Single nucleotide polymorphism analysis of the ITS2 region of two sympatric malaria mosquito species in Sweden: Anopheles daciae and Anopheles messeae.

Four species of the Anopheles maculipennis complex have previously been recorded in Sweden. A recent addition to the complex is Anopheles daciae, which is considered to be closely related to, but distinct from Anopheles messeae. The original designation of An. daciae was based on five genetic differences (161, 165, 167, 362 and 382) in the second internal transcribed spacer (ITS) 2 of the ribosomal RNA. Further studies have shown that only two nucleotide differences (362 and 382) robustly separate the species. Thirty-three An. maculipennis complex mosquitoes were collected in the province of U…

Does extreme asymmetric dominance promote hybridization between Anopheles coluzzii and Anopheles gambiae s.s. in seasonal malaria mosquito communities of West Africa?

BACKGROUND: Anopheles gambiae s.s. and An. coluzzii are two of the most important malaria vector species in sub-Saharan Africa. These recently-diverged sibling species do not exhibit intrinsic post-zygotic barriers to reproduction and are thought to be separated by strong assortative mating combined with selection against hybrids. At present, little is known about the ecological conditions that determine hybridization and introgression between these cryptic taxa. METHODS: Swarm segregation and assortative mating were studied in Western Burkina Faso in the villages of Vallée du Kou (VK7) and S…

Mapping Plasmodium transitions and interactions in the Anopheles female.

The human malaria parasite, Plasmodium falciparum, relies exclusively on Anopheles mosquitoes for transmission. Once ingested during blood feeding, most parasites die in the mosquito midgut lumen or during epithelium traversal1. How surviving ookinetes interact with midgut cells and form oocysts remains poorly understood, yet these steps are essential to initiate a remarkable growth process culminating in the production of thousands of infectious sporozoites2. Here, using single-cell RNA sequencing of both parasites and mosquito cells across different developmental stages and metabolic conditi…

A systematic review: is Anopheles vagus a species complex?

BACKGROUND: Anopheles vagus (subgenus Cellia) has been identified as a vector for malaria, filariasis, and Japanese encephalitis in Asia. Sporozoites of Plasmodium falciparum and Plasmodium vivax have been found in this zoophilic mosquito in Asia and Indonesia. This study systematically reviews publications regarding An. vagus species, variation, bio-ecology, and malaria transmission in various localities in Asia, especially Indonesia, to determine whether the current data support An. vagus as a species complex. METHODS: The databases Pubmed, Scopus, Europe PMC, and Proquest were searched to …

Swarms of the malaria vector Anopheles funestus in Tanzania.

BACKGROUND: Anopheles funestus mosquitoes currently contribute more than 85% of ongoing malaria transmission events in south-eastern Tanzania, even though they occur in lower densities than other vectors, such as Anopheles arabiensis. Unfortunately, the species ecology is minimally understood, partly because of difficulties in laboratory colonization. This study describes the first observations of An. funestus swarms in Tanzania, possibly heralding new opportunities for control. METHOD: Using systematic searches by community-based volunteers and expert entomologists, An. funestus swarms were …

Ecophysiology of Anopheles gambiae s.l.: persistence in the Sahel.

The dry-season biology of malaria vectors is poorly understood, especially in arid environments when no surface waters are available for several months, such as during the dry season in the Sahel. Here we reappraise results on the dry-season physiology of members of the Anopheles gambiae s.l. complex in the broad context of dormancy in insects and especially in mosquitoes. We examine evidence on seasonal changes in reproduction, metabolism, stress tolerance, nutrition, molecular regulation, and environmental conditions and determine if the current results are compatible with dry-season diapaus…

Is Anopheles mascarensis a new malaria vector in Madagascar?

Anopheles mascarensis De Meillon, 1947, a mosquito that is native to Madagascar, is reported for the first time to act as a vector of Plasmodium falciparum malaria. From September 1989 to March 1990, 2, 499 An. mascarensis specimens from different regions of Madagascar were tested by an enzyme-linked immunosorbent assay (ELISA), using monoclonal antibodies against the circumsporozoite (CS) protein of the four human species of Plasmodium. The salivary glands of 237 specimens were also dissected. Fourteen of 1,864 specimens obtained from Sainte Marie island on the Malagasy east coast were found …

Threshold of transmission of Brugia malayi by Anopheles sinensis.

In an area of central China where Brugia malayi is transmitted by Anopheles sinensis, three villages were followed for 4 years without any control measures. Diethylcarbamazine (DEC) had been used in a control programme reducing the parasite rate in Shuiwa from 12.72 to 0.59%, in Gubomen from 3.18 to 1.55% and in Moshi from 5.62 to 2.81% (although this fell the following year to 2.23%) up until the start of the trial. The village populations, the human biting rate, the parous and the gonotrophic cycle were comparable for all the areas, yet the microfilariae rate dropped to 0.18% in Shuiwa, 0.31…

Thioredoxin reductase from the malaria mosquito Anopheles gambiae.

Human pathogenic diseases received much attention recently due to their uncontrolled spread of antimicrobial resistance (AMR) which causes several threads every year. Effective alternate antimicrobials are urgently required to combat those disease causing infectious microbes. Halophilic actinobacteria revealed huge potentials and unexplored cultivable/non-cultivable actinobacterial species producing enormous antimicrobials have been proved in several genomics approaches. Potential gene clusters, PKS and NRPKS from Nocardia, Salinospora, Rhodococcus, and Streptomyces have wide range coding gene

Incrimination of Anopheles (Anopheles) intermedius Peryassú, An. (Nyssorhynchus) nuneztovari Gabaldón, An. (Nys.) oswaldoi Peryassú as natural vectors of Plasmodium falciparum in French Guiana.

Anopheles darlingi Root is the major vector of human malaria in the Neotropics and has been considered to be the sole malaria vector in French Guiana. The presence of other potential vectors suggests that malaria may be transmitted by other species under certain conditions. From 2006-2011, all anopheline specimens collected from 11 localities were assayed to determine if the Plasmodium circumsporozoite protein was present. In addition to An. darlingi, we found Anopheles oswaldoi, Anopheles intermedius and Anopheles nuneztovari specimens that were infected with Plasmodium sp. Further investigat…

mosGILT antibodies interfere with Plasmodium sporogony in Anopheles gambiae.

Plasmodium, the causative agents of malaria, are obtained by mosquitoes from an infected human. Following Plasmodium acquisition by Anopheles gambiae, mosquito gamma-interferon-inducible lysosomal thiol reductase (mosGILT) plays a critical role in its subsequent sporogony in the mosquito. A critical location for this development is the midgut, a tissue we show expresses mosGILT. Using membrane-feeding and murine infection models, we demonstrate that antibodies against mosGILT reduce the number of P. falciparum and P. berghei oocysts in the midgut and the infection prevalence of both species in…

Genomic diversity of the African malaria vector Anopheles funestus.

Anopheles funestus s.s. is a major human malaria vector across Africa. To study its evolution, especially under vector control pressure, we sequenced 656 modern specimens (collected 2014 to 2018) and 45 historic specimens (collected 1927 to 1967) from 16 African countries. Despite high genetic diversity, the species shows stable but considerable continental population structure. Although one population showed little differentiation over a century and 4000 kilometers, nearby, we found two genetically distinct ecotypes. Vector control has resulted in strong signals of selection, with some resist…

Sickle-trait hemoglobin does not influence Anopheles biting rates.

Children with sickle cell trait (HbAS) are protected against severe and symptomatic Plasmodium falciparum malaria. While several within-host resistance mechanisms have been investigated, it is unknown whether this protection may be attributable in part to reductions in exposure to P. falciparum parasites via mosquito bites. In a 15-month cohort in Western Kenya, we matched mosquito bloodmeals to human hosts based on short tandem repeat (STR) genotypes to determine individual mosquito biting rates. Using a multilevel multivariable model, we assessed mosquito biting behavior with respect to huma

Insights into malaria transmission among Anopheles funestus mosquitoes, Kenya.

BACKGROUND: Most malaria vectors belong to species complexes. Sibling species often exhibit divergent behaviors dictating the measures that can be deployed effectively in their control. Despite the importance of the Anopheles funestus complex in malaria transmission in sub-Saharan Africa, sibling species have rarely been identified in the past and their vectoring potential remains understudied. METHODS: We analyzed 1149 wild-caught An. funestus (senso lato) specimens from 21 sites in Kenya, covering the major malaria endemic areas including western, central and coastal areas. Indoor and outdo…
