Amylopectinosis disease isolated to the heart with normal glycogen branching enzyme activity and gene sequence.

We report a 17-month-old female patient with a rare cause of cardiomyopathy secondary to accumulation of amylopectin-like material (fibrillar glycogen) isolated to the heart. Evidence of amylopectinosis isolated to cardiac myocytes in this patient was demonstrated by histology and electron microscopy. Glycogen content, glycogen branching enzyme (GBE) activity, as well as phosphofructokinase enzyme activities measured in liver, skeletal muscle, fibroblasts and ex-transplanted heart tissue were all in the normal to lower normal ranges. Normal skeletal muscle and liver tissue histology and GBE ac

Measurement and characterization of external oil in the fried waxy maize starch granules using ATR-FTIR and XRD.

Concerns regarding increased dietary oil uptake have prompted efforts to investigate the oil absorption and distribution in fried starchy foods. In the present study, attenuated total reflection Fourier transform infrared (ATR-FTIR) spectroscopy, together with a chloroform-methanol method, was used to analyze the external and internal oil contents in fried starchy samples. The micromorphology of fried starchy samples was further investigated using scanning electron microscope (SEM), polarized light microscope (PLM) and confocal laser scanning microscopy (CLSM). The results indicated that large

Ultrastructure of in vivo-produced caryocysts containing the coccidian Caryospora bigenetica (Apicomplexa: Eimeriidae).

A cotton rat was inoculated orally with oocysts of Caryospora bigenetica from the feces of a rattlesnake. Sixteen days later the rat was euthanized, and portions of the scrotum, foot pad and muzzle were processed for histological sections and transmission electron microscopy. Sporozoites within caryocysts had typical coccidian features such as an anterior and posterior refractile body, centrally located nucleus, micronemes, rhoptries, a conoid, a micropore near the anterior refractile body, a posterior pore, amylopectin granules, lipid bodies, a Golgi-like body, a mitochondrion and subpellicul

Oral administration of Blautia wexlerae ameliorates obesity and type 2 diabetes via metabolic remodeling of the gut microbiota.

The gut microbiome is an important determinant in various diseases. Here we perform a cross-sectional study of Japanese adults and identify the Blautia genus, especially B. wexlerae, as a commensal bacterium that is inversely correlated with obesity and type 2 diabetes mellitus. Oral administration of B. wexlerae to mice induce metabolic changes and anti-inflammatory effects that decrease both high-fat diet-induced obesity and diabetes. The beneficial effects of B. wexlerae are correlated with unique amino-acid metabolism to produce S-adenosylmethionine, acetylcholine, and L-ornithine and carb

Salt-induced cellulose nanocrystal networks modulate the stability and digestion behavior of AMP/HPMC water-in-water Pickering emulsions.

Salt ions play a key role in governing the property of food system. In this work, the effects of salt ions on physicochemical property of cellulose nanocrystals (CNCs) and their stabilization behavior in Amylopectin/Hydroxypropyl methylcellulose (AMP/HPMC) water-in-water (W/W) emulsions were investigated. The addition of NaCl significantly reduced the absolute value of zeta potential of CNCs and resulted in particle aggregation, forming of network structures. These structure changes promoted the droplet clustering and bridging flocculation of AMP/HPMC W/W Pickering emulsions and enhanced their

A novel GBE1 gene variant in a child with glycogen storage disease type IV.

Glycogen storage disease type IV is an autosomal recessive disorder of carbohydrates caused by deficiency of amylo-1-4-glycanoglycosyltransferase, which leads to accumulation of amylopectin-like polysaccharides in tissues including liver, heart and neuromuscular system. More than 40 different mutations in the glycogen branching enzyme gene (GBE1) have been described. In this study, we report a 2-year-old boy who presented with developmental delay and muscle weakness. He subsequently was diagnosed with glycogen storage disease type IV based on a liver biopsy histology and electron microscopy. G

A Toxoplasma gondii Gluconeogenic Enzyme Contributes to Robust Central Carbon Metabolism and Is Essential for Replication and Virulence.

The expression of gluconeogenic enzymes is typically repressed when glucose is available. The protozoan parasite Toxoplasma gondii utilizes host glucose to sustain high rates of intracellular replication. However, despite their preferential utilization of glucose, intracellular parasites constitutively express two isoforms of the gluconeogenic enzyme fructose 1,6-bisphosphatase (TgFBP1 and TgFBP2). The rationale for constitutive expression of FBPases in T. gondii remains unclear. We find that conditional knockdown of TgFBP2 results in complete loss of intracellular growth in vitro under glucos

Molecular structure of citric acid cross-linked starch films.

The effect of citric acid (CA) on starch films has been examined. A new method to detect cross-linking of starch by CA in solution-cast films by molecular weight measurements is described. Furthermore, we managed to distinguished between free, mono- and di-esterified CA and quantify di-ester content within starch films by using a modification in the method of complexometric titration with copper(II)-sulfate. Cross-linking of starch by CA occurred at low temperature, 70°C, which we assumed is so far the lowest temperature reported where cross-linking reaction occurred. This is essential for sta

Antibodies against hydroxyethylstarch produced in rabbits by immunization with a protein-hydroxyetylstarch conjugate.

Antibodies (ABS) against hydroxyethylstarch (HES) were raised in rabbits by immunization with HES-bovine serum albumin conjugate. ABS against HES were demonstrable by gel diffusion, passive hemagglutination (PH) and passive cutaneous anaphylaxis (PCA); specificity was confirmed by inhibition of PH and neutralization of PCA. No ABS against starch could be induced in comparative experiments. Non-immunogenicity of starch was attributed to its structural similarity with glycogen, widely distributed in animal species. Absence of cross-reactivity of anti-HES ABS with starch, amylopectin and glycogen

High-amylose starch forms complexes with bioactive compounds present in the Amphipterygium adstringent ethanol extract.

Plants have been used for thousands of years to treat health problems. The bioactive compounds in parts of them are associated with cures for some diseases. The protection and release of bioactive compounds (bioaccessibility) are key issues in producing a therapeutic effect. Amylose and amylopectin, starch macromolecules, produce interactions with bioactive compounds, resulting in the formation of complexes. This study aimed to analyze the bioactive compounds in the complex produced between Amphipterygium adstringens (AA, cuachalalate) extract and high-amylose starch. Both the extract and the

Dietary starch structure modulates nitrogen metabolism in laying hens via modifying glucose release rate.

The objective of this study was to investigate the effects of starch structure (Amylopectin/Amylose, AP/AM) in a low-protein diet on production performance, nitrogen utilization efficiency, and cecal flora in laying hens. Four hundred eighty 45-wk-age Hy-Line Gray laying hens were randomly allocated to five dietary groups and subjected to a 12-wk feeding trial. The AP/AM ratios of the five experiment diets were 1.0, 1.5, 2.0, 3.0, and 4.0. The results indicated that compared to other groups, laying hens fed with AP/AM 4.0 diets showed significantly improved average egg weight and feed conversi

Finding granule-bound starch synthase enzymes with high activity using free-energy calculations.

Starch is a branched glucose polymer comprising amylopectin, of high molecular weight with many short branches, and amylose, of lower molecular weight and only a few long-chain branches. Granule-bound starch synthase I (GBSSI) is one of the main enzymes controlling amylose synthesis and structure. Production of different GBSSI mutants is laborious, this study retrospectively examined the relationship between mutation-induced changes in glucan binding and experimentally reported GBSSI activity. Atomistic molecular dynamics simulations and free-energy calculations were performed for GBSSI comple

Glycogenin-1 deficiency: a case report and review of the literature.

Pathogenic biallelic variants in GYG1, encoding for glycogenin-1, are associated with polyglucosan bodies myopathy characterized by muscle accumulation of deposits of amylopectin-like polysaccharides (MIM 616199). So far, only few cases (<50) with molecular defects in GYG1 have been reported. The proband is a 79-year-old Italian woman presenting with subacute onset of diffuse soreness, weakness in the upper limbs and diffuse muscle atrophy without cardiac or respiratory involvement. Electromyography showed myopathic features. Muscle biopsy revealed several type I muscle fibers containing inten

Effect of freeze-thaw dehydration pretreatment and bacterial cellulose addition on 3D printing performance of whole potato flour complex gel as dysphagia diets.

In this study, whole potato flour (WP) was modified through freeze-thaw dehydration (FTD) and the incorporation of bacterial cellulose (BC) to enhance the 3D printability of its gel. The effects of FTD pretreatment and BC addition on 3D printing performance of whole potato flour complex gel were systematically investigated. The results demonstrated that FTD treatment led to an increase in the amylopectin content of whole potato flour gel, while BC addition enhanced the short-range order, relative crystallinity and aggregate structure of the starch. These effects synergistically improved the st

PTPA Governs Stress-Responsive Differentiation and Metabolic Homeostasis in Toxoplasma gondii.

The protozoan parasite Toxoplasma gondii transitions between acute (tachyzoite) and chronic (bradyzoite) stages, enabling lifelong persistence in hosts. Iron depletion triggers bradyzoite differentiation, with the phosphotyrosyl phosphatase activator (PTPA) identified as a key regulator. Here, we define PTPA's role in T. gondii pathogenesis. PTPA forms a ternary complex with PP2A A/C subunits, validated by reciprocal pull-down assays. Depleting PTPA impaired tachyzoite proliferation, invasion, and gliding motility, while stress-induced bradyzoites exhibited defective cyst formation and vacuola

Multiple pathways for glucose phosphate transport and utilization support growth of Cryptosporidium parvum.

Cryptosporidium parvum is an obligate intracellular parasite with a highly reduced mitochondrion that lacks the tricarboxylic acid cycle and the ability to generate ATP, making the parasite reliant on glycolysis. Genetic ablation experiments demonstrated that neither of the two putative glucose transporters CpGT1 and CpGT2 were essential for growth. Surprisingly, hexokinase was also dispensable for parasite growth while the downstream enzyme aldolase was required, suggesting the parasite has an alternative way of obtaining phosphorylated hexose. Complementation studies in E. coli support a rol

Two cases of a non-progressive hepatic form of glycogen storage disease type IV with atypical liver pathology.

Glycogen storage disease type IV (GSD IV) is a rare inborn metabolic disorder characterized by the accumulation of amylopectin-like glycogen in the liver or other organs. The hepatic subtype may appear normal at birth but rapidly develops to liver cirrhosis in infancy. Liver pathological findings help diagnose the hepatic form of the disease, supported by analyses of enzyme activity and GBE1 gene variants. Pathology usually shows periodic acid-Schiff (PAS) positive hepatocytes resistant to diastase. We report two cases of hepatic GSD IV with pathology showing PAS positive hepatocytes that were

Detoxification of cadmium (Cd) by a novel Cd-associated and Cd-induced molecule in the stem of common reed.

Common reed (Phragmites australis) is a phytoremediator tolerant to heavy metals. In this study, we found that 70% of the cadmium (Cd) found in the stem of common reed exists in a soluble form, with more than half of the soluble Cd in the 10- to 50-kDa fraction. Based on an enzyme degradation assay, the major component of the Cd-associated molecule is assumed to be an amylopectin-like α-glucan. This molecule may associate with Cd via the carboxyl group, rather than the thiol group. The conditions required for the disengagement of Cd from the 10- to 50-kDa fraction indicated that disulfide bond
