Deoxygenative Amination of Unactivated Primary Alcohols Using Sulfonimidoyl Fluorides as an Ammonia Surrogate.

Herein, we describe a novel strategy for the highly selective amination of inactivated primary alcohols with bench-stable, readily available N-Boc-benzenesulfonimidoyl fluoride as a new ammonia surrogate, providing easy access to amines bearing easily removable N-Boc and N-benzenesulfonyl groups. The deoxygenative amination involves a key sulfonimidate ester intermediate generated via a sulfur(VI) fluoride exchange (SuFEx) click reaction, which subsequently undergoes an unprecedented [1,3] O-to-N rearrangement. The reaction displays high tolerance toward a wide array of functional groups, whic…

Direct C-H amination of indoles enabled by tert-butyl chlorosulfonylcarbamate with diisopropylethylamine.

Herein, we report a direct and catalyst-free C-H amination for the construction of C-N bonds at the C2 position of indoles. This method employs readily available tert-butyl chlorosulfonylcarbamate and diisopropylethylamine under very mild conditions, proceeds rapidly without indole prefunctionalization, and enables the efficient synthesis of diverse Boc-protected 2-aminoindole derivatives with a broad substrate scope.

Nitrous acid deaminative cleavage of heparins using reductive amination tagging by sulfanilic acid.

Heparins and low-molecular-weight heparins are widely used anticoagulants whose structural complexity challenges complete characterization. Chromatographic building block analyses and NMR are key techniques. Depolymerization is typically performed by heparinase digestion, though deaminative cleavage with nitrous acid remains essential for preserving the C5 configuration of uronic acids. We revisited nitrous acid depolymerization and addressed detection limitations by implementing sulfanilic acid tagging via reductive amination. This enabled UV-based detection and quantification of disaccharide…

Light-Promoted Dearomative Amination of Nonactivated Arenes via Redox-Neutral Chromium-Nitrenoid Transfer.

A light-driven dearomative amination of nonactivated arenes via redox-neutral Cr-nitrenoid transfer is reported. We designed a novel Cr(0) redox-neutral mode that overcomes traditional Cr(0)/Cr(II) redox pathways in dearomatization chemistry using (η6-arene)Cr(CO)3, thereby shifting the electrophile scope from obligatory cationic species to neutral nitrenes for the first time. This strategy enables both dearomative 1,2-hydroamination and 1,2-carboamination with exclusive regio- and stereocontrol, broad functional-group tolerance, and preferential site-selectivity for benzene rings over heteroa…

Catalytic Asymmetric Synthesis of β-Amino α-Tertiary Alcohol through Borrowing Hydrogen Amination.

The first enantioconvergent transition-metal-catalyzed amination of racemic α-tertiary 1,2-diols providing access to vicinal β-amino α-tertiary alcohols is disclosed. The iridium-catalyzed amination reaction proceeds through a chiral phosphoric acid-mediated borrowing hydrogen pathway with excellent yields and enantioselectivities for a range of amine nucleophiles and α-tertiary 1,2-diols. An array of β-amino α-tertiary alcohols were obtained with high yields and enantioselectivities (50 examples with up to 91% yield and up to 99% ee). These important chiral amino alcohol products can be easil…

Investigations of amination reactions on an antimalarial 1,2,4-triazolo[4,3-a]pyrazine scaffold.

1,2,4-Triazolo[4,3-a]pyrazines have previously been explored by the Open Source Malaria project as potent in vitro and in vivo antimalarial drug leads. With a view to generating a library of unique antimalarial 1,2,4-triazolo[4,3-a]pyrazines and exploring regiochemical preference for nucleophilic amines, we utilised the known synthetic 5-chloro-3-(4-chlorophenyl)-[1,2,4]triazolo[4,3-a]pyrazine (1) as a scaffold for aminations with 14 commercially available primary amines. Reacting scaffold 1 with excess primary amine at room temperature for 16 h generated the desired amine analogues in respect…

C-H Amination of Heteroarenes Promoted by Cyclic Hypervalent Iodine(III) Reagents (cHIR).

Herein, an efficient strategy for synthesizing novel cyclic hypervalent iodine(III) reagents (HIRs), featuring a transferable bis-sulfonimido group, has been developed. Based on a sulfonamide skeleton, these reagents are air- and moisture-stable, allowing for easy handling. Further studies have demonstrated that these reagents enable the direct C-H bond amination of diverse heteroaromatic compounds. Mechanistic studies suggest that the reaction proceeds via a radical pathway. This work advances research on cyclic HIRs and provides a practical strategy for the synthesis of valuable nitrogen-con…

Microwave-Assisted Reductive Amination of Aldehydes and Ketones Over Rhodium-Based Heterogeneous Catalysts.

Microwave (MW)-assisted reductive aminations of aldehydes and ketones were carried out in the presence of commercial and homemade heterogeneous Rh-based catalysts. Ultrasound (US) was used to improve dispersion and stability of metal nanoparticles, while commercial activated carbon and carbon nanofibers were used as supports. Moreover, various bio-derived molecules were selected as substrates, and aqueous ammonia was used as a cheap and non-toxic reagent. MW combined with heterogeneous Rh catalysts gave a 98.2 % yield in benzylamine at 80 °C with 10 bar H2 for 1 h; and a 43.3 % yield in phenyl…

Facile access to versatile aza-macrolides through iridium-catalysed cascade allyl-amination/macrolactonization.

Direct access to benzo-fused aza-macrolides was successfully achieved through iridium-catalysed intermolecular decarboxylative coupling of vinylethylene carbonates with isatoic anhydrides under relatively mild reaction conditions. Notably, this reaction proceeded through sequential allyl-amination/macrolactonization upon extrusion of CO2. Moreover, favourable fluorescence properties could be observed in the title macrocyclic products.

Organocatalytic Reductive Amination of Aldehydes with 2-Propanol: Efficient Access to N-Alkylated Amines.

Herein, we report on the development of a one-pot organocatalytic approach for the synthesis of N-alkylated secondary and tertiary amines. The reaction proceeds via the reduction of in situ generated imines, where the solvent acts as a hydride surrogate in the presence of TMSOTf, thereby facilitating the alkylation process. Mechanistic insights derived from control experiments and deuterium labeling studies reveal a solvent-mediated hydride transfer pathway. This methodology offers a streamlined approach to amine alkylation, eliminating the need for metal catalysts and providing a potentially

Integrating Au Catalysis and Engineered Amine Dehydrogenase for the Chemoenzymatic Synthesis of Chiral Aliphatic Amines.

Direct synthesis of aliphatic amines from alkynes is highly desirable due to its atom economy and high stereoselectivity but still challenging, especially for the long-chain members. Here, a combination of Au-catalyzed alkyne hydration and amine dehydrogenase-catalyzed (AmDH) reductive amination was constructed, enabling sequential conversion of alkynes into chiral amines in aqueous solutions, particularly for the synthesis of long-chain aliphatic amines on a large scale. The production of chiral aliphatic amines with more than 6 carbons reached 36-60 g/L. A suitable biocatalyst [PtAmDH (A113G…

Direct Asymmetric Reductive Amination of Alkyl (Hetero)Aryl Ketones by an Engineered Amine Dehydrogenase.

The direct asymmetric reductive amination of heteroaryl ketones has been a long-standing synthetic challenge. Here we report the engineering of an amine dehydrogenase (AmDH) from Jeotgalicoccus aerolatus for the asymmetric synthesis of chiral α-(hetero)aryl primary amines in excellent conversions (up to 99 %) and enantioselectivities (up to 99 % ee). The best AmDH variant (Ja-AmDH-M33 ) exhibited high activity and specificity toward alkyl (hetero)aryl ketones, even for those bearing a bulky alkyl chain. An efficient directed evolution approach based on molecular docking was implemented to enla…

Iridium-Catalyzed Direct Reductive Amination of Ketones and Secondary Amines: Breaking the Aliphatic Wall.

Direct reductive amination (DRA) is a ubiquitous reaction in organic chemistry. This transformation between a carbonyl group and an amine is most often achieved by using a super stoichiometric amount of hazardous hydride reagents, thus being incompatible with many sensitive functional groups. DRA could also be achieved by means of chemo- or biocatalysis, thereby attracting the interest of industry as well as academic laboratories due to the virtually perfect atom economy. Although DRAs are well-established for substrate pairs such as aldehydes with either 1° or 2° amines as well as ketones wit…

Labeling of Amine-Modified RNA.

RNA labeling enables high-contrast visualization and quantitative measurements. This chapter describes practical guidance for fluorescent dye labeling of RNA via the NHS ester-amine coupling. We detail reaction setup, cleanup by ethanol precipitation, and evaluation of labeling efficiency by absorbance spectrum analysis or by high-performance liquid chromatography (HPLC). Notes summarize common failure modes and remedies, and RNase-free practice is emphasized throughout. Although the workflow demonstrates fluorescent labeling of 3'-amine-modified RNA, the protocol is broadly applicable to 5'-e

Deaminative Allylation of Primary Amines.

Nitrogen atoms play pivotal roles in governing molecular properties and biological activity. Accordingly, nitrogen-atom manipulation has emerged as a powerful strategy for skeletal remodeling. However, its integration into classical reactions to redirect their inherent pathways and outcomes remains largely unexplored. Here, we report a deaminative allylation enabled by embedding a photoinduced N-atom deletion process within a classical N-allylation reaction. This reaction-reprogramming strategy transforms ubiquitous alkylamines into α-substituted acrylates and acrylamides-privileged motifs in

Receptors of mammalian trace amines.

The discovery of a family of G-protein coupled receptors, some of which bind and are activated by biogenic trace amines, has prompted speculation as to the physiological role of these receptors. Observations associated with the distribution of these trace amine associated receptors (TAARs) suggest that they may be involved in depression, attention-deficit hyperactivity disorder, eating disorders, migraine headaches, and Parkinson's disease. Preliminary in vitro data, obtained using cloned receptors, also suggest a role for TAARs in the function of hallucinogens.

Trace amines and Tourette's syndrome.

There has been considerable interest in recent years in possible neurochemical abnormalities in Tourette's Syndrome (TS). In studies combining neuropsychological and neurochemical measurements, we have investigated the possible roles of trace amines in this disorder. Urinary levels of free beta-phenylethylamine (PEA) and plasma levels of its precursor amino acid phenylalanine were decreased in TS patients when compared to values in normal children. These urinary PEA levels in TS patients were inversely related to several scores from the Tourette's Syndrome Global Scale (TSGS). Further investig

Interaction of biogenic amines with ethanol.

Ethanol through its primary catabolite, acetaldehyde, competitively inhibits oxidation of aldehyde dehydrogenase substrates. As a consequence biogenic amines form increased quantities of alcohols rather than the corresponding acids. During this biotransformation, condensation reactions between deaminated and intact amines may occur which can yield tetrahydropapaverolines. These compounds are closely related to precursors of opioids which is cause to link ethanol abuse to morphine addiction. There is, however, no pharmacological or clinical evidence suggesting similarities between ethanol depen
