Interaction of metals at the reaction center of lipoamidase.

Various metals have been shown to inhibit porcine brain lipoamidase activity at 0.1 mM, but not ferrous and ferric ions. However, in the presence of ethylenediamine tetraacetic acid (EDTA) 0.1 mM iron ions did inhibit the activity. No other metals exhibited this type of increased inhibition with the addition of EDTA. The ferric- and ferrous-EDTA compounds were equally effective. Various Fe-containing compounds also inhibited the enzyme activity, the order of inhibition being: EDTA greater than o-phenanthroline greater than azide greater than citrate. Hemin also inhibited the enzyme activity st

Factors affecting aspartase activity.

Depue, Robert H. (Hahnemann Medical College, Philadelphia), and Albert G. Moat. Factors affecting aspartase activity. J. Bacteriol. 82:383-386. 1961.-Cells of Escherichia coli grown in a glucose-mineral salts medium contain about one-fifth the amount of aspartase activity observed in cells grown in a yeast extract-peptone medium. The aspartase activity of the cells grown in glucose-salts medium would appear to be too low to provide a mechanism for synthesis of amino groups. Aspartase was purified approximately eightfold by ammonium sulfate precipitation and column chromatography of cell-free e

Characterization of a Novel Amidase and Catabolic Pathway in Paraburkholderia sp. AP1-9 Involved in Acetaminophen Biodegradation.

Acetaminophen (APAP) is a ubiquitous pharmaceutical contaminant frequently detected in agricultural environments and crop produce due to its extensive consumption and continuous release. Although microbial degradation is a major pathway for APAP removal, the molecular mechanisms underlying APAP catabolism in Paraburkholderia remain unexplored. In this study, an efficient APAP-degrading bacterium, Paraburkholderia sp. AP1-9, was isolated from activated sludge and characterized through genomic, biochemical, and metabolite analyses. Genome analysis identified acdH, encoding a previously uncharact

Chitin deacetylase 3 of Cryptococcus neoformans produces chitosans with low degree and random pattern of acetylation.

The fungal pathogen Cryptococcus neoformans poses a significant global health concern, accounting for more than 100,000 fatalities among individuals with AIDS alone every year. In contrast to other Basidiomycetes, C. neoformans actively deacetylates the chitin in its cell wall to chitosans. The deacetylation reaction is catalyzed by four chitin deacetylases (CnCDAs 1-4), presumably to evade the chitin-triggered host immunity. In this study, we conducted a comprehensive in vitro characterization of recombinant CnCDA3, revealing a preference for deacetylated units at its -1 subsite, higher activ

Deep Intronic SVA_E Retrotransposition as a Novel Factor in Canavan Disease Pathogenesis.

Canavan disease (CD) is a rare autosomal recessive leukodystrophy caused by biallelic pathogenic variants in the ASPA gene. CD is characterized by developmental delay, macrocephaly, and abnormal muscle tone. The biochemical diagnosis is confirmed by increased N-acetylaspartic acid levels. The phenotypic presentation varies, with 85-90% of individuals exhibiting the severe, typical form, while 10-15% present with a milder, atypical form. Here we report on five patients with a clinical and biochemically proven diagnosis in whom a second pathogenic variant had not yet been identified. Targeted lo

Production Enhancement of Surface-Expressed D-hydantoinase and D-carbamoylase in E. Coli by Developing a Novel Coculture Process for the Efficient Biotransformation of D-p-hydroxyphenylglycine.

D-p-hydroxyphenyl glycine (D-PHPG) is a D-amino acid used as an intermediate in the synthesis of semi-synthetic antibiotics. It is synthesized from hydantoin derivatives through two sequential enzymatic reactions involving D-hydantoinase (D-hase) and D-carbamoylase (D-case). Although whole-cell biocatalysis of D-PHPG is cost-effective, its efficiency suffers from transport obstacles, intracellular degradation, and limited substrate solubility. This study utilized a bacterial surface display system to express D-hase and D-case in Escherichia coli for D-PHPG production. Enzyme production optimiz

A systematic review and meta-analysis of nitric oxide-associated arginine metabolites in schizophrenia.

There is increasing interest in the pathophysiological role of arginine metabolism in schizophrenia, particularly in relation to the modulation of the endogenous messenger nitric oxide (NO). The assessment of specific arginine metabolites that, unlike NO, are stable can provide useful insights into NO regulatory enzymes such as isoform 1 of dimethylarginine dimethylaminohydrolase (DDAH1) and arginase. We investigated the role of arginine metabolomics in schizophrenia by conducting a systematic review and meta-analysis of the circulating concentrations of arginine metabolites associated with DD

Detection of Bacterial Neutral Ceramidase in Diabetic Foot Ulcers with an Optimized Substrate and Chemoenzymatic Probes.

Ceramidases (CDases) are important in controlling skin barrier integrity by regulating ceramide composition and affording downstream signal molecules. While the functions of epidermal CDases are known, roles of neutral CDases secreted by skin-residing microbes are undefined. Here, we developed a one-step fluorogenic substrate, S-B, for specific detection of bacterial CDase activity and inhibitor screening. We identified a non-hydrolyzable substrate mimic, C6, as the best hit. Based on C6, we designed a photoaffinity probe, JX-1, which efficiently detects bacterial CDases. Using JX-1, we identi

Fatty Acid Amide Hydrolase: An Integrative Clinical Perspective.

Introduction: Fatty acid amide hydrolase (FAAH) is one of the main terminating enzymes of the endocannabinoid system (ECS). Since being discovered in 1996, the modulation of FAAH has been viewed as a compelling alternative strategy to obtain the beneficial effect of the ECS. With a considerable amount of FAAH-related publication over time, the next step would be to comprehend the proximity of this evidence for clinical application. Objective: This review intends to highlight the rationale of FAAH modulation and provide the latest evidence from clinical studies. Methods: Publication searches we

Divergent Dimethylarginine Dimethylaminohydrolase Isoenzyme Expression in the Central Nervous System.

The endogenous methylated derivative of ʟ-arginine, Nω,Nω'-dimethyl-ʟ-arginine (asymmetric dimethylarginine, ADMA), an independent risk factor in many diseases, inhibits the activity of nitric oxide synthases and, consequently, modulates the availability of nitric oxide. While most studies on the biological role of ADMA have focused on endothelial and inducible nitric oxide synthases modulation and its contribution to cardiovascular, metabolic, and renal diseases, a role in regulating neuronal nitric oxide synthases and pathologies of the central nervous system is less understood. The two isof

A mutation of aspartoacylase gene in a Turkish patient with Canavan disease.

Canavan disease (CD) is an autosomal recessive inherited disorder characterized by spongy degeneration of the brain. The deficiency of aspartoacylase (ASPA), resulting in the accumulation of N-acetyl aspartic acid (NAA) in the brain, plays an important role in the pathogenesis of the disease. The cardinal features of this neurodegenerative disease are macrocephaly, mental retardation, and hypotonia. Magnetic resonance imaging (MRI) of the brain generally shows diffuse white matter degeneration and also elevated excretion of urinary NAA is usually seen. A large number of mutations were identifi

Relation of beta-lactamase activity and cellular location to resistance of Enterobacter to penicillins and cephalosporins.

The Enterobacter species E. aerogenes, E. cloacae, and E. hafnia were examined for resistance to penicillin and cephalosporin derivatives. All were resistant to benzyl penicillin, ampicillin, 6 [d(-)alpha-amino-p-hydroxyphenylacetamido] penicillanic acid, cephaloridine, cephalothin, and cephalexin. A significant number were sensitive to carbenicillin and 6 [d(-)alpha-carboxy-3-thienylacetamido] penicillanic acid. No differences among the three species were noted. The beta-lactamase activity was cell-bound, and was not released by osmotic shock, toluene treatment, or diphenylamine treatment. It

Functional characterization of putrescine biosynthesis in the early development of conchospores in the red seaweed Pyropia yezoensis.

The current seedling production system for Pyropia primarily relies on the collection of conchospores, which serve as seeds to produce germlings for open-sea aquaculture. Following their release from shell-borne conchocelis, conchospores undergo adhesion to seedling ropes, cell polarization, meiosis to form tetrads, and subsequent mitotic divisions to develop into germlings. the efficiency of conchospore-to-germling development is critical for aquaculture yield. In this study, we demonstrate that putrescine (PUT) plays important roles during the early development of conchospores. We analyzed d

Electrochemical Determination of Creatinine Based on Multienzyme Cascade-Modified Nafion/Gold Nanoparticles/Screen-Printed Carbon Composite Biosensors.

Creatinine serves as a crucial diagnostic biomarker for assessing kidney disease. This work developed portable non-enzymatic and multienzyme-modified electrochemical biosensors for the detection of creatinine based on commercial screen-printed carbon electrodes (SPCEs). The non-enzymatic creatinine sensor was constructed by the electrochemical deposition of AuNPs onto the surface of a pre-activated SPCE by electrochemical activation, followed by the surface modification of a Nafion membrane. The developed AuNPs/SCPE exhibited excellent reproducibility, and the proposed Nafion/AuNPs/SPCE sensor

Linker Editing of Pneumococcal Lysin ClyJ Conveys Improved Bactericidal Activity.

Streptococcus pneumoniae is a leading human pathogen uniquely characterized by choline moieties on the bacterial surface. Our previous work reported a pneumococcus-specific chimeric lysin, ClyJ, which combines the CHAP (cysteine, histidine-dependent amidohydrolase/peptidase) enzymatically active domain (EAD) from the PlyC lysin and the cell wall binding domain (CBD) from the phage SPSL1 lysin, which imparts choline binding specificity. Here, we demonstrate that the lytic activity of ClyJ can be further improved by editing the linker sequence adjoining the EAD and CBD. Keeping the net charge of

Preservation of conjugated primary bile acids by oxygenation of the small intestinal microbiota in vitro.

UNLABELLED: Bile acids play a critical role in the emulsification of dietary lipids, a critical step in the primary function of the small intestine, which is the digestion and absorption of food. Primary bile acids delivered into the small intestine are conjugated to enhance functionality, in part, by increasing aqueous solubility and preventing passive diffusion of bile acids out of the gut lumen. Bile acid function can be disrupted by the gut microbiota via the deconjugation of primary bile acids by bile salt hydrolases (BSHs), leading to their conversion into secondary bile acids through th
