Suppression of alcohol preference in high alcohol drinking rats: efficacy of amperozide versus naltrexone.

The selectively bred high alcohol drinking (HAD) line of rat is considered as a potential model of one type of alcoholism. The purpose of the present experiments was to compare the efficacy of two drugs on the volitional drinking of the HAD rats: the 5-HT2A receptor antagonist, amperozide, and a nonselective antagonist of opiate receptors, naltrexone. To determine the pattern of alcohol drinking of the HAD rats, a standard preference test was used in which water was offered with alcohol increased in concentrations from 3% to 30% over 11 days. The maximally preferred concentration of alcohol of…

Increased alcohol selection in rats after alcohol drinking paired with recovery from thiamine deficiency.

Repeated pairings of novel alcohol solutions (5% or 7.5% w/v) with IP injections of 267 microgram/kg thiamine in thiamine-deficient male Sprague-Dawley rats resulted in the ingestion of pharmacologically active doses of alcohol in association with recovery. Mean alcohol intake in a postrecovery fluid choice situation exceeded metabolic capacity for 7--10 days; such intake was not observed in nondeficient pair-fed rats or in formely deficient rats whose recovery was not paired with alcohol drinking. Alcohol was self-selected in the presence of water under conditions where the rats had no experi…

The Loughborough Alcohol Drinking Questionnaire (LoAD-Q) for assessment of emotional, external, and restrained drinking behaviour.

Researching alcohol-related addictive behaviours typically utilises questionnaires which measure an individual's level of alcohol use. However, these questionnaires typically either rely on a participant reporting the amount of alcohol they drink in units or a measure of disorderly drinking behaviour. Such measures do not necessarily capture the essence of drinking behaviours in the general population. This study aimed to validate a new alcohol drinking questionnaire and evaluate its internal consistency, factor structure, and test-retest reliability. Exploratory factor analysis revealed a 3-c…

Contingent gambling-drinking patterns and problem drinking severity moderate implicit gambling-alcohol associations in problem gamblers.

Although problem gambling and problem drinking often co-occur, the processes underlying this association are not well understood. This study investigated the effects of contingent gambling-drinking patterns and problem drinking severity on implicit gambling-alcohol associations. Participants were 144 (34 female) problem gamblers. The South Oaks Gambling Screen (SOGS) measured severity of problem gambling. The Brief Michigan Alcohol Screening Test (BMAST) measured severity of problem drinking. The Implicit Association Test (Greenwald, McGhee, & Schwartz, 1998, Journal of Personality and Social

Combining energy drinks and alcohol - a recipe for trouble?

BACKGROUND: Combining energy drinks (such as 'Red Bull(®)') with alcohol is becoming increasingly popular, particularly among young people. However, as yet, limited research has been conducted examining the harms associated with this form of drinking. OBJECTIVE: To review current evidence associated with combining energy drinks with alcohol and provide recommendations for addressing this issue within primary care. DISCUSSION: Combining alcohol with energy drinks can mask the signs of alcohol intoxication, resulting in greater levels of alcohol intake, dehydration, more severe and prolonged h

Drinking by alcoholic patients following hospital admission.

Ad lib fluid intake of abstinent alcoholics and alcoholics in acute withdrawal exceeded that of psychiatric controls for 3 d following admission to hospital. The urinary output of abstinent alcoholics reflected their high fluid intake, but alcoholics in withdrawal displayed an initial oliguria. Routine blood and urine analysis did not provide any evidence of dehydration among alcoholics in withdrawal. Some possible causal factors contributing to a generalized alcoholic polydipsia are outlined.

Differences and similarities in development of drinking behavior between alcoholic offspring of alcoholics and alcoholic offspring of non-alcoholics.

Self-reported initial, early, and long-term drinking behaviors, experiences, and consequences were obtained from male alcoholics completing inpatient treatment. Subjects were recruited and selected on the basis that they met DSM-III criteria for diagnosis of alcohol dependency and that their biological fathers were alcoholic (FHP; Family History Positive) or that they had no biological family history of alcoholism (FHN; Family History Negative). Results indicated that FHP subjects rated their initial taste of beer higher than FHN subjects, that FHP subjects began tasting and subsequently regul

Effect of single binge alcohol consumption on alcohol-naïve and -exposed drinking mouse models.

Binge alcohol exposure is underestimated compared to chronic intake, particularly in young or alcohol-naïve individuals. This study investigates the effects of single binge drinking, on both alcohol-naïve and alcohol-dependent drinking subjects. C57BL/6J mice received a binge ethanol dose (6 g/kg body weight; 40 % ethanol(v/v)), and subsequent haematological, hepatic, transcriptomic, and metabolomic changes were analyzed. In naïve mice, binge alcohol elevated serum AST levels, indicating acute liver injury. In ethanol-dependent mice (20 g/kg/day; 6 % ethanol(v/v)), single binge episode altered

Drinking motives and alcohol sensitivity mediate multi-dimensional genetic influences on alcohol use behaviors.

BACKGROUND: Genetic influences account for a substantial proportion of individual differences in alcohol use behaviors (AUBs). However, multiple distinct sets of genes are linked to different AUBs, which may explain their dramatic variability in risk factors and manifestations. In this study, we explore whether intermediate neurobiological traits and alcohol-related cognitions mediate the relationship between polygenic scores (PGS) and multiple AUBs, with the aim to better understand processes captured by different genetic profiles. METHODS: Using results from prior genome-wide association st

Sources of alcohol and associations with drinking frequency and binge drinking among a large sample of adolescents.

BACKGROUND: Underage drinking remains a major public health concern worldwide, contributing to a wide range of health and social harms. While adolescent alcohol use has been widely studied, there is limited evidence on how adolescents obtain alcohol and how these sources relate to drinking behaviours. METHODS: This cross-sectional analysis used data from 8,507 Grades 9-12 students participating in the 2022-2023 COMPASS study across 66 schools in Canada. Analyses were restricted to students who reported alcohol use in the past 12 months. Proportional odds mixed models assessed associations bet

Effects of heavy drinking, binge drinking, and family history of alcoholism on regional brain metabolites.

BACKGROUND: The main goals are to investigate the effects of chronic active heavy drinking on N-acetylaspartate (NAA) and other metabolites throughout the brain and to determine whether they are affected by family history (FH) of alcoholism and long-term drinking pattern. METHODS: Forty-six chronic heavy drinkers (HD) and 52 light drinkers (LD) were recruited from the community and compared on measures of regional brain structure using magnetic resonance imaging and measures of common brain metabolites in gray matter (GM) and white matter (WM) of the major lobes, subcortical nuclei, brainstem

Parental supply of sips and whole drinks of alcohol to adolescents and associations with binge drinking and alcohol-related harms: A prospective cohort study.

BACKGROUND: Parents frequently supply alcohol to their children, often only sips. We investigated whether supply of sips and whole drinks, from parents and other sources, are differentially associated with subsequent drinking outcomes. METHODS: A cohort of 1910 adolescents (mean age 12.9yrs) were surveyed annually over seven years from 2010-11. We examined prospective, adjusted associations between the quantity of supply from parental and non-parental sources in the preceding 12 months and five outcomes in the subsequent year, over several consecutive years: binge drinking; alcohol-related ha

Relationships of alcohol dehydrogenase 1B (ADH1B) and aldehyde dehydrogenase 2 (ALDH2) genotypes with alcohol sensitivity, drinking behavior and problem drinking in Japanese older men.

OBJECTIVES: Many East Asians have the genetic polymorphisms rs1229984 in alcohol dehydrogenase 1B (ADH1B) and rs671 in aldehyde dehydrogenase 2 (ALDH2). Here we analyzed the relationships of the two genotypes with alcohol sensitivity, drinking behavior and problem drinking among older and younger men living in rural areas of Japan. METHODS: The subjects were 718 Japanese men aged 63.3 ± 10.8 (mean ± SD), categorized into the older (≥65 years, n = 357) and younger (<65 years, n = 361) groups. Facial flushing frequency, drinking behavior and positive CAGE results were compared among the genotyp

Engagement in alcohol-free activities, community-level economic disadvantage, and heavy drinking following alcohol treatment.

The reinforcer pathology model proposed by Warren Bickel and colleagues focuses on within-individual motivational processes involving overvaluation of smaller immediate rewards relative to larger delayed rewards (delay discounting) and high drug reinforcing value (drug demand) as determinants of unhealthy substance use. We propose an extension of this model to incorporate broader socioecological factors (e.g., community-level economic environment) that may influence reward preference. Using a secondary data analysis of 1,726 individuals enrolled in a randomized clinical trial for alcohol use d

Global prevalence of alcohol use and binge drinking during pregnancy, and fetal alcohol spectrum disorder.

Alcohol use during pregnancy is an established cause of fetal alcohol spectrum disorder (FASD), with heavy drinking during pregnancy being explicitly linked to fetal alcohol syndrome (FAS). This paper presents recent estimates of the prevalence of: (i) any amount of alcohol use during pregnancy; (ii) one or more binge drinking episode(s) (4 or more standard drinks on a single occasion) during pregnancy; (iii) FAS; and (iv) FASD among the general population globally and by World Health Organization region. It is apparent, based on the presented estimates, that alcohol use and binge drinking occ
