Burkholderia pseudomallei induces cell fusion and actin-associated membrane protrusion: a possible mechanism for cell-to-cell spreading.

Burkholderia pseudomallei, a facultative intracellular bacterium, is the causative agent of a broad spectrum of diseases collectively known … ferent cell markers (CMFDA or CMTMR), mixed, and then infected with B. pseudomallei, direct cell-to-cell fusion could be observed, leading to MNGC formation. Staining of the infected cells with rhodamine-conjugated phalloidin indicated that immediately after the infection, actin rearrangement into a comet tail appearance occurred, similar to that described ea …

Tin oxide nanowires suppress herpes simplex virus-1 entry and cell-to-cell membrane fusion.

The advent of nanotechnology has ushered in the use of modified nanoparticles as potential antiviral agents against diseases such as herpes simplex virus 1 and 2 (HSV-1) (HSV-2), human immunodeficiency virus (HIV), monkeypox virus, and hepatitis B virus. Here we describe the application of tin oxide (SnO(2)) nanowires as an effective treatment against HSV-1 infection. SnO(2) nanowires work as a carrier of negatively charged structures that compete with HSV-1 attachment to cell bound heparan sulfate (HS), therefore inhibiting entry and subsequent cell-to-cell spread. This promising new approach

LIN-39/Hox triggers cell division and represses EFF-1/fusogen-dependent vulval cell fusion.

General mechanisms by which Hox genes establish cell fates are known. However, a few Hox effectors mediating cell behaviors have been identified. Here we found the first effector of LIN-39/HoxD4/Dfd in Caenorhabditis elegans. In specific vulval precursor cells (VPCs), LIN-39 represses early and late expression of EFF-1, a membrane protein essential for cell fusion. Repression of eff-1 is also achieved by the activity of CEH-20/Exd/Pbx, a known cofactor of Hox proteins. Unfused VPCs in lin-39(-);eff-1(-) double mutants fail to divide but migrate, executing vulval fates. Thus, lin-39 is essentia…

Establishment of a ZFP64::NCOA3 fusion-positive spindle cell/sclerosing rhabdomyosarcoma organoid model and evaluation of fusion-gene dependency.

Spindle cell/sclerosing rhabdomyosarcoma (SRMS) comprises a heterogeneous group of tumors characterized by distinct molecular alterations and clinical behaviors. Among them, ZFP64::NCOA3 fusion-positive SRMS represents an extremely rare subtype, and its biological characteristics remain poorly understood because of the lack of representative experimental models. In this study, we established a patient-derived organoid (PDO) line, designated OICI-RMS-1275, together with organoid-derived xenograft (ODX) models from a previously reported case of ZFP64::NCOA3 fusion-positive SRMS using a modified

Recombinant vesicular stomatitis virus expressing respiratory syncytial virus (RSV) glycoproteins: RSV fusion protein can mediate infection and cell fusion.

The genes encoding the respiratory syncytial virus (RSV) attachment (G) and fusion (F) envelope glycoproteins were expressed separately as additional genes in recombinant vesicular stomatitis viruses (VSV). Cells infected with the VSV-RSV F recombinant formed large syncytia illustrating the fusion activity of F in absence of other RSV proteins. Both F and G glycoproteins were expressed at the cell surface and incorporated into virions. Incorporation of these proteins did not require cytoplasmic tail sequences of VSV G. Using a compound, ammonium chloride, that raises the endosomal pH, we showe

[Expression of ATR-Fc fusion protein in CHO cells].

ATR-Fc is a fusion protein consisting of extracellular domain of human anthrax toxin receptor (ATR) and a fragment (hinge, CH2, and CH3 domains) of the Fc of human IgG1. The aim of ATR-Fc expression is to get an antibody-like molecule binding to protective antigen (PA), a component of anthrax toxins, this fusion protein may compete with cell surface receptor for PA binding, and block the transport of lethal factor (LF) and edema factor (EF) into cells, thereby act as an antitoxin to prevent and treat anthrax infection. A DNA fragment encoding N-terminal amino acids 1-227 of ATR and human IgG1

Atypical germ cells in a child with splenic-gonadal fusion.

Splenic-gonadal fusion is a rare congenital anomaly associated with other anomalies, particularly limb defects. This is the first reported case of splenic-gonadal fusion, tetraperomelia (congenital malformations involving all 4 extremities), and spinal abnormality with neurogenic bladder, in which focal atypia of germ cells was found following orchiectomy. Testicular biopsy with frozen section examination should be performed if the diagnosis of splenic-gonadal fusion is made, especially if associated with cryptorchidism and an orchiopexy is planned.

ABL1 fusions in T-cell acute lymphoblastic leukemia.

Mutant tyrosine kinases are common in cancer and can be therapeutically targeted with kinase inhibitors. To obtain insight in the contribution of activated kinases to the pathogenesis ofT-cell acute lymphoblastic leukemia (T-ALL), we studied the NUP214-ABL1 fusion gene that is found in 6% of T-ALL and EML1-ABL1 that we identified in one T-ALL patient. NUP214-ABL1 and EML1-ABL1 display constitutive kinase activity and are sensitive to the kinase inhibitor imatinib. Both proteins transform hematopoietic cells and we established mouse models of EML1-ABL1 and NUP214-ABL1 induced T-ALL. Interesting

A mathematical model for temperature effects on virus-mediated cell fusion.

Syncytia are formed by fusion of multiple cells into a multi-nucleated cell body. Viral-induced syncytia formation is used by certain viruses to propagate without leaving the cell. The ability to propagate without leaving the cell allows viruses to enhance replication and evade some of the host's immune responses. Currently, it is not known how temperature affects syncytia formation rate. This study uses a cell-to-cell fusion mathematical model to analyze data from previously published cell-cell fusion assays of HIV and SARS-CoV-2 at various temperatures. Parameters were estimated via minimiza…

Sickle Cell Disease Association with Premature Suture Fusion in Young Children.

UNLABELLED: Sickle cell disease (SCD) leads to the formation of an atypical hemoglobin tetramer with reduced capacity to carry oxygen. Although correlation between SCD and craniosynostosis (CS) has been mentioned, these are mostly small series or case reports. This article aimed to study any correlation between these entities in a large pediatric population. METHODS: We retrospectively reviewed head CT scans of SCD patients from 0 to 8 years of age who required a CT for issues unrelated to their head shape between 2012 and 2020. We excluded patients with known history of CS or any CS-related

Novel EWSR1::UBP1 fusion expands the spectrum of spindle cell rhabdomyosarcomas.

Over the last decade, the development of next-generation sequencing techniques has led to the molecular dismantlement of adult and pediatric sarcoma, with the identification of multiple gene fusions associated with specific subtypes and currently integrated into diagnostic classifications. In this report, we describe and discuss the identification of a novel EWSR1-UBP1 gene fusion in an adult patient presenting with multi-metastatic sarcoma. Extensive pathological, transcriptomic, and genomic characterization of this tumor in comparison with a cohort of different subtypes of pediatric and adul

Primary retroperitoneal renal cell carcinoma associated with transcription EB gene fusion.

BACKGROUND: Renal cell carcinoma (RCC) with the distinct type of t(6;11) (p21;q12) translocation (transcription factor EB, TFEB) is a rare neoplasm. It is even less when talks about primary retroperitoneal TFEB RCCs. To our knowledge, no previous literature has been reported about this kind of RCCs. In this article, we report a case of primary retroperitoneal renal cell carcinoma associated with transcription EB gene fusion. METHODS: A 73-year-old male patient presented with a retroperitoneal mass for more than one month. RESULTS: Pathologically, the mass was soft and colorful, tumor cells s

Pediatric renal cell carcinoma associated with Xp11.2 translocation/TFE3 gene fusion.

Renal cell carcinoma (RCC) in children and young adults is rare and pathologically problematic. RCC can be either hereditary or sporadic and has a guarded prognosis because appropriate management has not been established. A case of RCC in an 11-year-old is reported. The clinical presentation was a right abdominal mass, hematuria, urinary tract infection, and wasting. Radio-logically, the mass was found within the right kidney with calcification and paraaortic lymphadenopathy. The postsurgical diagnosis was Wilms' tumor stage T4N2M0. On gross inspection, the tumor was ill defined, extending acr

Cell-mediated immune responses to MUC1 fusion protein coupled to mannan.

The immunotherapy of cancer requires the definition of a suitable target for and the induction of a CD8+ cytotoxic lymphocyte reaction. In breast cancer, particularly mucins (MUC1) of the variable number of tandem repeat sequence may be a suitable target, but there has been a problem in inducing a cytotoxic response. MUC1 peptides, conjugated to carriers (keyhole-limpet hemocyanin or diphtheria toxoid) induce a humoral response and give poor tumor protection in mice and there is little cellular immunity. However, when MUC1 fusion protein is conjugated to mannan under oxidizing conditions, a ce

[Expression of an Epstein-Barr virus early antigen and cell fusion].

Superinfection of Raji cells with Epstein-Barr virus (EBV) derived from P3HR-1 (P3H-EBV) leads to syncytium formation. Studies using inhibitors of DNA and protein synthesis suggest that the fusion-inducing factor belongs to the early antigen of virus-specified proteins. Induction of early antigen (EA) in Raji cells by using various chemicals also leads to syncytium formation. However, A2L/AH hybrid cells, which were prepared by fusion between epithelial cells of adenoid origin and lymphocytes transformed by B95-8 virus, could not be superinfected with P3H-EBV. But, the syncytium formation was

RET fusion partners dictate oncogenic potential in undifferentiated spindle cell sarcomas.

BACKGROUND: Spindle cell tumors with RET rearrangements exhibit variable clinical behaviors, ranging from benign to highly aggressive malignancies. The underlying heterogeneity is suspected to be associated with distinct fusion partner genes. Understanding the impact of these fusion partners on oncogenic potential is crucial for precision therapy. METHODS: We report a case of a spindle cell tumor harboring an MYH10::RET fusion, which initially responded to anti-RET therapy but relapsed due to an NTRK1 fusion. Isogenic cell lines expressing MYH10::RET and CCDC6::RET were established. Functiona
