Interleukin 4 responses in acute leukaemia patients with severe chemotherapy-induced leucopenia.

T lymphocyte functions in acute leukaemia patients with severe chemotherapy-induced leucopenia were investigated using 3 different approaches: (i) analysis of serum concentrations of the T cell cytokine interleukin 4 (IL4) demonstrated that serum IL4 levels increased during complicating bacterial infections. However, this response was modulated by a concomitant increase in serum levels of the potential IL4 antagonist soluble IL4 receptor alpha chain (sIL4R alpha). (ii) Even during leucopenia a subset of T lymphocytes derived from leucopenic patients expressed the activation markers CD25 (IL2 r

Thymic function as a central determinant of immune competence and clinical outcomes.

The thymus is a central regulator of immune competence, sustaining the continuous generation of a diverse and self-tolerant T-cell repertoire throughout life. Although traditionally regarded as primarily relevant during early development, accumulating evidence highlights its persistent role in shaping adaptive immunity across physiological and pathological conditions. In this review, we examine the role of thymic function as a determinant of immune competence and treatment efficacy across conditions in which adaptive immunity is challenged or therapeutically reshaped, including hematopoietic c

IL-17A/F double producing T cells, unstable Tregs and quiescent TRMs in clinically healed lesions are potential cellular candidates for recurrence of psoriasis.

Biological antibodies targeting key cytokines such as IL-17 and IL-23 have revolutionized psoriasis outcome. However, the recurrence remains an urgent challenge to be addressed. Currently, most of the descriptions of skin T-cell characteristics in psoriasis are derived from lesional and non-lesional skin, and their characteristics in resolved lesions (clinically healed lesions) remain vague. In order to further elucidate the cellular mechanism of recurrence, we performed single-cell sequencing and multiplexed immunohistochemical staining of T-cell subsets in autologous resolved lesion (RL), on

Memory CD8 T cell inflation vs tissue-resident memory T cells: Same patrollers, same controllers?

The induction of long-lived populations of memory T cells residing in peripheral tissues is of considerable interest for T cell-based vaccines, as they can execute immediate effector functions and thus provide protection in case of pathogen encounter at mucosal and barrier sites. Cytomegalovirus (CMV)-based vaccines support the induction and accumulation of a large population of effector memory CD8 T cells in peripheral tissues, in a process called memory inflation. Tissue-resident memory (TRM ) T cells, induced by various infections and vaccination regimens, constitute another subset of memor

Murine hepatitis caused by lymphocytic choriomeningitis virus. II. Cells involved in pathogenesis.

BACKGROUND: Human virus hepatitides are often assumed to result from pathogenic immune responses rather than from direct viral cytopathic effects, but the details are largely unknown. Hepatitis of the mouse undergoing infection with lymphocytic choriomeningitis (LCM) virus is an immunopathologic phenomenon, and its analysis may help us to understand some of the events leading to the human illnesses. EXPERIMENTAL DESIGN: Mice were infected with LCM virus and were depleted of T cells or their subsets by inoculation of monoclonal antibodies; other infected mice lacked all T lymphocytes or the CD

Irisin Pretreatment Reveals Enhanced Protective Response Upon Hematopoietic Reconstitution in the Acquired Aplastic Anemia Mouse Model.

Acquired aplastic anemia (AA) is a prevalent nonmalignant hematologic disorder characterized by primary bone marrow failure (BMF), ineffective hematopoiesis, and consequent pancytopenia associated with abnormal immune responses and autoreactive T lymphocyte-mediated dysfunction of hematopoietic stem and progenitor cells (HSPCs). Recently, we observed a remarkable decrease in serum irisin levels in patients with AA, as well as beneficial effects of irisin administration on HSPC stemness and pancytopenia-related dysimmunity during BMF; however, the optimal protective effect of irisin on AA remai

MicroRNA-21-5p participates in IgA nephropathy by driving T helper cell polarization.

BACKGROUND: Previous studies have revealed abnormal lymphocyte subsets in IgA nephropathy (IgAN). Some microRNAs have been reported to influence T helper differentiation. Here, we explored the underlying mechanism regarding how miRNAs regulate lymphocyte subsets in IgAN. METHODS: First, miRNA and mRNA profiles in PBMCs from IgAN patients and controls were obtained by next-generation sequencing and gene expression array. The target miRNAs and mRNAs were identified through combined analysis. Then, in an independent population, we detected the expression of target miRNA in CD3+ T cells and CD19+

Generation of primary human intestinal T cell transcriptomes reveals differential expression at genetic risk loci for immune-mediated disease.

OBJECTIVE: Genome-wide association studies (GWAS) have identified genetic variants within multiple risk loci as predisposing to intestinal inflammatory diseases, including Crohn's disease, ulcerative colitis and coeliac disease. Most risk variants affect regulation of transcription, but a critical challenge is to identify which genes and which cell types these variants affect. We aimed to characterise whole transcriptomes for each common T lymphocyte subset resident within the gut mucosa, and use these to infer biological insights and highlight candidate genes of interest within GWAS risk loci

In neonates with vitamin D deficiency, low lymphocyte activation markers are risk factors for infection.

Background: Vitamin D has regulatory effects on different cells of the immune system and low levels are associated with several immune-mediated diseases. Aim: To investigate the association between neonatal 25-hydroxy vitamin D (25-OHD) level and the expression of lymphocyte activation markers (HLA-DR, CD69, CD25, CD45RA) on T-lymphocyte subpopulations and its impact in neonatal infection. Methods: 25-OHD level was measured in the cord blood of 56 neonates and their mothers using an enzyme immune-assay method. Based on the 25-OHD level, infants were categorised into four groups: severe deficie

Flow cytometric analysis of TCR Vβ repertoire in patients with 22q11.2 deletion syndrome.

In 22q11.2 deletion patients, the normal decrease in T lymphocyte counts after 1-2 years is blunted such that relatively T lymphocyte numbers increase over early childhood, probably via post-thymic expansion of peripheral lymphocytes. This may leave less T lymphocyte receptor (TCR) diversity than when derived from naive thymic emigrants. We analysed TCR Vβ repertoire on 27 22q11.2 chromosome deletion patients. No patient had infection at sampling. CD3(+) CD4(+) recent thymic emigrants (RTEs) were identified by CD45RA and CD31 expression. TCR Vβ repertoire was determined using four-colour flow

Minimally expanded breast cancer tumor-infiltrating-lymphocytes provide guidance for therapeutic selection.

INTRODUCTION: The analysis of tumor-infiltrating lymphocytes often requires techniques that expand their numbers, potentially introducing bias. To address this, we performed a detailed analysis of minimally cultured TILs to evaluate whether this approach better preserves their characteristics. METHODS: The TIL culture method was based solely on tumor tissue with low IL-2 supplementation to minimize artificial alterations. The validity of this approach was confirmed by the correlation between CD3+ T cell percentages in cultures and infiltration patterns observed by immunohistochemistry. Immuno

Synovial T lymphocyte-specific immune response to Chlamydia trachomatis in Reiter's disease.

We studied the lymphocyte proliferative response to Chlamydia trachomatis in Reiter's syndrome (RS) compared with that in other rheumatic diseases. RS patients showed significantly increased C trachomatis-specific synovial fluid (SF) T cell proliferation. Proliferating cells were found in both CD4+ and CD8+ T cell subsets. The SF lymphocyte proliferative response to C trachomatis in RS was inhibited by anti-class I and class II major histocompatibility complex monoclonal antibodies, while the response to tuberculin purified protein derivative was inhibited only by anti-class II monoclonal anti

Systemic immune cell response in rats after pulmonary exposure to manganese-containing particles collected from welding aerosols.

Welding fume inhalation affects the immune system of exposed workers. Manganese (Mn) in welding fume may induce immunosuppressive effects. The goal was to determine if Mn in welding fume alters immunity by reducing the number of circulating total leukocytes and specific leukocyte sub-populations. Sprague-Dawley rats were treated by intratracheal instillation (ITI) with either a single dose (2.00 mg/rat) or repeated doses (0.125 or 2.00 mg/rat for 7 weeks) with welding fumes that contained different levels of Mn. Additional rats were treated by ITI once a week for 7 weeks with the two doses of

Effect of surface antigen-1 (SA-1) immune lymphocyte subsets and naive cell subsets in protecting scid mice from initial and persistent infection with Cryptosporidium parvum.

We tested the hypothesis that adoptive transfer of immune spleen cell subsets from Cryptosporidium parvum antigen immunized, immunocompetent BALB/c mice would prevent initial infection or terminate persistent infection in severe combined immunodeficient (scid) mice. Cell donor mice were immunized with either solubilized C. parvum oocysts and sporozoites (positive control) or a surface antigen-1 (SA-1) enriched C. parvum antigen fraction. Both groups of BALB/c cell donor mice immunized with C. parvum antigens had increased antibody titers and lymphoproliferative responses when compared with neg

[Lymphocyte subpopulation changes in children with chronic virus hepatitis D and B].

We observed 367 children with chronic viral hepatitis Delta (CVHD) and B (CVHB) and found strict secondary T-immunodeficiency, more expressed in children with CVHD. We have found also the most characterizing changes of lymphocytes subpopulation and natural killer-cells depending on etiology of CVH. As for CVHD it is characterized by more significant decrease in CD4-lymphocytes, as for CVHB--CD8-lymphocytes. The above mentioned changes of lymphocytes subpopulation in patients with CVHD and CVHB show different pathological mechanisms of these two diseases, that should be taken into account treat

Immunophenotypic characterization of tumor infiltrating lymphocytes and peripheral blood lymphocytes isolated from melanomatous and non-melanomatous Sinclair miniature swine.

Selectively-bred Sinclair miniature swine exhibit a high incidence of congenital malignant melanoma which either proves fatal (10-15% of tumor-bearing piglets) or spontaneously regresses with a biphasic immunological phenomenon (85-90%) and no recurrence of malignancy. Mononuclear leukocytes were isolated from cutaneous melanomas and peripheral blood specimens collected from melanomatous (tumor-bearing) Sinclair swine during second-phase regression, and from peripheral blood specimens collected from non-melanomatous (tumor-free) Sinclair swine and control Hanford swine. Leukocyte identities we

Lymphocyte and lymphocyte subset numbers in blood and in bronchoalveolar lavage and pleural fluid in various forms of human pulmonary tuberculosis at presentation and during recovery.

BACKGROUND: Lymphocytes have a central role in human defences against mycobacteria. A study was designed to assess the relation between lymphocyte responses and clinical pattern of disease, nutrition and recovery during treatment in patients with tuberculosis. METHODS: Lymphocyte numbers and subsets (on the basis of CD3, CD4, and CD8 monoclonal antibodies) were measured in peripheral blood and, where appropriate, bronchoalveolar lavage or pleural fluid of patients with different forms of pulmonary tuberculosis. Eleven had localised pulmonary tuberculosis, 18 miliary tuberculosis and seven a t

T cells: an emerging cast of roles in bipolar disorder.

Bipolar disorder (BD) is a distinctly heterogeneous and multifactorial disorder with a high individual and social burden. Immune pathway dysregulation is an important pathophysiological feature of BD. Recent studies have suggested a potential role for T lymphocytes in the pathogenesis of BD. Therefore, greater insight into T lymphocytes' functioning in patients with BD is essential. In this narrative review, we describe the presence of an imbalance in the ratio and altered function of T lymphocyte subsets in BD patients, mainly in T helper (Th) 1, Th2, Th17 cells and regulatory T cells, and al

The possible role of circulating immune complexes in the deficiency of cell-mediated immunity in paracoccidioidomycosis.

1. The nature and extent of immune abnormalities was studied in 28 untreated patients with a chronic moderate form of paracoccidioidomycosis (PCM). 2. The patients presented hyporeactivity to skin tests, diminished lymphocyte transformation by mitogens such as phytohemagglutinin-P and concanavalin A and by Paracoccidioides brasiliensis protein antigen. They also presented peripheral blood leukocytosis but normal absolute numbers of T-cell and T-cell subsets. 3. The patients had increased serum levels of C3d, as well as high levels of circulating immune complexes (CIC) detected by C1q-binding a
