Purified human C3 was biotinylated using the biotinyl-N-hydroxysuccinimide imidoester (BNHS). Depending on the input of BNHS, from three to six molecules of biotin were incorporated per C3 molecule. A procedure is described for preparing a polychelating agent-biotin conjugate through the interaction of biotin-maleimide with poly-L-lysine modified with multiple residues of diethylenetriaminepentaacetic acid and containing amino-terminal pyridylthio-propionate group. The conjugate can be easily loaded with multiple metal atoms, such as 111In, and preserves its ability to specifically interact with avidin (as avidin-agarose). Polychelate-biotin conjugate may be used for signal amplification in visualization and analytical protocols involving the application of the avidin-biotin system.

Tamavidin 2 is a biotin-binding protein of the avidin-like family, discovered in Tamogitake mushrooms, that has found a number of uses in biotechnology. Formalin-fixed, paraffin-embedded tissue samples of canine amelanotic melanomas and normal canine tissues were studied immunohistochemically for the presence of S100 protein. Use of the avidin-biotin complex procedure demonstrated variable amounts of S100 protein in the tumor cell cytoplasm and nuclei in 26 of 31 tumors.
