Impact of ethanol and acetaldehyde on DNA and cell viability of cultured neurones.

Ethanol consumption has long been associated with brain damage. However, the mechanism underlying this deleterious effect remains unclear. Among different hypotheses, acetaldehyde is regarded by certain authors as playing a major role in the expression of ethanol toxicity, but there are still some uncertainties about the exact nature of its implication. We therefore tried to characterize the profile of the alterations of neuronal viability and DNA integrity obtained after either a direct exposure to ethanol or to acetaldehyde. Ethanol at concentrations within the range of blood alcohol levels …

Role of acetaldehyde in ethanol-induced conditioned taste aversion in rats.

RATIONALE: In spite of many recent studies on the effects of acetaldehyde, it is still unclear whether acetaldehyde mediates the reinforcing and/or aversive effects of ethanol. OBJECTIVES: The present study reexamined the role of acetaldehyde in ethanol-induced conditioned taste aversion (CTA). A first experiment compared ethanol- and acetaldehyde-induced CTA. In a second experiment, cyanamide, an aldehyde dehydrogenase inhibitor, was administered before conditioning with either ethanol or acetaldehyde to investigate the effects of acetaldehyde accumulation. METHODS: A classic CTA protocol w…

Role of catalase in in vitro acetaldehyde formation by human colonic contents.

Ingested ethanol is transported to the colon via blood circulation, and intracolonic ethanol levels are equal to those of the blood ethanol levels. In the large intestine, ethanol is oxidized by colonic bacteria, and this can lead to extraordinarily high acetaldehyde levels that might be responsible, in part, for ethanol-associated carcinogenicity and gastrointestinal symptoms. It is believed that bacterial acetaldehyde formation is mediated via microbial alcohol dehydrogenases (ADHs). However, almost all cytochrome-containing aerobic and facultative anaerobic bacteria possess catalase activit…

[The dynamics of 14C-ethanol and 14C-acetaldehyde distribution in rat testes].

Changes in radioactivity were studied during one day in common proteins, karyoplasm and chromatin of testis tissue and in the blood serum of rats after administration of (14C)ethanol and (14C)acetaldehyde. In a series of experiments the acetaldehyde involvement dynamics was determined on the background of disulphiram action. It has been found that the rate of elimination of indicator doses of ethanol and acetaldehyde from the blood varies within a day. In the interval (2-4 h) the ethanol utilization rate is higher than that of acetaldehyde. It is, probably, caused by the activity of ethanol tr…

Possible role of acetaldehyde in ethanol-related rectal cocarcinogenesis in the rat.

Prospective epidemiologic studies have reported an increased risk of rectal cancer following chronic ethanol ingestion. The effect of ethanol on chemically induced colorectal carcinogenesis is controversial depending on the experimental conditions. In the present study the effect of chronic ethanol administration on acetoxymethylmethylnitrosamine-induced rectal cancer and the possible role of acetaldehyde in this process were investigated. Chronic ethanol administration resulted in an earlier occurrence of rectal tumors in this animal model. Because the concomitant administration of cyanamide,…

Use of cyanamide to determine localization of acetaldehyde metabolism in rat liver.

Various techniques have been employed previously to show that acetaldehyde is primarily oxidized in the mitochondrial matrix of rat liver. In this study, a new approach was tested. Mitochondrial low-Km aldehyde dehydrogenase (ALDH) was partially inactivated and the effect on acetaldehyde oxidation measured. Cyanamide was chosen as the ALDH inhibitor. An enzymatic activation of cyanamide, probably by catalase, was necessary for the drug to inhibit ALDH activity. The level of remaining ALDH activity after cyanamide treatment was correlated with the ability of either rat liver mitochondria or liv…

Acetaldehyde-mediated alcohol sensitivity and elevation of plasma catecholamine in man.

According to the presence and absence of aldehyde dehydrogenase (ALDH) I isozyme which had low Km for acetaldehyde, subjects were divided into two groups: the former, the usual ALDH group and the latter, the unusual ALDH one. Blood alcohol and acetaldehyde levels, plasma norepinephrine and epinephrine levels, and urinary excretion of norepinephrine, epinephrine, dopamine, vanillylmandelic acid (VMA) and 3-methoxy-4-hydroxyphenylglycol (MHPG) were determined; and the differences in these values and cardiovascular symptoms after alcohol intake between the two groups were investigated. Fifty-six …

Effects of naloxone on ethanol and acetaldehyde-induced electroencephalographic changes in rabbits.

Rabbits treated with a single IV dose of ethanol, 0.6 g/kg, exhibited rapid EEG synchronization and behavioral changes that were reversed and, in part, prevented by the opiate antagonist, naloxone, 40 micrograms/kg. The continuous intracerebroventricular infusion of acetaldehyde, 10 or 120 micrograms/min. caused biphasic changes with EEG activation and severe bradycardia followed by overt synchrony during the post-infusion period. The results of the present study provide further evidence for the ability of naloxone to counteract some acute effects of ethanol. Moreover, they do not support a ro…

The teratogenic effect of acetaldehyde: implications for the study of the fetal alcohol syndrome.

Pregnant mice were injected intravenously with either saline alone (controls) or 1% or 2% acetaldehyde in saline, on days 7, 8 and 9 of gestation. The uterine contents were examined either on day 10 or on day 19. Acetaldehyde-treated females had more resorptions both at mid-gestation and at term, though no significant difference in maternal weight gain was observed. Experimental embryos examined on day 10 were smaller, and had a higher incidence of developmental delay, as compared to controls. Anomalies of closure of the cranial and caudal regions of the neural tube were the most commonly note…

The effects of fructose and other substances on acetaldehyde and ethanol metabolism in the rat.

The purpose of this study was to test the comparative effectiveness of fructose, glucose, sucrose and alanine in increasing the rates of ethanol and acetaldehyde clearance. Intravenously administered fructose was found to be most effective, followed by alanine in increasing the rate of blood alcohol clearance. Administration of any one of the test substances during ethanol metabolism did not result in a significant change in the levels of blood acetaldehyde. Fructose partially prevented the ethanol-mediated changes in the concentration ratios of lactate/pyruvate and beta-hydroxybutyrate/acetoa…

Alcoholic cardiomyopathy: the effect of ethanol and acetaldehyde on cardiac protein synthesis.

The occurrence of cardiomy opathy in chronic alcoholics is well known, but the causes are as yet unclear (Mitchell and Cohen, 1970). Metabolic effects of ethanol, such as accumulation of triglycerides despite a decrease in fatty acid extraction (Regan et al., 1966; 1969), have been suggested as a cause of ultimate impairment of myofibrillar function. The suggestion has also been made that the detrimental effects of ethanol may actually be an acetaldehyde effect, mediated through the release of norepinephrine causing chronic chronotropic and inotropic effects which may often play a role in the …

Generation of protein adducts with malondialdehyde and acetaldehyde in muscles with predominantly type I or type II fibers in rats exposed to ethanol and the acetaldehyde dehydrogenase inhibitor cyanamide.

BACKGROUND: Alcoholic myopathy is known to primarily affect type II muscle fibers (glycolytic, fast-twitch, anaerobic), whereas type I fibers (oxidative, slow-twitch, aerobic) are relatively protected. OBJECTIVE: We investigated whether aldehyde-derived adducts of proteins with malondialdehyde and acetaldehyde are formed in muscle of rats as a result of acute exposure to ethanol and acetaldehyde. The differences between type I muscle, type II muscle, and liver tissue were also assessed. DESIGN: The formation and distribution of malondialdehyde- and acetaldehyde-protein adducts were studied w…

Relationship between acetaldehyde concentration in mouth air and tongue coating volume.

OBJECTIVE: Acetaldehyde is the first metabolite of ethanol and is produced in the epithelium by mucosal ALDH, while higher levels are derived from microbial oxidation of ethanol by oral microflora such as Candida species. However, it is uncertain whether acetaldehyde concentration in human breath is related to oral condition or local production of acetaldehyde by oral microflora. The aim of this pilot study was to investigate the relationship between physiological acetaldehyde concentration and oral condition in healthy volunteers. MATERIAL AND METHODS: Sixty-five volunteers (51 males and 14 …

Human blood acetaldehyde levels: with improved methods, a clearer picture emerges.

New reliable methods for the determination of acetaldehyde in human blood, either from separated plasma or from acid-precipitated whole blood, demonstrate that the blood of healthy Caucasians contains at most only extremely small amounts of acetaldehyde (less than 1 microM) after moderate alcohol intoxication. On the other hand, among about 50% of the Japanese population ethanol ingestion results in elevated blood acetaldehyde levels (10-50 microM) with consequent unpleasant cardiovascular responses such as facial flushing and tachycardia, apparently because of a lack of one of the acetaldehyd…

Capacity of Candida species to produce acetaldehyde at various concentrations of alcohol.

BACKGROUND: Along with tobacco use, alcohol consumption is one of the crucial factors for oral cancer. Acetaldehyde (ACH), a byproduct of alcohol, is reported as carcinogenic. One of the producers of ACH from alcohol is Candida species. The aim of the study was to quantify the ACH produced by Candida species at various concentrations of alcohol. MATERIALS AND METHODS: Clinical isolates of Candida, namely Candida albicans, Candida krusei and Candida tropicalis and C. albicans ATCC 18,804, were subjected to various concentrations of alcohol. Alcohol dehydrogenase and ACH were estimated using sp…

Kinetics and mechanism of haloacetaldehyde formation from the reaction of acetaldehyde and chlorine.

Haloacetaldehydes (HALs) are the third prevalent group of disinfection by-products (DBPs) by weight in drinking water, and their cytotoxicity and genotoxicity are higher than regulated DBPs. In order to understand their formation mechanism during chlorination and ozonation-chlorination, this study examined the reaction kinetics of chloral hydrate (CH), dichloroacetaldehyde (DCA), chloroacetaldehyde (CA) and acetaldehyde by chlorine at different pH values and chlorine doses. The results showed that the reaction rate constants increased with pH and chlorine dose, except that the degradation of C…
