Species and life-stage specific differences in cadmium accumulation and cadmium induced oxidative stress, metallothionein and heat shock protein responses in white sturgeon and rainbow trout.

Understanding the mechanistic basis of differences in the sensitivity of fishes to metals is important for developing informed ecological ri … erent life-stages of two evolutionarily distinct fish species, namely rainbow (Oncorhynchus mykiss) trout and white sturgeon (Acipenser transmontanus). These two species have been shown to differ significantly in their physiological and apical responses to Cd exposure. The findings of the present study suggest that species-specific differences in the se …

Transcriptomic signatures differentiating carrier and dead-end fish hosts of proliferative kidney disease.

INTRODUCTION: Tetracapsuloides bryosalmonae is the causative agent of proliferative kidney disease (PKD) in salmonids. Brown trout function as a carrier host, while rainbow trout serve as a dead-end host. However, the molecular mechanisms underlying these host-specific differences remain poorly understood. Therefore, in the present study, we aimed to identify host-specific transcriptomic signatures distinguishing carrier and dead- end host. METHOD: Comparative transcriptomic analysis of infected kidneys from brown trout and rainbow trout was performed to identify differential gene expression …

Effects of a Plant-Based Diet During the First Month of Feeding on Alevin Rainbow Trout (Oncorhynchus mykiss) in the Development of Tongue Sensory System Regulating Feeding Behavior.

Taste perception is essential for animals to detect nutrients, providing critical dietary information necessary for growth and survival. Since the early growth performance of alevin rainbow trout (Oncorhynchus mykiss) can be affected by food intake influenced by terrestrial ingredients without fish meal and fish oil, our study aimed to evaluate the role of taste receptors in nutrient detection and the associated signaling pathways leading to central nervous system activation in the regulation of feeding behavior. We conducted a nutritional experiment from the first feeding to 30 days, comparin…

Changes in the EPA and DHA content and lipids quality parameters of rainbow trout (Oncorhynchus mykiss, Walbaum) and carp (Cyprinus carpio, L.) at individual stages of hot smoking.

The aim of this study was to evaluate qualitative changes in lipids of two most popular freshwater farmed fish (rainbow trout and carp) at particular stages of hot smoking process (brining, drying, smoking and heating). In order to characterize qualitative changes, the amount of peroxides (PV), secondary oxidation products (AsV) and the degree of hydrolysis of lipids (AV) was determined during the smoking process. The studies were carried out both in the fraction of lipids extracted using chloroform-based method (free lipids), as well as the fraction extracted by Bligh and Dyer method of the 1…

Regulation of oocyte maturation in the rainbow trout,Salmo gairdneri: role of cyclic AMP in the mechanism of action of the maturation inducing steroid (MIS), 17α-hydroxy, 20β-dihydroprogesterone.

In fish, oocyte maturation (resumption of meiosis after completion of vitellogenesis and before ovulation) is triggered by maturation inducing steroids (MIS) which generally appear to be secreted in the ovary in response to stimulation by a pituitary maturational gonadotropin. Converging data from different laboratories show that 17α-hydroxy, 20β-dihydroprogesterone (17α, 20β-OH-P) is the principal MIS in salmonoids; but clear identification remains to be done in other taxonomic groups. The experiments reported here in the rainbow troutSalmo gairdneri examine the possible involvement of oocyte …

Hematological adaptations in diploid and triploid Salvelinus fontinalis and diploid Oncorhynchus mykiss (Salmonidae, Teleostei) in response to long-term exposure to elevated temperature.

Hematology is a simple but reliable method to determine the influence of environmental stressors on the physiology and performance of teleost fish. Understanding the detailed impacts of elevated temperature on the hematology of fish can help us to evaluate fish responses to climate change. Therefore, cellular and biochemical changes in peripheral blood were investigated in diploid (2n) and triploid (3n) brook trout and in 2n rainbow trout exposed to 20 °C for 32 days in comparison to fish acclimated to 9 °C. Additionally, survival and growth rates and the relative mRNA expression of heat-shock…

Analyses of signal transduction cascades reveal an essential role of calcium ions for regulation of melatonin biosynthesis in the light-sensitive pineal organ of the rainbow trout (Oncorhynchus mykiss).

Signal transduction processes regulating melatonin production in the light-sensitive trout pineal organ were investigated by immunocytochemical and immunochemical demonstration of phosphorylated cyclic AMP-responsive element-binding protein (pCREB) and measurements of cyclic AMP, melatonin, and calcium levels. Melatonin levels were tightly controlled by light and darkness. Elevation of cyclic AMP levels by 8-bromo-cyclic AMP, forskolin, and 3-isobutyl-1-methylxanthine increased the levels of pCREB and melatonin in light- or dark-adapted pineal organs in vitro. Without pharmacological treatment…

The linkage between Na+ uptake and ammonia excretion in rainbow trout: kinetic analysis, the effects of (NH4)2SO4 and NH4HCO3 infusion and the influence of gill boundary layer pH.

The nature of the linkage between between branchial ammonia excretion (JAmm) and unidirectional Na+ influx (JNain) was studied in the freshwater rainbow trout (Oncorhynchus mykiss). Arterial plasma total [ammonia], PNH3 and JAmm were all elevated approximately threefold by intravascular infusion for 24 h with either 70 mmol l-1 (NH4)2SO4 or 140 mmol l-1 NH4HCO3 at a rate of approximately 400 micromol kg-1 h-1. Both treatments markedly stimulated JNain. NH4HCO3 induced metabolic alkalosis in the blood plasma, whereas (NH4)2SO4 caused a slight metabolic acidosis. Experiments with Hepes-buffered …

An oral DNA vaccine against infectious haematopoietic necrosis virus (IHNV) encapsulated in alginate microspheres induces dose-dependent immune responses and significant protection in rainbow trout (Oncorrhynchus mykiss).

Administered by intramuscular injection, a DNA vaccine (pIRF1A-G) containing the promoter regions upstream of the rainbow trout interferon regulatory factor 1A gene (IRF1A) driven the expression of the infectious hematopoietic necrosis virus (IHNV) glycoprotein (G) elicited protective immune responses in rainbow trout (Oncorhynchus mykiss). However, less laborious and cost-effective routes of DNA vaccine delivery are required to vaccinate large numbers of susceptible farmed fish. In this study, the pIRF1A-G vaccine was encapsulated into alginate microspheres and orally administered to rainbow …

Influence of band 3 protein absence and skeletal structures on amphiphile- and Ca(2+)-induced shape alterations in erythrocytes: a study with lamprey (Lampetra fluviatilis), trout (Onchorhynchus mykiss) and human erythrocytes.

Amphiphiles which induce either spiculated (echinocytic) or invaginated (stomatocytic) shapes in human erythrocytes, and ionophore A23187 plus Ca(2+), were studied for their capacity to induce shape alterations, vesiculation and hemolysis in the morphologically and structurally different lamprey and trout erythrocytes. Both qualitative and quantitative differences were found. Amphiphiles induced no gross morphological changes in the non-axisymmetric stomatocyte-like lamprey erythrocyte or in the flat ellipsoidal trout erythrocyte, besides a rounding up at higher amphiphile concentrations. No s…

Isotopic and chemical characteristics of mercury in organs and tissues of fish in a mercury-polluted lake: Evidence for fractionation of mercury isotopes by physiological processes.

Organs and tissues of whitefish and trout from mercury (Hg)-polluted Lake Ontario were analyzed for Hg isotopes, methylmercury (CH3 Hg+ ), and inorganic Hg to investigate possible mass-dependent fractionation (MDF) and mass-independent fractionation (MIF) of Hg isotopes by physiological processes of the fish. Isotope signatures of different body parts were defined by δ-values of 198 Hg/202 Hg, 199 Hg/202 Hg, 200 Hg/202 Hg, and 201 Hg/202 Hg ratios and by Δ-values representing effects of MIF on 199 Hg/202 Hg and 201 Hg/202 Hg ratios. The research yielded the following evidence for MDF and MIF, …

Nasal immunity is an ancient arm of the mucosal immune system of vertebrates.

The mucosal surfaces of all vertebrates have been exposed to similar evolutionary pressures for millions of years. In terrestrial vertebrates such as birds and mammals, the nasopharynx-associated lymphoid tissue (NALT) represents a first line of immune defence. Here we propose that NALT is an ancient arm of the mucosal immune system not restricted to terrestrial vertebrates. We find that NALT is present in rainbow trout and that it resembles other teleost mucosa-associated lymphoid tissues. Trout NALT consists of diffuse lymphoid cells and lacks tonsils and adenoids. The predominant B-cell sub…
