Bacteriocins from Limosilactobacillus and Ligilactobacillus: ecological logic, mechanistic diversity, and translational potential in the post-Lactobacillus taxonomy.

Bacteriocins are ribosomally synthesized antimicrobial peptides that contribute to microbial competition, niche establishment, and community structure. The 2020 taxonomic reorganization of the former broad Lactobacillus genus provides a useful framework for reinterpreting bacteriocin diversity in lineage-specific ecological contexts. This review focuses on bacteriocins produced by species now assigned to Limosilactobacillus and Ligilactobacillus, two host- and food-associated genera that include several reported bacteriocin producers. These genera were selected because they contain bacteriocin…

Lactic acid bacteria and their bacteriocins: promising food safety tools for controlling the foodborne pathogen Listeria monocytogenes.

Listeria monocytogenes is an important foodborne pathogen and is a common contaminant of raw milk and meat products. It is also found in man … tive method that involves the use of lactic acid bacteria strains and/or their metabolites, such as bacteriocins, to ensure the food safety of fermented foods without altering their organoleptic properties. Here, we discuss how lactic acid bacteria and their bacteriocins affect the proliferation of foodborne L. monocytogenes and spoilage microorganisms in cold …

Comprehensive analysis of bacteriocins produced by clinical enterococcal isolates and their antibacterial activity against Enterococci including VRE.

It is well-known that Enterococcus species produce bacteriocins that have antibacterial activity. However, a comprehensive analysis of the bacteriocin distribution among Enterococcus strains has not been conducted. In this study, we identified the bacteriocin genes from 80 Enterococcus faecalis and 38 Enterococcus faecium strains and investigated their antibacterial activity. In the 80 E. faecalis strains, the cytolysin gene (61.3%), enterolysin A gene (27.5%) and BacL1 gene (45.0%) were identified. In the 38 E. faecium strains, the enterocin A gene (97.4%), enterocin B gene (2.6%), enterocin …

Characterization of Enterococcus faecium E86 bacteriocins and their inhibition properties against Listeria monocytogenes and vancomycin-resistant Enterococcus.

In the present scenario of a major demand for new compounds with antimicrobial activity, bacteriocin and bacteriocin-like inhibitory substances (BLIS) are promising tools against deteriorating and pathogenic microorganisms, thus having potential applications in both the food industry and infectious disease control. In the present report, we describe the genetic and phenotypic characteristics of BLIS produced by Enterococcus faecium E86, a strain previously isolated and sequenced by our group, focusing on the structural genes of two bacteriocins identified: enterocin TW21 and enterocin P. Trans…

An enhanced split intein-mediated ligation (SIML) platform for rapid discovery and functional screening of circular bacteriocins.

Bacteriocins are ribosomally synthesized antimicrobial peptides with promising applications in biotechnology, particularly in food preservation and animal and human health. Circular bacteriocins are especially attractive due to their head-to-tail cyclized structure, which confers enhanced stability and antimicrobial potency relative to linear peptides. Here, we report an in vitro cell-free protein synthesis system coupled with an enhanced Split Intein-Mediated Ligation platform (IV-CFPS/SIML) for the efficient synthesis of circular bacteriocins through systematic evaluation of cyclization site…

Bacteriocins (meningocins) in Norwegian isolates of Neisseria meningitidis: possible role in the course of a meningococcal epidemic.

In the Neisseria meningitidis strain MC58 (serogroup B; ET-5 complex) genome three putative islands of horizontally transferred DNA (IHTs) have been identified. IHT-A2 codes for eight hypothetical proteins and two disrupted open reading frames with similarity to a secretion protein (NMB0097) and an ABC transporter (NMB0098). The strains MC58 and 44/76 (shown here) are meningocin resistant/weakly sensitive. None of these strains are meningocin producers. However, NMB0097 and NMB0098 homologues with open reading frames are found in meningocin producers (N. meningitidis P241 (serogroup A; systemi

Increasing prevalence of bacteriocin carriage in a six-year hospital cohort of E. faecium.

UNLABELLED: Vancomycin resistant enterococci (VRE) are important pathogens in hospitalized patients, however, the factors involved in VRE colonization of hospitalized patients are not well characterized. Bacteriocins provide a competitive advantage to enterococci in experimental models of colonization, but little is known about bacteriocin content in samples derived from humans and even less is known about their dynamics in the clinical setting. To identify bacteriocins which may be relevant in the transmission of VRE, we present a systematic analysis of bacteriocin content in the genomes of 2…

Bacteriocin-Producing Probiotic Lactic Acid Bacteria in Controlling Dysbiosis of the Gut Microbiota.

Several strains of lactic acid bacteria are potent probiotics and can cure a variety of diseases using different modes of actions. These bacteria produce antimicrobial peptides, bacteriocins, which inhibit or kill generally closely related bacterial strains and other pathogenic bacteria such as Listeria, Clostridium, and Salmonella. Bacteriocins are cationic peptides that kill the target cells by pore formation and the dissipation of cytosolic contents, leading to cell death. Bacteriocins are also known to modulate native microbiota and host immunity, affecting several health-promoting functio…

Bacteriocin-Like Inhibitory Substances from Probiotics as Therapeutic Agents for Candida Vulvovaginitis.

Probiotics can potentially prevent and treat diseases. We examined the inhibitory activity of bacteriocin-like inhibitory substances (BLISs) from potentially probiotic lactobacilli and streptococci on Candida albicans and non-Candida albicans clinical isolates from women with vulvovaginitis. Using agar well diffusion assays, BLISs inhibited both Candida albicans and non-Candida albicans isolates. The BLIS from L. pentosus isolates had the highest anti-Candida activity (33/45; 73.3%), followed by BLISs from isolates of L. paracasei subsp. paracasei (31/45; 68.9%), L. rhamnosus I (30/45; 66.7%),

Partial Characterization of an Anti-Listerial Bacteriocin from Enterococcus faecium CJNU 2524.

Listeria monocytogenes is a representative foodborne pathogen and causes listeriosis. Enterococcus faecium CJNU 2524 was confirmed to produce a bacteriocin with anti-listerial activity. To establish optimal culture conditions for the production of the bacteriocin from E. faecium CJNU 2524, different media (MRS and BHI broth) and temperatures (25°C, 30°C, and 37°C) were investigated. The results showed that the optimal culture conditions were MRS broth and 25°C or 30°C temperatures. The crude bacteriocin was stable in a broad range of pH conditions (2.0-10.0), temperatures (60°C-100°C), and org

Control of Propionibacterium acnes by natural antimicrobial substances: Role of the bacteriocin AS-48 and lysozyme.

We report the high susceptibility of several clinical isolates of Propionibacterium acnes from different sources (skin, bone, wound exudates, abscess or blood contamination) to the head-to-tail cyclized bacteriocin AS-48. This peptide is a feasible candidate for further pharmacological development against this bacterium, due to its physicochemical and biological characteristics, even when it is growing in a biofilm. Thus, the treatment of pre-formed biofilms with AS-48 resulted in a dose- and time-dependent disruption of the biofilm architecture beside the decrease of bacterial viability. Furt

Effects of the bacteriocin PsVP-10 produced by Pseudomonas sp. on sensitive bacterial strains.

The bacteriocin PsVP-10 is a 2.6 Kda peptide which was isolated and purified from Pseudomonas sp. This bacteriocin possesses lethal activity over Enterococcus faecalis, Salmonella typhimurium and Shigella flexneri. The experimental assays showed that the bacteriocin is able to be adsorbed by all cells of these bacterial species and also by their isolated cell walls. It was observed that the resistant mutants and their respective cell walls are unable to adsorb the bacteriocin. Assays performed with spheroplasts obtained from sensitive bacterial species and their resistant mutants show a rapid
