During the latter months of 1998, cases of sleeping sickness caused by Trypanosoma brucei rhodesiense presented in Soroti district, eastern Uganda, a region which had not previously experienced cases of the disease. Cattle are the main reservoir for T b rhodesiense, by contrast with sleeping sickness caused by Trypanosoma brucei gambiense in west Africa where there appears to be no epidemiologically significant animal reservoir. Several factors have been identified that interacted to produce ideal conditions for the establishment of a new disease focus. After a period of civil unrest, Soroti,

Human African trypanosomiasis is an essentially rural disease. Occurrence in urban areas is uncommon except in cities that reproduce rural conditions conducive to the survival of glossinidae, i.e., forest and water. This is the case in neighborhoods near the zoo in Brazzaville, People's Republic of the Congo and in the residual mangrove forest in Conakry, Guinea. In Kinshasa, Democratic Republic of the Congo, an average of 39 cases were reported annually from 1970 to 1995. This figure increased to 254 in 1996 and 226 in 1997. This sharp rise led authorities to organize screening operations in

Monitor lizards were sampled along the shores of Lake Victoria to detect natural infections of potentially human-infective trypanosomes. In an area with endemic rhodesian sleeping sickness, one of 19 lizards was infected (Busia, Kenya). Six of ten lizards also showed indirect evidence of infection with Trypanosoma brucei (antibody ELISA). In an area with no recent history of human disease (Rusinga Island), no parasites were found and no antibodies to T. brucei were detected. The isolate was identified as T. brucei through xenodiagnosis (completion of the life cycle in the salivary glands of ts

The relationship between serum concentrations of the prophylactic trypanocidal drug isometamidium chloride and protection against tsetse challenge with two populations of Trypanosoma congolense was investigated in Boran (Bos indicus) cattle, using an isometamidium-ELISA. Isometamidium chloride (Samorin) was administered to cattle at a dose rate of 1.0 mg/kg body weight by deep intramuscular injection. Thereafter, the animals were challenged at monthly intervals with either a drug-sensitive clone (T. congolense IL 1180) or a clone expressing a moderate level of resistance to isometamidium (T. c

Trypanosomes were isolated from the midguts of Glossina palpalis palpalis, G. pallicera pallicera and G. nigrofusca nigrofusca captured around the village of Guediboua, South West of Daloa in Côte d'Ivoire. Seventy of the 124 isolates, obtained from 688 flies, were examined for four different kinds of trypanosome using the Polymerase Chain Reaction (PCR). Prevalences were: Trypanozoon 46%, riverine-forest T. congolense 86% and savannah T. congolense 54%. Only 29 samples were examined for T. simiae but it was not detected. Just 30% of the infections involved a single kind of trypanosome; the re

Historical profiles can be used to portray past pathways of agricultural development, the factors that affected pest status and the responses made by farmers in the form of pest management. Understanding the key factors affecting these historical developments is thought to be crucial for identifying likely future scenarios and associated opportunities and constraints for improving pest management. Evidence for this view is provided by four case studies: brassica pests in the United Kingdom; tsetse fly and trypanosomiasis management in The Gambia; rice pest management in the Lop-Buri area of Th

Teneral Glossina morsitans centralis Machado were fed on the flanks of the African buffalo (Syncerus caffer Sparrman), N'Dama (Bos taurus L.) or Boran (Bos indicus L.) cattle infected with Trypanosoma congolense Broden. The infected tsetse were maintained on rabbits and on day 30 after the infected feed, the surviving tsetse were dissected to determine the infection rates. The mean infection rates (% +/- SE) in the midgut of tsetse fed on buffalo, N'Damas and Borans were 23.5 +/- 3.3, 31.6 +/- 2.7 and 33.7 +/- 4.6, respectively. The differences were not significant. However, the mean mature in

Genetic exchange is now known to occur during the life-cycle of many parasitic protozoa, including malaria parasites, coccidia and trypanosomes. The process is studied by making deliberate crosses between cloned organisms differing in clearly defined markers. In malaria parasites, crosses have been made between parasites differing in characters such as isoenzymes, antigens and other proteins, drug sensitivity, and chromosome and other DNA polymorphisms. Crosses are made by transmitting a mixture of gametes of each clone through mosquitoes to allow cross-fertilization to take place, and examini

The duration of a single isometamidium chloride (Samorin) prophylactic treatment against Trypanosoma congolense ILNat. 3.1 and T. congolense IL 285 was examined in 24 Boran steers with regard to (1) the dose of drug, (2) the level of metacyclic challenge and (3) the influence of infection with an unrelated serodeme at the time of treatment. The cattle were repeatedly challenged at monthly intervals between 2 and 7 months following treatment, either by five infected Glossina morsitans centralis or by intradermal inoculation of 5 X 10(3) or 5 X 10(5) in vitro-derived metacyclic trypanosomes. The development and distribution of Trypanosoma congolense, T vivax and T brucei in the skin of goats was examined after the animals were bitten by infected Glossina morsitans centralis. Following the tsetse bite, the trypanosomes in the skin multiplied, reaching maximum numbers when the skin reaction (chancre) of the host attained its maximum size. In goats infected with T vivax and T brucei, trypanosomes were observed circulating in the blood before the peak of the chancre, while in T congolense-infected goats microscopically detectable parasites were found in blood only during the decline o

The epidemiology of bovine trypanosomiasis on Galana Ranch, Kenya was studied for one year (July 1980--June 1981), using measurements from an experimental population of 3 types of Boran cattle. The results were applied to the estimated ranch population at risk, and an attempt was made to measure the potential economic loss due to trypanosomiasis. The potential losses in beef production due to trypanosomiasis in the ranch population at risk at Galana were estimated at around K.Shs. 8900/km2, when the stocking rate was 14.2 Tropical Livestock Units per km2. The estimated potential loss in the to

The effect of chemoprophylaxis on developing and mature Trypanosoma vivax in Glossina tachinoides and G. palpalis palpalis was evaluated. Newly emerged G. tachinoides and G.p. palpalis flies were infected with T. vivax by allowing them to feed on parasitaemic animals. Three experiments were conducted and in each the flies were divided into two groups. One group of infected flies was fed on animals treated 1 day previously with isometamidium (1 mg/kg body wt. in a 4% soln.) after which the surviving flies were dissected and examined for trypanosomes. The other group was fed on untreated animals

Forty-nine Trypanozoon stocks isolated from camels in five different areas of Kenya were screened for isoenzyme patterns of 11 enzymes using thin-layer starch gel electrophoresis. The results were compared with those obtained in other studies with Trypanosoma (Trypanozoon) evansi stocks from South America, Nigeria, Sudan and Kuwait. Most of the Kenyan camel stocks were found to be either identical or closely similar to T (T) evansi stocks from elsewhere and were therefore designated T (T) evansi also; these Kenyan stocks grew well in rats and were monomorphic. However, four other Kenyan stocks

The number of trypanosome-infected bites received by cattle grazed around an 8 ha area of forest harbouring a semi-isolated population of Glossina pallidipes Austen was estimated. The absolute size of the tsetse population was determined by mark-release-recapture techniques, the tsetse host range by the identification of blood-meals, and trypanosome infection rates by dissection of samples of tsetse. Feeding frequency was estimated and the number of cattle present was known. It was estimated that each cow received, from G. pallidipes, one infective inoculum of T. congolense every 5.8 days duri

A study was made of village goat and sheep production systems in the humid tropical zone of south-west Nigeria. A summary of the herd and flock population dynamics is given followed by a discussion of the importance of goat and sheep production in southern Nigeria and suggestions for methods of improvement. The problems of livestock production in areas of endemic trypanosomiasis are discussed and it is concluded that, where cattle are excluded due to this disease and where human population pressure is high, goats and sheep have a continuing and increasing role to play in providing meat for the

Trypanosoma brucei, the causative agent of human and animal African trypanosomiasis, cycles between a mammalian host and a tsetse fly vector. The parasite undergoes huge changes in morphology and metabolism during adaptation to each host environment. These changes are reflected in the different transcriptomes of parasites living in each host. However, it remains unclear whether chromatin-interacting proteins help mediate these changes. Bromodomain proteins localize to transcription start sites in bloodstream parasites, but whether the localization of bromodomain proteins changes as parasites d

Glossina pallidipes is the main vector of animal African trypanosomiasis and a potential vector of human African trypanosomiasis in eastern Africa where it poses a large economic burden and public health threat. Vector control efforts have succeeded in reducing infection rates, but recent resurgence in tsetse fly population density raises concerns that vector control programs require improved strategic planning over larger geographic and temporal scales. Detailed knowledge of population structure and dispersal patterns can provide the required information to improve planning. To this end, we i

BACKGROUND: Several modeling studies have been undertaken to assess the feasibility of the WHO goal of eliminating gambiense human African trypanosomiasis (g-HAT) by 2030. However, these studies have generally overlooked the effect of vector migration on disease transmission and control. Here, we evaluated the impact of vector migration on the feasibility of interrupting transmission in different g-HAT foci.
METHODS: We developed a g-HAT transmission model of a single tsetse population cluster that accounts for migration of tsetse fly into this population. We used a model calibration approach
