A rabbit model for evaluation of chlorpromazine-induced orthostatic hypotension.

The present study was conducted to develop an experimental model for evaluation of chlorpromazine-induced orthostatic hypotension in rabbits. In addition, the alpha-adrenoceptor blocking effect of chlorpromazine was investigated in isolated rabbit aorta and saphenous vein in comparison with prazosin. Chlorpromazine (0.1 and 1 mg/kg, i.v.) potentiated significantly a decrease in mean blood pressure at 1 min after the onset of head-up tilt in rabbits anesthetized with urethane alone, urethane+alpha-chloralose or nitrous oxide alone, but not in conscious and morphine+urethane+alpha-chloralose-ane…

Peripheral vascular responses to hyperthermia in the rat.

To investigate the sequence and nature of the peripheral vascular responses during the prodromal period of heat stroke, rats were implanted with Doppler flow probes on the superior mesenteric (SMA), left iliac (LIA) or left renal (LRA), and external caudal (ECA) arteries. Studies were performed in unanesthetized rats (n = 6) exposed to 46 degrees C and in chloralose-anesthetized animals (n = 11) at 40 degrees C. Core (Tc) and tail-skin temperatures, heart rate, and mean arterial blood pressure (MAP) were also monitored. In both groups, prolonged (70-150 min) exposure progressively elevated Tc …

Responses to bladder distension were not observed after the administration of chloralose anesthesia. We conclude that after cervical spinal transection the rat exhibits autonomic hyperreflexia similar to that seen in humans with spinal injury. Furthermore, autonomic hyperreflexia is completely established within 24 h after CST in the ra …

Naloxone or haloperidol but not yohimbine reverse apomorphine-induced respiratory depression.

Intravenous injection of low doses of naloxone was found to reverse the respiratory depression induced by apomorphine in chloralose-anesthesized dogs. Similar results were obtained with haloperidol, whereas yohimbine remained ineffective. These data suggest that apomorphine depresses respiratory rate through a mechanism involving both dopamine and opiate mechanisms but not alpha 2-adrenoceptors. Respiratory arrest induced by overdosages of apomorphine can be treated with naloxone.

Electrolyte changes in plasma and urine during tender coconut water infusion in dogs.

Intravenous infusion of tender coconut water to paraldehyde and chloralose-urethane anaesthetised dogs at a rate of 0.5 ml/kg/min to a total dose of 100 ml/kg body weight decreased serum Na and increased K and Ca levels. Simultaneously, urinary Na excretion decreased and K and Ca excretion increased and there was glycosuria. Serum electrolyte changes were more prominent in paraldehyde anaesthetised dogs.

Cardiovascular depressant effects of the kappa opioid receptor agonists U50488H and spiradoline mesylate.

The cardiovascular effects of the selective kappa opioid receptor agonists U50488H and spiradoline mesylate were examined in pentobarbital-anesthetized mongrel dogs and chloralose-anesthetized cats. In the dog studies, U50488H (0.01-3.0 mg/kg, i.v.) produced a dose-related depression in mean arterial pressure (MAP), left ventricular systolic pressure (LVSP), left ventricular dp/dt, and heart rate. These effects were completely reversed by a 1 mg/kg i.v. dose of the opioid receptor antagonist naloxone. A second kappa agonist, spiradoline mesylate, also produced a naloxone-reversible cardiovascu…

Pharmacological observations on the hypotensive action of extracts of teleost fish Urophyses (urotensin I) in the rat.

1 Intravenous injections of urotensin I regularly caused a long-lasting, dose-related, lowering of blood pressure and an increase in heart rate in conscious rats, or a reduction in perfusion pressure in the isolated hind limb of the rat.2 After subcutaneous administration, the hypotensive effect of urotensin I was greater in extent and in duration (> 24 hours).3 Anaesthesia with ether, chloralose, pentobarbitone and thiobarbitone caused a decrease in blood pressure and only slightly diminished the hypotensive effect of urotensin.4 Mecamylamine, hexamethonium, atropine, phenoxybenzamine, propra…

Interactions between the neuromuscular blocking drug Org NC 45 and some anaesthetic, analgesic and antimicrobial agents.

The effects of some anaesthetics and other agents used in anaesthetic practice on the neuromuscular blocking activity of cumulative doses of Org NC 45 have been studied in cats anaesthetized with chloralose. As expected from studies with other neuromuscular blocking drugs, both inhalation and i.v. anaesthetics reduced the doses of Org NC 45 required for neuromuscular blockade. An apparently specific potentiating interaction between Org NC 45 and metronidazole was observed.

[Opiate component in the realization of the vascular effects of clonidine].

Systemic and regional hemodynamic changes (microsphere technique) were studied in urethan-chloralose anesthetized cats after intravenous clonidine and naloxone administration in different sequence. It was shown that antihypertensive effect of clonidine was caused by a decrease in the cardiac output and an increase in the total peripheral vascular conduction. The ability of clonidine to increase total peripheral conduction was completely blocked with naloxone preadministration. It was concluded that endogenous opiate receptors take part in the vasodilating clonidine effect in abdominal organs.

Drug administration into the third ventricle of the brain of anesthetized and conscious cats.

A method is described for the administration of drugs into the third ventricle of the brain of conscious and anesthetized cats. Noradrenaline administered by this route produced cardiovascular stimulant effects as against depressor effects when administered into a lateral cerebral ventricle. The pressor responses evoked by third ventricular administration of noradrenaline were antagonized by spinal section in anesthetized cats and by hexamethonium in conscious animals. Conscious cats were more sensitive to the pressor effects of third ventricular noradrenaline than chloralose-anesthetized anim…

Structural analysis of heart rhythm spectrogram in narcotized cats.

Eight types of peaks were revealed in the cardiac rhythm spectrum during acute experiments on vagotomized cats. Some peaks had no physiological nature and resulted from specificity of ECG processing by Fourier analysis, while others reflected myogenic reaction of the sinoatrial node (length-dependence of automaticity) to changes in venous return caused by respiratory-induced and other variations of the blood flow in the cardiovascular system.

EEG in anaesthetized rats.

The constancy of rat EEG during surgical anaesthesia induced by phenobarbital, cholorose, paraldehyde, and inactin was investigated. The results showed that intraindividual variations and spontaneity of EEG were not abolished in the anaestheized animals. However, at the higher of the two doses used, the EEG was less variable. Our findings thus indicate that in the study of drug actions on the EEG of anaesthetized animals, the results obtained not only represent the interaction of the anaesthetic and the drug on the animal EEG but is also a reflection of the dosage of the anaesthetic employed.

Lack of effect by substance P at sites in the substantia gelatinosa where Met-enkephalin reduces the transmission of nociceptive impulses.

In alpha-chloralose anaesthetized spinal cats, Met-enkephalin amide (M-ENKA) and substance P were administered electrophoretically in the substantia gelatinosa while studying the excitation of deeper spinal neurones by noxious and non-noxious cutaneous stimuli. At sites where Met-enkephalin selectively reduced excitation by noxious skin stimuli, substance P was without effect. This result does not support the hypothesis that enkephalins are released at axo-axonic synapses on the terminals of substance P releasing primary afferent fibres.

Central indomethacin enhances volume-dependent vasopressin release.

The effect of centrally administered indomethacin on hemorrhage-induced vasopressin release was studied in the morphine-sedated, urethan/chloralose-anesthetized dog. Ventriculocisternal perfusion of indomethacin 1) significantly reduced the amount of prostaglandin E2 in the effluent from the cisterna magna, 2) significantly enhanced the vasopressin response to volume depletion, and led to a greater fall in mean arterial blood pressure during severe hemorrhage. The results suggest that central prostaglandins may have an inhibitory effect on vasopressin secretion during volume depletion.

Suppression of ouabain-induced atrial arrhythmias by carotid sinus stimulation.

In dogs anaesthetized with chloralose ectopic atrial arrhythmias were produced by subepicardial injection of ouabain. Stimulation of the right carotid sinus abruptly suppressed the ectopic arrhythmias. They returned on cessation of stimulation and sometimes already during the period of stimulation. It is suggested that in view of its response to carotid sinus stimulation the ouabain-induced arrhythmia resembles paroxysmal atrial tachycardia and that it is distinct from the aconitine-induced arrhythmia which on account of its response to vagal stimulation has been classified as atrial flutter.

The impact of four different classes of anesthetics on the mechanisms of blood pressure regulation in normotensive and spontaneously hypertensive rats.

Most anesthetics induce characteristic hemodynamic changes leading to blood pressure (BP) reduction but the role of renin-angiotensin system (RAS), sympathetic nervous system (SNS) and nitric oxide (NO) synthesis in this BP reduction is unknown. We therefore studied the influence of four widely used anesthetics - pentobarbital (P), isoflurane (ISO), ketamine-xylazine (KX) and chloralose-urethane (CU) - on the participation of these vasoactive systems in BP maintenance. BP effects elicited by the acute sequential blockade of RAS (captopril), SNS (pentolinium) and NO synthase (L-NAME) were compa…
