Hormone-sensitive lipase deficiency in mice causes lipid storage in the adrenal cortex and impaired corticosterone response to corticotropin stimulation.

Hormone-sensitive lipase (HSL, E.C.3.1.1.3, gene designation Lipe) is reportedly the major cholesteryl esterase of adrenal cortex. Because of the potential importance of cholesteryl ester hydrolysis in steroidogenesis, gene-targeted HSL-deficient mice were assessed for adrenal cortical morphology and function. Compared with control animals, HSL deficiency results in a marked accumulation of lipid droplets both in zona glomerulosa and zona fasciculata. In the zona fasciculata, lipid accumulation was observed progressively from the outer to the inner regions, culminating near the corticomedullar

[Caryometric studies on the cortex of the adrenal gland of parathyroidectomized and thyroparathyroidectomized female rats both during and without hormonal substitution (author's transl)].

80 female Wistar rats were either parathyroidectomized or thyroparathyroidectomized and subsequently substituted with one of the following hormone regimes: thyroxine, parathormone, calcitonin, thyroxine + parathormone, or thyroxine + calcitonin. On the fifteenth day following the operation the animals were killed, the adrenals immediately removed, fixed in Bouin's fluid, dehydrated through methylbenzoate and embedded in paraffin. 5 micrometer thick sections were stained with hematoxylin--Organe G--phosphotumgstic acid--Aniline blue (HOPA). From 5 different animals in each experimental group, 2

The effect of season on the circulating concentrations of anterior pituitary, ovarian and adrenal cortex hormones and hormone binding proteins in the subarctic area; evidence of increased activity of the pituitary-ovarian axis in spring.

To evaluate the effects of season on the function of the pituitary-ovarian axis and the adrenal cortex in a northern area with great seasonal variation in the length of daylight, 10 healthy women were investigated over 1 menstrual cycle in spring (May-June), autumn (August-September), early winter (November-December) and late winter (February-March). Serum concentrations of LH, FSH, prolactin, estradiol, progesterone, total and free testosterone, cortisol, sex hormone binding globulin (SHBG) and cortisol binding globulin (CBG) were measured, and the indices of free estradiol (FEI), free androg

[Replacement therapy of adrenal insufficiency].

The adrenal cortex produces three principal categories of steroid hormones that regulate a wide variety of physiologic process from fetal to adult. Three steroid hormones are glucocorticoids (principally cortisol), mineralocorticoids (principally aldosterone) and adrenal androgens. Glucocorticoids are named for their carbohydrate mobilizing activity but are ubiquitous physiologic regulators influencing a wide variety of bodily functions. In case of deficiency of endogenous adrenal cortex steroid hormones, glucocorticoid replacement therapy is employed. There are two pathologic forms in steroid

Adrenal cortical hyperplasia: diagnostic workup, subtypes, imaging features and mimics.

Adrenal cortical hyperplasia manifests radiologically as a non-malignant growth, or enlargement, of the adrenal glands, specifically the cortex, although the cortex cannot be definitively identified by conventional imaging. Controlled by the pituitary gland, the adrenal cortex drives critical processes, such as the production of cortisol, mineralocorticoid and sex hormones. Any disruption in the multiple enzymes and hormones involved in these pathways may cause serious or life-threatening symptoms, often associated with anatomical changes in the adrenal glands. Diagnosis and treatment of adren

Adrenal cortex senescence: an ageing-related pathology?

The adrenal glands are a pair of endocrine organs that produce and secrete mineralocorticoids, glucocorticoids, sex hormones, adrenaline, and noradrenaline. They have a vital role in a range of physiological processes including regulating electrolyte balance, blood pressure and metabolism, immunomodulation, sexual development and the stress response. Adrenal cortex senescence describes the ageing-related decline in the normal functioning of the adrenal cortex, characterised by an alteration in the output of adrenal cortical hormones, in particular reduced secretion of dehydroepiandrosterone (D

Polycythemia and adrenal cortex tumor in children: a case report and literature review.

Brazil has the world's highest incidence of adrenal cortex tumor in childhood, mainly in Parana and Sao Paulo states. The diagnosis in children is done by the signs and symptoms secondary to the production of adrenal cortex hormones. The occurrence of polycythemia in patients with adrenal cancer is very rare, especially in children. The authors describe a case of adrenal cortex tumor in a child diagnosed during investigation of polycythemia. In the case described, the polycythemia was justified by increased production of erythropoietin.

Transgenic Mouse Models to Study the Development and Maintenance of the Adrenal Cortex.

The cortex of the adrenal gland is organized into concentric zones that produce distinct steroid hormones essential for body homeostasis in mammals. Mechanisms leading to the development, zonation and maintenance of the adrenal cortex are complex and have been studied since the 1800s. However, the advent of genetic manipulation and transgenic mouse models over the past 30 years has revolutionized our understanding of these mechanisms. This review lists and details the distinct Cre recombinase mouse strains available to study the adrenal cortex, and the remarkable progress total and conditional

Prolactin as an adrenocorticotropic hormone: Prolactin signalling is a conserved key regulator of sexually dimorphic adrenal gland function in health and disease.

A large number of previous reports described an effect of the pituitary hormone prolactin (PRL) on steroid hormone production by the adrenal cortex. However, those studies remained anecdotal and were never converted into a conceptual and mechanistic framework, let alone being translated into clinical care. In the light of our recently published landmark study where we described PRL signalling as a pivotal regulator of the sexually dimorphic adrenal phenotype in mouse and of adrenal androgen production in humans, we present here the overarching hypothesis that PRL signalling increases the activ

Adrenal insufficiency in the term and preterm neonate.

The adrenal gland is part of the endocrine system and produces hormones that are essential for survival. The adrenal cortex, the largest part of the adrenal gland, produces three major hormones. Glucocorticoids regulate metabolism of glucose, protein, and fat and allow the body to respond to stress by increasing blood glucose levels and cardiac output. Mineralocorticoids regulate fluid and electrolyte balance and help maintain blood pressure. Androgens are responsible for sexual differentiation in the fetus. Insufficient production of these hormones can occur for many reasons and can have a pr

Immunohistochemical demonstration of LH/CG receptors in non-neoplastic human adrenal cortex and adrenocortical tumors.

INTRODUCTION: Numerous data indicate that luteinizing hormone and/or chorionic gonadotropin (LH/CG) exert direct actions on the adrenal cortex and are involved in the adrenal pathology. However, the immunohistochemical studies on the expression of LH/CG receptors (LH/CGR) in the human adrenal cortex and in the adrenocortical tumors are scarce. MATERIAL AND METHODS: Paraffin sections of samples of 6 human non-neoplastic adrenal cortex and 25 adrenocortical tumors were immunostained with anti-LH/CGR polyclonal antibody. RESULTS: All zones of the human non-neoplastic adrenal cortex present a po

Age-related Changes in the Adrenal Cortex: Insights and Implications.

Aging is characterized by a gradual decline in physiological function. This process affects all organs including the adrenal cortex, which normally functions to produce essential steroid hormones including mineralocorticoids, glucocorticoids, and androgens. With increasing age, features such as reduced adrenal cortex size, altered zonation, and increased myeloid immune cell infiltration substantially alter the structure and function of the adrenal cortex. Many of these hallmark features of adrenal cortex aging occur both in males and females, yet are more enhanced in males. Hormonally, a subst

Peptidergic G Protein-Coupled Receptor Regulation of Adrenal Function: Bench to Bedside and Back.

An altered secretion of adrenocortical and adrenomedullary hormones plays a role in the clinical syndromes of primary aldosteronism (PA), Cushing, and pheochromocytoma. Moreover, an altered production of adrenocortical hormones and/or an abnormal release of factors by the adrenal medulla are involved in several other diseases, including high blood pressure, congestive heart failure, liver cirrhosis, nephrotic syndrome, primary reninism, renovascular hypertension, Addison disease, Bartter, Gitelman, and virilization syndromes. Understanding the regulation of adrenal function and the interaction

Adrenal Cortex Development and Maintenance: Knowledge Acquired From Mouse Models.

The adrenal cortex is an endocrine organ organized into concentric zones that are specialized to produce specific steroid hormones essential for life. The development and maintenance of the adrenal cortex are complex, as a fetal adrenal is first formed from a common primordium with the gonads, followed by its separation in a distinct primordium, the invasion of the adrenal primordium by neural crest-derived cells to form the medulla, and finally its encapsulation. The fetal cortex is then replaced by a definitive cortex, which will establish zonation and be maintained throughout life by regene
