Intracellular Ca2+ and the phospholipid PIP2 regulate the taste transduction ion channel TRPM5.

The transduction of taste is a fundamental process that allows animals to discriminate nutritious from noxious substances. Three taste modalities, bitter, sweet, and amino acid, are mediated by G protein-coupled receptors that signal through a common transduction cascade: activation of phospholipase C beta2, leading to a breakdown of phosphatidylinositol-4,5-bisphosphate (PIP2) into diacylglycerol and inositol 1,4,5-trisphosphate, which causes release of Ca2+ from intracellular stores. The ion channel, TRPM5, is an essential component of this cascade; however, the mechanism by which it is acti

Studies on the mechanism of passive cation fluxes activated by deoxygenation of sickle cells.

Sickle cells exhibit a striking increase in monovalent cation content upon deoxygenation. These deoxy cation fluxes are reversible upon reoxygenation, balanced (Na flux = K flux), activated under physiologic conditions of pH and PO2, and insensitive to ouabain and furosemide. Our recent evidence indicates that deoxy cation fluxes and the Na/K pump, acting in concert, contribute to cation depletion in sickle cells in vitro. The resultant dehydration worsens their rigidity and increases their tendency to sickle upon deoxygenation. Here we report our finding that deoxy cation fluxes are inhibited

Cations reduce antimicrobial efficacy of lysozyme-chelator combinations.

Reduction of the antimicrobial efficacy of lysozyme-chelator combinations against two Escherichia coli O157:H7 strains on addition of mineral salts was studied. The objective of the study was to determine the effect of type and concentration of mono-, di-, and trivalent mineral salts on the antimicrobial effectiveness of lysozyme and various chelators against E. coli O157:H7. Seven salts (Al3+, Ca2+, Fe2+, Fe3+, K+, Mg2+, and Na+) at 1 to 10 mM were added to aqueous solutions of lysozyme and disodium ethylenediamine tetraacetic acid (EDTA), disodium pyrophosphate (DSPP), or pentasodium tripoly

A Coppoborylene Stabilized by Multicenter Covalent Bonding and Its Amphoteric Reactivity to CO.

A cationic copper-stabilized coppoborylene was prepared and structurally characterized via infrared photodissociation spectroscopy and density functional theory calculations. This structure exemplifies a new class of borylenes stabilized by three-center-two-electron metal-boron-metal covalent bonding interaction, displaying exceptional σ-acidity and unparalleled π-donor capability for CO activation that outperforms all of the known transition metal cations and is comparable or even superior to the documented base-trapped borylenes. Its neutral form represents a monovalent boron compound with a

Qualitative evaluation of maternal milk and commercial infant formulas via LIBS.

This study focuses on the use of laser-induced breakdown spectroscopy (LIBS) for the evaluation of the nutrients in maternal milk and some commercially available infant formulas. The results of such evaluation are vital for adequate and healthy feeding for babies during lactation period. Laser-induced breakdown spectroscopy offers special advantages in comparison to the other conventional analytical techniques. Specifically, LIBS is a straightforward technique that can be used in situ to provide qualitative analytical information in few minutes for the samples under investigation without prepa

Structure of the potassium form of CGCGAATTCGCG: DNA deformation by electrostatic collapse around inorganic cations.

The potassium form of d(CGCGAATTCGCG) solved by X-ray diffraction to 1.75 A resolution indicates that monovalent cations penetrate the primary and secondary layers of the "spine of hydration". Both the sodium [Shui, X., McFail-Isom, L., Hu, G. G., and Williams, L. D. (1998) Biochemistry 37, 8341-8355] and the potassium forms of the dodecamer at high resolution indicate that the original description of the spine, only two layers deep and with full occupancy by water molecules, requires substantive revision. The spine is merely the bottom two layers of a four layer solvent structure. The four la

Dynamic high-resolution computer simulation of electrophoretic enantiomer separations with neutral cyclodextrins as chiral selectors.

GENTRANS, a comprehensive one-dimensional dynamic simulator for electrophoretic separations and transport, was extended for handling electrokinetic chiral separations with a neutral ligand. The code can be employed to study the 1:1 interaction of monovalent weak and strong acids and bases with a single monovalent weak or strong acid or base additive, including a neutral cyclodextrin, under real experimental conditions. It is a tool to investigate the dynamics of chiral separations and to provide insight into the buffer systems used in chiral capillary zone electrophoresis (CZE) and chiral isot

Rubidium-82 myocardial uptake and extraction after transient ischemia: PET characteristics.

The effects of transient regional ischemia on monovalent cation uptake and extraction by the myocardium were studied in seven open chest dogs. Following a 10-20% stenosis of the left anterior descending coronary artery to prevent reactive hyperemia, the regional myocardial uptake of the monovalent cation 82Rb (ml/min/g X extraction) was measured before, during, and at 10, 30, and 60-80 min after a 10 min total occlusion of the left anterior descending coronary artery using positron emission tomography. Regional myocardial blood flow was also measured at the same time interval using radiolabele

Alkali Metal Mediated Redox Tuning in Bis(μ-OH) Dinickel(II) Complexes.

Redox-inactive metal ions act in tandem with redox-active transition metals in biological, molecular, and solid-state systems to precisely tune reactivity. Notwithstanding, mildly Lewis acidic alkali cations are rarely studied in this context, despite their prevalence in both cellular and electrochemical media. Herein, we disclose a structurally characterized series of monovalent Lewis acids (Li+, Na+, K+, and Cs+) adducted to a bis(μ-OH) dinickel diamond core. Spectroscopic characterization demonstrates that the oxygenic binding pocket of the tripodal NNNO-chelate maintains this structure in

Disease-associated missense mutations in the pore loop of polycystin-2 alter its ion channel function in a heterologous expression system.

Polycystin-2 (PC2) is mutated in ∼15% of patients with autosomal dominant polycystic kidney disease (ADPKD). PC2 belongs to the family of transient receptor potential (TRP) channels and can function as a homotetramer. We investigated whether three disease-associated mutations (F629S, C632R, or R638C) localized in the channel's pore loop alter ion channel properties of human PC2 expressed in Xenopus laevis oocytes. Expression of wild-type (WT) PC2 typically resulted in small but measurable Na+ inward currents in the absence of extracellular divalent cations. These currents were no longer observ

Adsorption of reovirus to clay minerals: effects of cation-exchange capacity, cation saturation, and surface area.

The adsorption of reovirus to clay minerals has been reported by several investigators, but the mechanisms defining this association have been studied only minimally. The purpose of this investigation was to elucidate the mechanisms involved with this interaction. More reovirus type 3 was adsorbed, in both distilled and synthetic estuarine water, by low concentrations of montmorillonite than by comparable concentrations of kaolinite containing a mixed complement of cations on the exchange complex. Adsorption to the clays was essentially immediate and was correlated with the cation-exchange cap

Electrostatic shielding chemistry enabling reversible four-electron aqueous zinc-iodine batteries.

A divalent organic cation-mediated electrolyte provides stronger confinement of polyiodide species and interhalogen intermediates than its monovalent analogue, promoting reversible Br--assisted four-electron iodine redox while suppressing side reactions and regulating Zn deposition. Consequently, aqueous Zn-I2 batteries exhibit high capacity, enhanced rate capability and stable cycling over 2500 cycles.

Effect of metal ion exchange on the photocurrent response from bacteriorhodopsin on tin oxide electrodes.

The transient photocurrent response from bacteriorhodopsin (bR) on tin oxide electrodes was strongly influenced by metal ions bound to bR molecules. The photocurrent polarity reversal pH, which corresponded to the pH value for the reversal of the proton release/uptake sequence in the bR photocycle, of cation-substituted purple membrane (PM) was shifted to lower pH with the increase in the cation affinities to carboxyl groups and a close correlation was noted between the two values. This suggests that the metal ion present in the extracellular region of a bR molecule modulates the pK(a) of prot

The renal cGMP-gated cation channel: its molecular structure and physiological role.

Cyclic nucleotide-gated cation channels, which are permeable to monovalent and divalent cations, are expressed in a number of tissues. cDNAs encoding cGMP-gated cation channel subunits have been cloned in retinal rods, cones, olfactory neuroepithelium, pineal gland, aorta, testis, heart, and most recently kidney. Patch clamp studies have identified and characterized cGMP-gated cation channels in the cortical collecting duct (CCD) and inner medullary collecting duct (IMCD). cGMP-gated cation channels in kidney share many biophysical and molecular properties with the retinal rod cGMP-gated chann

Mechanistic evaluation of factors affecting compound loading into ion-exchange fibers.

Donnan theory was applied to gain mechanistic understanding on the factors affecting drug loading process, compound-fiber affinity and subsequent release from fibrous ion-exchangers. Impact of initial loading solution concentration on fiber occupancy and loading efficiency of compounds were assessed experimentally and theoretically. Relative affinity towards the anion-exchange fibers was studied by dual loading of monovalent salicylic acid and either more lipophilic 3-isopropylsalicylic acid or divalent 5-hydroxyisophthalic acid. The effect of fiber framework on compound binding was evaluated

Ion selectivity of scorpion toxin-induced pores in cardiac myocytes.

The lytic activity of parabutoporin (PP) and opistoporin 1 (OP1) on mammalian and bacterial membranes have been described. We investigated pore-formation and ion selectivity in cardiac myocytes by measuring the whole cell leak current by means of the patch clamp technique. Pore formation was observed as the induction of leak currents. Ion selectivity of the pores was indicated by the shift of the reversal potential (E(rev)) upon substitution of intra- and extra-cellular ions. Results were compared with the effect of gramicidin A (gramA). PP and OP1 induced a fluctuating leak current and indica

Role of cations as components of jelly coats in Bufo arenarum fertilization.

The role of monovalent (Na+, K+) and divalent (Ca2+, Mg2+) cations in Bufo arenarum fertilization was analysed. Our results showed that the highest fertilization percentages were obtained when strings of uterine oocytes (UO) were inseminated. Under these conditions, full jelly (FJ), which represents the jelly coats surrounding the oocytes at the time of deposition, contained 68.5 +/- 7.0 mM Na+, 27.4 +/- 2.4 mM K+, 6.3 +/- 0.9 mM Ca2+ and 6.9 +/- 0.9 mM Mg2+. When the strings of oocytes were washed in deionized water, these cations diffused into the liquid medium surrounding them. There was a
