Comparative developmental and maternal neurotoxicity following acute gestational exposure to chlorpyrifos in rats.

Chlorpyrifos (CPF), an organophosphorus (OP) insecticide, exerts toxicity through inhibition of acetylcholinesterase (AChE). In the present study, pregnant Sprague-Dawley rats were given CPF (200 mg/kg, sc) as a single dose on gestation d 12 (GD12) and then sacrificed on either GD16, GD20, or postnatal d 3 (PND3) for measurement of maternal and developmental indicators of toxicity. While most CPF-treated rats exhibited no overt signs, a subset (4/28) showed moderate to severe signs of "cholinergic" toxicity at 2-3 d after treatment, and these rats were omitted from further studies. Mechanistic insights into chlorpyrifos-induced nephrotoxicity revealed by network toxicology and molecular docking.

Chlorpyrifos (CPF), a widely used organophosphorus pesticide, poses potential nephrotoxicity risks, yet its molecular mechanisms remain incompletely elucidated. This study integrated network toxicology, molecular docking, molecular dynamics (MD) simulations, and in vitro experiments to systematically explore CPF-induced renal injury mechanisms. ADMET and toxicity predictions indicated CPF's potential nephrotoxic risk. Through cross-database screening, 33 overlapping targets were identified, and seven core targets, MPO, XDH, TLR4, NFE2L2, NOS2, NOS3, and SOD2, were further selected via protein-

GC-MS/MS analysis of chlorpyrifos in forensic samples with varied survival time.

Organophosphorus pesticides are extensively used in the agricultural sector to kill insects, worms, and other pests. Many people may be poisoned by chlorpyrifos either accidentally or intentionally, including accidental, suicidal, and homicidal poisoning cases in India. The effect of chlorpyrifos on human health depends on factors such as the time, amount and frequency of exposure, the individual's health, and certain environmental conditions. The main objective of this investigation is to identify the post-mortem biological sample that shows the longest detection window, enabling precise chlo

Chlorpyrifos effects on integrin alpha v and beta 3 in implantation window phase.

Chlorpyrifos (CPF), as a worldwide pesticide, can effect on the integrins αv and β3 which play a main role in the implantation window. Therefore, the aim of this study was to consider CPF effects on integrin alpha v and beta 3 in implantation window phase. Thirty female NMRI mice were separated into groups of CPF, sham, and control. After 6 weeks, each group was mated, and on the 5th day of gestation, all mice were euthanized. Estradiol and progesterone levels were detected by the enzyme-linked immunosorbent assay (ELISA) test; two subunits of integrins (αv and β3) genes and proteins of endome

Seven-year neurodevelopmental scores and prenatal exposure to chlorpyrifos, a common agricultural pesticide.

BACKGROUND: In a longitudinal birth cohort study of inner-city mothers and children (Columbia Center for Children's Environmental Health), we have previously reported that prenatal exposure to chlorpyrifos (CPF) was associated with neurodevelopmental problems at 3 years of age. OBJECTIVE: The goal of the study was to estimate the relationship between prenatal CPF exposure and neurodevelopment among cohort children at 7 years of age. METHODS: In a sample of 265 children, participants in a prospective study of air pollution, we measured prenatal CPF exposure using umbilical cord blood plasma (

Chlorpyrifos exposure induces inflammatory and immune dysregulation: An integrative in silico and in vitro analysis.

Chlorpyrifos (CPF) is a widely used organophosphate insecticide that has been associated with chronic inflammatory and immune-related health effects. However, the molecular mechanisms underlying CPF-induced immune-inflammatory dysregulation remain incompletely defined. In this study, an integrated in silico and in vitro strategy was employed to elucidate the signaling pathways involved in CPF-mediated macrophage inflammatory activation. Network-based target prediction and pathway enrichment analysis identified MAPK14, which encodes p38 MAPK, and RELA, which encodes the NF-κB p65 subunit, as ce

An aptazyme driven dual-FRET nanoplatform for simultaneous detection of chlorpyrifos and lead ions.

The co-occurrence of pesticides and heavy metal ions in water resources poses significant health risks, necessitating the development of advanced detection methods capable of simultaneously monitoring these contaminants. Sometimes, traditional biosensors for multiple target detection may face challenges in detecting chemically distinct analytes due to their signal interference and interdependent detection mechanisms. To overcome these challenges, a simple dual-target optical biosensor was fabricated for sensitive and efficient measurement of chlorpyrifos and lead ions (Pb2+) by utilizing a För

An Aptasensor Based on a Flexible Screen-Printed Silver Electrode for the Rapid Detection of Chlorpyrifos.

In this work, we propose a novel disposable flexible and screen-printed electrochemical aptamer-based sensor (aptasensor) for the rapid detection of chlorpyrifos (CPF). To optimize the process, various characterization procedures were employed, including Fourier transform infrared spectroscopy (FT-IR), electrochemical impedance spectroscopy (EIS), and cyclic voltammetry (CV). Initially, the aptasensor was optimized in terms of electrolyte pH, aptamer concentration, and incubation time for chlorpyrifos. Under optimal conditions, the aptasensor showed a wide linear range from 1 to 105 ng/mL with

Thioglycolic acid-modified AuNPs as a colorimetric sensor for the rapid determination of the pesticide chlorpyrifos.

Herein, we report the preparation of a gold nanoparticle-thioglycolic acid (TGA@AuNP) suspension for the label-free colorimetric detection of the organophosphorus pesticide chlorpyrifos. This colorimetric sensor was synthesized by a one-pot method, providing thioglycolic acid (TGA)-modified gold nanoparticles with a negatively charged surface. The formation of TGA@AuNPs was confirmed by Fourier transform infrared spectroscopy (FTIR), transmission electron microscopy (TEM), and dynamic light scattering (DLS). In addition, chlorpyrifos was quantitatively determined by UV-vis spectrophotometry. T

Longitudinal assessment of occupational determinants of chlorpyrifos exposure in adolescent pesticide workers in Egypt.

Chlorpyrifos (CPF) is an organophosphourus insecticide applied to cotton fields by adolescents employed by the Egyptian Ministry of Agriculture. Urinary 3,5,6-trichloro-2-pyridinol (TCPy) is a biomarker of CPF exposure that has substantial variability among these applicators. In order to identify predictors of CPF exposure, we conducted a longitudinal study of 43 adolescent pesticide applicators in Egypt from April 2010 to January 2011 in Egypt. Urinary TCPy was quantified at 25 time-points, prior to, during, and following application. We used log-linear regression and a best subset selection

Effect of chlorpyrifos-methyl on steroid and thyroid hormones in rat F0- and F1-generations.

Chlorpyrifos-methyl (CPM) suppressed androgenic activity in Hershberger assay using castrated rats. Acute oral lowest-observed-adverse-effect-level (LOAEL) and no-observed-adverse-effect-level (NOAEL) was evaluated as 12 and 0.1 mg/kg bw, respectively, based on its major effect of cholinesterase inhibition. Also, repeated oral NOAEL was 0.1 mg/kg bw/day based on adrenal damage in rats. We investigated one-generation reproductive toxicity of CPM focusing on endocrine-disrupting effects by the administration of 1, 10 and 100 mg/kg bw/day CPM to mature SD rats (F0) through pre-mating, mating, ges

Lower birth weight as a critical effect of chlorpyrifos: a comparison of human and animal data.

Chlorpyrifos is an irreversible inhibitor of cholinesterase (ChE), and inhibition of ChE is believed to be the most sensitive effect in all animal species evaluated and in humans. Recent epidemiology studies reported associations between umbilical cord plasma chlorpyrifos levels and fetal birth weight decreases among minority women living in New York City during pregnancy. These associations raise questions whether impaired fetal development is the critical effect rather than the inhibition of ChE as is currently believed so. We analyze the available information from epidemiology studies and a

Alterations in serotonin transporter expression in brain regions of rats exposed neonatally to chlorpyrifos.

Chlorpyrifos (CPF), one of the most widely-used organophosphate pesticides, is a suspected neuroteratogen. We administered CPF to neonatal rats on postnatal days (PN) 1-4 (1 mg/kg) or PN11-14 (5 mg/kg), treatments devoid of overt toxicity. At the end of the treatment period (PN5 and 15, respectively) and 5-7 days later, we then examined the effects on paroxetine (PXT) binding to the presynaptic 5HT high-affinity transporter, a marker for serotonin (5HT) projections. In males, we found a persistent decrease in PXT binding across the two different treatment regimens, with deficits apparent in a

Thermoregulatory effects of chlorpyrifos in the rat: long-term changes in cholinergic and noradrenergic sensitivity.

Subcutaneous injection of a sublethal dose of chlorpyrifos (CHLP), an organophosphate (OP) pesticide, causes long-term inhibition in cholinesterase activity (ChE) of brain, blood, and other tissues. Such prolonged inhibition in ChE should lead to marked behavioral and autonomic thermoregulatory patterns, especially in terms of altered noradrenergic and cholinergic sensitivity. To evaluate the behavioral and autonomic effects of long-term ChE inhibition, Long-Evans rats were implanted with radiotelemetry transmitters that continuously monitored core temperature (Tc), heart rate (HR), and motor
