both of which are brain areas associated with memory and cognition. The association of synaptic loss and ADD was established in the late 1980s, and it has been estimated that 30-50% of neocortical synaptic protein is lost in ADD, but there has not yet been a quantitative profiling of different synaptic proteins in different brain regions in ADD fr…

Neuromuscular synapse electrophysiology in myasthenia gravis animal models.

The neuromuscular junction (NMJ) forms the synaptic connection between a motor neuron and a skeletal muscle fiber. In order to achieve a sustained muscle contraction, this synapse has to reliably transmit motor neuronal action potentials onto the muscle fiber. To guarantee successful transmission even during intense activation of the NMJ, a safety factor of neuromuscular transmission exists. In the neuromuscular disorder myasthenia gravis (MG), autoantibodies are directed against acetylcholine receptors or, in the rarer variants, against other postsynaptic NMJ proteins. This causes loss of fun

Inhibition of phosphatase and tensin homolog induces chemical long-term potentiation in the dentate gyrus of hippocampus.

Phosphatase and Tensin Homolog (PTEN) is a tumor suppressor gene that regulates cell growth, proliferation, and survival. Although the role of PTEN in cellular processes has been well-established, its involvement in synaptic plasticity is still poorly understood. This study examined the impact of bisperoxovanadium (bpV, PTEN inhibitor, 10 µM) on basal synaptic transmission and metaplasticity in the dentate gyrus of the hippocampus. A total of 48 male Wistar rats weighing between 240 and 360 g were used for all experiments. We recorded extracellular postsynaptic potentials (EPSPs) in the dentat…

α2δ-1-Linked NMDA and AMPA Receptors in Neuropathic Pain and Gabapentinoid Action.

Chronic neuropathic pain is a debilitating condition that presents a significant therapeutic challenge. Developmental Changes in the Protein Expression Levels and Localization of the Vesicular Zinc Transporter (ZnT3) in the Mouse Central Auditory System.

Vesicular (synaptic) zinc is a neuromodulator that fine-tunes synaptic transmission and sensory processing across many brain areas, including the brainstem, hippocampus, amygdala, and neocortex. Throughout the central auditory system, synaptic zinc plays a crucial role in modulating neurotransmission as well as baseline and adaptive sound processing. However, the developmental changes in the protein expression levels and localization of the vesicular zinc transporter (ZnT3), which loads synaptic zinc into presynaptic vesicles, remain unknown-due in part to the lack of robust ZnT3 antibodies. T…

Endogenous dopamine and endocannabinoid signaling mediate cocaine-induced reversal of AMPAR synaptic potentiation in the nucleus accumbens shell.

Repeated exposure to drugs of abuse alters the structure and function of neural circuits mediating reward, generating maladaptive plasticity in circuits critical for motivated behavior. Within meso-corticolimbic dopamine circuitry, repeated exposure to cocaine induces progressive alterations in AMPAR-mediated glutamatergic synaptic transmission. During a 10-14 day period of abstinence from cocaine, AMPAR signaling is potentiated at synapses on nucleus accumbens (NAc) medium spiny neurons (MSNs), promoting a state of heightened synaptic excitability. Re-exposure to cocaine during abstinence, ho…

Regional hippocampal alteration associated with cognitive deficit following experimental brain injury: a systems, network and cellular evaluation.

Cognitive deficits persist in patients who survive traumatic brain injury (TBI). Lateral fluid percussion brain injury in the mouse, a model of human TBI, results in hippocampal-dependent cognitive impairment, similar to retrograde amnesia often associated with TBI. To identify potential substrates of the cognitive impairment, we evaluated regional neuronal loss, regional hippocampal excitability and inhibitory synaptic transmission. Design-based stereology demonstrated an approximate 40% loss of neurons through all subregions of the hippocampus following injury compared with sham. Input/outpu…

Actions of remimazolam on inhibitory transmission of rat spinal dorsal horn neurons.

Remimazolam is an ultra-short benzodiazepine that acts on the benzodiazepine site of γ-aminobutyric acid (GABA) receptors in the brain and induces sedation. Although GABA receptors are found localized in the spinal dorsal horn, no previous studies have reported the analgesic effects or investigated the cellular mechanisms of remimazolam on the spinal dorsal horn. Behavioral measures, immunohistochemistry, and in vitro whole-cell patch-clamp recordings of dorsal horn neurons were used to assess synaptic transmission. Intrathecal injection of remimazolam induced behavioral analgesia in inflammat…

An overview of the human brain myelin proteome and differences associated with schizophrenia.

OBJECTIVES: Disturbances in the myelin sheath drive disruptions in neural transmission and brain connectivity as seen in schizophrenia. Here, the myelin proteome was characterised in schizophrenia patients and healthy controls to visualise differences in proteomic profiles. METHODS: A liquid chromatography tandem mass spectrometry-based shotgun proteomic analysis was performed of a myelin-enriched fraction of postmortem brain samples from schizophrenia patients (n = 12) and mentally healthy controls (n = 8). In silico pathway analyses were performed on the resulting data. RESULTS: The presen

Preparation of Newborn Rat Brain Tissue for Ultrastructural Morphometric Analysis of Synaptic Vesicle Distribution at Nerve Terminals.

Our laboratory and many others have exploited the high resolving power of transmission electron microscopy to study the morphology and spatial organization of synaptic vesicles. In order to obtain high-quality electron micrographs that can yield the degree of morphological detail necessary for quantitative analysis of pre-synaptic vesicle distribution, optimal specimen preparation is critical. Chemical fixation is the first step in the process of specimen preparation, and of utmost importance to preserve fine ultrastructure. Vascular fixation with a glutaraldehyde-formaldehyde solution, follow
