Involvement of all-trans-retinal in acute light-induced retinopathy of mice.

Exposure to bright light can cause visual dysfunction and retinal photoreceptor damage in humans and experimental animals, but the mechanism(s) remain unclear. We investigated whether the retinoid cycle (i.e. the series of biochemical reactions required for vision through continuous generation of 11-cis-retinal and clearance of all-trans-retinal, respectively) might be involved. Previously, we reported that mice lacking two enzymes responsible for clearing all-trans-retinal, namely photoreceptor-specific ABCA4 (ATP-binding cassette transporter 4) and RDH8 (retinol dehydrogenase 8), manifested

Cis-trans isomers of vitamin A and retinene in the rhodopsin system.

Vitamin A and retinene, the carotenoid precursors of rhodopsin, occur in a variety of molecular shapes, cis-trans isomers of one another. For the synthesis of rhodopsin a specific cis isomer of vitamin A is needed. Ordinary crystalline vitamin A, as also the commercial synthetic product, both primarily all-trans, are ineffective. The main site of isomer specificity is the coupling of retinene with opsin. It is this reaction that requires a specific cis isomer of retinene. The oxidation of vitamin A to retinene by the alcohol dehydrogenase-cozymase system displays only a low degree of isomer sp

PGC-1α, mitochondrial dysfunction, and Huntington's disease.

The constant high energy demand of neurons makes them rely heavily on their mitochondria. Dysfunction of mitochondrial energy metabolism leads to reduced ATP production, impaired calcium buffering, and generation of reactive oxygen species. There is strong evidence that mitochondrial dysfunction results in neurodegeneration and may contribute to the pathogenesis of Huntington's disease (HD). Studies over the past few years have implicated an impaired function of peroxisome proliferator-activated receptor (PPAR)-γ coactivator-1α (PGC-1α), a transcriptional master coregulator of mitochondrial bi

[Hyperlipidemia and peroxisome proliferator-activated receptor (PPAR)--regulation of the PPARalpha gene by CLOCK: BMAL1].

The main pathological findings in atherosclerosis include abnormal reactions of neutrophils, lymphocytes and monocytes/macrophages, vascular smooth muscle cells and vascular endothelial cells, and the accumulation of cholesterol ester in the arterial wall. Therefore, investigating the effects of these abnormal reactions on the arterial wall may improve understanding of the mechanisms underlying atherosclerosis. Three types of peroxisome proliferator-activated receptors (PPARs): PPARalpha, PPARbeta/delta, and PPARgamma are expressed in endothelial cells. In endothelial cells, the ligands/activa

Structural basis for a filamentous morpheein model of human cystathionine beta-synthase.

Human cystathionine beta-synthase (CBS) is a vital enzyme that regulates sulfur amino acid metabolism, hydrogen sulfide production, and cellular redox balance. Using a multidisciplinary approach, we demonstrate that CBS functions as a filamentous morpheein, with its stability, turnover, and activity governed by dynamic quaternary structural transitions. Three distinct filamentous assemblies were resolved by cryo-EM and are mediated by the oligomerization loop (residues 516-525): (i) ligand-free trans-dimers that form trans-basal filaments with basal stability and activity, (ii) adenosylornithi

CTLA-4 haploinsufficiency leads to late-onset autoimmune pathology in mice.

In humans, the loss of function in a single copy of the CTLA4 gene leads to an immune disorder characterized by a spectrum of immunodeficient and autoimmune manifestations. To investigate the pathogenicity of CTLA-4 haploinsufficiency, we utilized a heterozygous mouse model carrying a patient-derived Y139C mutation. We found that young heterozygous mice exhibited lymphoproliferative features and an activated T cell phenotype, without displaying autoimmunity. Co-culture assays revealed impaired trans-endocytosis. Despite this dysfunction, they remained resistant to induced inflammation. With ag

Ternary Neurexin-T178-PTPR complexes represent a pre-synaptic core-module of neuronal synapse organization.

Synapses, prototypic sites for neuronal communication, are key to brain function. Their organization and properties are instructed by synaptic cell adhesion molecules (sCAMs) that may operate independently or in coordination through yet unknown linker proteins. Here, we used multi-epitope affinity-purifications combined with quantitative mass spectrometry and immuno-EM to comprehensively map synaptic protein networks in the mouse brain. We identify a pre-synaptic core-module assembled from the major sCAMs, Neurexins1-3 and LAR-type receptor protein-tyrosine-phosphatases (PTPRs), and the previo

The Protective Roles of IL-6 Trans-Signaling Regulated by ADAM9 on the Liver in Carbon Tetrachloride-Induced Liver Injury in Mice.

Our study was undertaken to evaluate the important role that a disintegrin and metalloproteinase 9 (ADAM9) regulates IL-6 trans-signaling in carbon tetrachloride (CCl4)-induced liver injury in mice. Mice were divided into four groups. Each group respectively received mineral oil injection, CCl4 injection, anti-ADAM9 monoclonal antibody (mAb) pretreatment and CCl4 injection, anti-ADAM9 mAb and recombinant mouse ADAM9 molecules pretreatment with CCl4 injection. Our results showed that anti-ADAM9 mAb pretreatment significantly aggravated liver injury, inhibited IL-6 trans-signaling, which led to

Pre-catalyst resting states: a kinetic, thermodynamic and quantum mechanical analyses of [PdCl2(2-oxazoline)2] complexes.

The treatment of cold ( approximately 3 degrees C) methanolic solutions of Li(2)PdCl(4) with two equivalents of 2-phenyl-2-oxazoline (Phox) results in the isolation of [PdCl(2)(Phox)(2)] (3). This complex undergoes remarkably slow isomerisation (CHCl(3)-d) at room temperature to a corresponding thermodynamic form. In addition to a theoretical treatment (DFT), the isomerisation behaviour has been analysed both kinetically and thermodynamically. These investigations lead to the conclusion that the initially formed (i.e. kinetic) isomer of 3 is the cis-form which undergoes conversion to the corre

Juvenile xanthogranuloma of the proximal nail fold.

BACKGROUND: Volatile organic compounds (VOCs), commonly emitted from vehicle exhaust and industrial activities, are prevalent air pollutants in urban environments. These compounds have been reported to cause various health effects through mechanisms such as oxidative stress and neurotoxicity. Recently, air pollution has attracted attention as a potential risk factor for age-related macular degeneration (AMD); however, the association between VOC exposure and AMD remains unclear. OBJECTIVE: This study aimed to assess VOC exposure levels among urban-dwelling AMD patients by quantifying urinary

PU.1-activated genomic regions define low-risk MDS subsets characterized by immune dysregulation and disease progression.

Myelodysplastic syndromes (MDS) are heterogeneous myeloid neoplasms associated with an increased risk of progression to secondary acute myeloid leukemia (sAML). This study investigates the genomic correlates of disease progression in MDS by profiling active genomic regulatory regions and their transcriptional impact using H3K27ac chromatin immunoprecipitation-sequencing and RNA-sequencing analysis on CD34+ bone marrow progenitors cells isolated from a prospective cohort of 86 and 357 patients, respectively. Our analysis revealed distinct patterns of genomic region activation and transcriptiona

When less is more and when it isn't: Microglial Spi1 and the limits of what we know.

Microglia are key players in Alzheimer's disease, but the transcriptional control of their phagocytic function remains unclear. Kim et al. show that mouse microglial Spi1 deletion worsens amyloid pathology by impairing Aβ clearance through Syk, Lyn, and Fcgr1, providing new insight into PU.1-dependent regulatory networks and microglial functions in neurodegeneration.

A secreted AIP-Like peptide from Helcococcus kunzii inhibits the Agr quorum sensing system of Staphylococcus aureus.

Staphylococcus aureus is a major human pathogen whose virulence is tightly regulated by the Agr quorum sensing system. In this study, we investigated the impact of Adh2, a secreted protein from the commensal bacterium Helcococcus kunzii, on S. aureus physiology and pathogenicity. Adh2 shares structural similarity with native auto-inducing peptides (AIPs), including the conserved CDFIM motif characteristic of Agr group I. We hypothesized that Adh2 interferes with Agr signaling by competitively binding the AgrC receptor. Exposure to Adh2 significantly repressed agrA and its downstream α-hemolysi

Evolution of PqsE as a Pseudomonas aeruginosa-specific regulator of LuxR-type receptors: insights from Pseudomonas and Burkholderia.

UNLABELLED: Pseudomonas aeruginosa is a Gram-negative opportunistic pathogen that poses a significant public health threat, particularly in healthcare settings. A key determinant of P. aeruginosa virulence is the regulated synthesis and release of extracellular products, which is controlled by a cell density-dependent signaling system known as quorum sensing (QS). P. aeruginosa uses a complex QS network, including two systems that rely on diffusible N-acylhomoserine lactone (AHL) signal molecules. The LuxR-type receptor RhlR is unique in that it requires not only its cognate AHL but also the a

Effects of low intracellular glutathione and CbrA-CbrB-Crc signaling on methylglyoxal sensitivity in Pseudomonas aeruginosa LasR-deficient mutants.

Pseudomonas aeruginosa, which causes many types of infections, often has loss-of-function mutations in the gene encoding LasR, a transcription factor involved in quorum sensing. Here, we report that LasR- strains are more sensitive to the electrophile methylglyoxal (MGO), which is produced by many cells, including those involved in immune defense. MGO, due to its high reactivity, must be rapidly detoxified to prevent the formation of damaging adducts on macromolecules. GloA3 is a GSH-dependent MGO glyoxalase involved in MGO detoxification, and we found that intracellular GSH levels were lower

Identification of nonsense-mediated decay inhibitors that alter the tumor immune landscape.

Despite exciting developments in cancer immunotherapy, its broad application is limited by the paucity of targetable antigens on the tumor cell surface. As an intrinsic cellular pathway, nonsense-mediated decay (NMD) conceals neoantigens through the destruction of the RNA products from genes harboring truncating mutations. We developed and conducted a high-throughput screen, based on the ratiometric analysis of transcripts, to identify critical mediators of NMD in human cells. This screen implicated disruption of kinase SMG1's phosphorylation of UPF1 as a potential disruptor of NMD. This led u

Molecular basis and therapeutic implications of binary YAPOn/YAPOff cancer classes.

Cancers have traditionally been classified based on their tissue of origin. However, with advances in sophisticated genome sequencing techniques and progression toward an era of precision medicine, it has become increasingly clear that classifying tumors based on unifying molecular features instead of tissue of origin may hold the key to improving patient outcomes. Various efforts have been undertaken to address this critical aspect of cancer biology, but it is still unclear as to the best approach to stratify tumors into different molecular classes. One approach is to define many small subcla

Cited2 is a key regulator of placental development and plasticity.

The biology of trophoblast cell lineage development and placentation is characterized by the involvement of several known transcription factors. Central to the action of a subset of these transcriptional regulators is CBP-p300 interacting transactivator with Glu/Asp-rich carboxy-terminal domain 2 (CITED2). CITED2 acts as a coregulator modulating transcription factor activities and affecting placental development and adaptations to physiological stressors. These actions of CITED2 on the trophoblast cell lineage and placentation are conserved across the mouse, rat, and human. Thus, aspects of CI

Unveiling signaling pathways inducing MHC class II expression in neutrophils.

INTRODUCTION: Gram-negative bacillary bacteremia poses a significant threat, ranking among the most severe infectious diseases capable of triggering life-threatening sepsis. Despite the unambiguous involvement of neutrophils in this potentially fatal disease, there are limited data about the molecular signaling mechanisms, phenotype, and function of human neutrophils during the early phase of gram-negative bacillary bacteremia. METHODS: By using an unbiased proteomics and flow cytometry approach, we identified an antigen-presenting cell (APC)-like phenotype in human peripheral blood neutrophi
