ABC protein CgABCF2 is required for asexual and sexual development, appressorial formation and plant infection in Colletotrichum gloeosporioides.

ATP-binding cassette (ABC) proteins are exclusively found in both prokaryotes and eukaryotes. In this study, we have characterized a gene from Glomerella leaf spot pathogen Colletotrichum gloeosporioides that encodes an ABC protein, whose function to date remains unknown. We designated this gene as CgABCF2. Deletion of CgABCF2 showed drastic reduction both growing rate and conidial production in C. gloeosporioides. The Δcgabcf2 mutant did not form the appressoria, lost the capability to infect apple and failed to form lesions on the wounded leaves and fruits. The C. gloeosporioides native CgAB…

Entomopathogenic activity of a variety of the fungus, Colletotrichum acutatum, recovered from the elongate hemlock scale, Fiorinia externa.

A fungal epizootic in populations of Fiorinia externa Ferris (Hemiptera: Diaspididae) infesting hemlock trees, Tsuga canadensis (L.) Carrière (Pinales: Pinaceae) in forests of the Northeastern US has been recently detected. The current known distribution of the epizootic spans 36 sites in New York, Pennsylvania, New Jersey and Connecticut. Colletotrichum acutatum Simmonds var. fioriniae Marcelino and Gouli var. nov. inedit. (Phyllachorales: Phyllachoraceae) was the most prevalent fungus recovered from infected scales. Bioassays indicated that this C. acutatum variety is highly pathogenic to F.…

Pathogenic characteristics and chemical control of soybean leaf anthracnose caused by Colletotrichum sojae newly found in Northeast China.

Soybean (Glycine max) leaf anthracnose, caused by diverse pathogenic agents, poses a significant threat to global soybean production. In 2025, a pathogenic fungus was isolated from symptomatic soybean leaves collected in a major soybean-producing region of China. Morphological and molecular characterization confirmed that the isolates belong to Colletotrichum sojae. To our knowledge, this is the first report of C. sojae causing soybean leaf anthracnose in Northeast China. Among 15 tested C. sojae isolates tested, moderately virulent strains were predominant, accounting for approximately 53.3%.…

Niemann-Pick Type C Proteins Are Required for Sterol Transport and Appressorium-Mediated Plant Penetration of Colletotrichum orbiculare.

Many biotrophic and hemibiotrophic fungal pathogens use appressoria to directly penetrate the host plant surface. In the cucumber anthracnose fungus Colletotrichum orbiculare, differentiation of appressoria requires a proper G1/S cell cycle progression, regulated by the GTPase-activating protein complex CoBub2-CoBfa1 and its downstream GTPase CoTem1. To explore the mechanisms by which the CoTem1 cascade regulates plant infection, we screened for CoTem1 interaction factors and identified a Niemann-Pick type C2 homolog (CoNpc2). Niemann-Pick type C proteins NPC1 and NPC2 are sterol-binding prote…

Ammonium secretion during Colletotrichum coccodes infection modulates salicylic and jasmonic acid pathways of ripe and unripe tomato fruit.

The postharvest pathogens Colletotrichum coccodes remains quiescent after infection of unripe fruit. However, during fruit ripening, the pathogen assumes a necrotrophic life style, rapidly colonizing the tissue. C. coccodes secretes ammonium during germination and colonization of host tissue that induces host programmed cell death. We further examined the role of ammonia in the infection process by analyzing transcriptome expression from infected and ammonia-treated fruit tissue compared with healthy tissue. The analysis revealed 82 and 237 common upregulated and downregulated genes, respectiv…

CgrBrlA, CgrAbaA, and CgrWetA form a developmental regulatory cascade controlling conidiation, spore integrity, and virulence in Colletotrichum graminicola.

Colletotrichum graminicola, the causal agent of maize anthracnose, relies on asexual spores for disease initiation and spread, yet the transcriptional mechanisms controlling conidiation and infection-related development remain incompletely understood. Here, we functionally characterize three putative homologs of the conserved fungal developmental regulators BrlA, AbaA, and WetA in C. graminicola. Targeted deletion analyses showed that CgrbrlA is essential for conidiophore formation and conidium production, while CgrabaA is required for proper conidiophore differentiation and normal sporulation…

A Positive Regulator CsPR10-9 Confers Resistance to Anthracnose (Colletotrichum gloeosporioides) Is Negatively Regulated by CsMYB72 in Tea Plants.

Anthracnose is a devastating fungal disease in tea plants caused by the pathogen Colletotrichum species. It severely affects tea yield and quality. Pathogenesis-related (PR) genes play indispensable roles in plant resistance to fungal pathogens. Our transcriptome data showed that PR10 family genes are involved in the immune response to anthracnose infection in tea plants; however, the underlying regulatory mechanisms remain unclear. Here, we identified a PR10 family gene, CsPR10-9, which was highly induced by Colletotrichum gloeosporioides infection and salicylic acid (SA)/methyl jasmonate (Me…

Ornithine decarboxylase of the fungal pathogen Colletotrichum higginsianum plays an important role in regulating global metabolic pathways and virulence.

Colletotrichum higginsianum is an important fungal pathogen causing anthracnose disease of cruciferous plants. In this study, we characterized a putative orthologue of yeast SPE1 in C. higginsianum, named ChODC. Deletion mutants of ChODC were defective in hyphal and conidial development. Importantly, deletion of ChODC significantly affected appressorium-mediated penetration in C. higginsianum. However, polyamines partially restore appressorium function and virulence indicating that loss of ChODC caused significantly decreased virulence by the crosstalk between polyamines and other metabolic pa…

Postharvest detection of anthracnose (Colletotrichum asianum) on mango fruit (Mangifera indica L. cv Namdokmai Sithong) using near-infrared response.

Anthracnose disease, caused by fungi of the genus Colletotrichum, poses a major threat to mango production and export industries, with Colletotrichum asianum being among the most significant pathogenic species. This work proposes the hypothesis that the simple difference in absorption between anthracnose-infected and noninfected mangoes illustrated by the average near-infrared (NIR) spectra obtained from hyperspectral images could be used for simple differentiation of the two groups. The method of depositing fungal spores by spraying the spores over the fruit surface, not a small area or speci…

GSNOR plays roles in growth, pathogenicity, and stress resistance by modulating mitochondrial protein COX6B S-nitrosylation in Colletotrichum gloeosporioides.

Nitric oxide (NO) serves as a versatile signaling molecule governing diverse biological processes, primarily through post-translational modifications such as S-nitrosylation. The enzyme glutathione S-nitrosoglutathione reductase (GSNOR) plays a central role in NO homeostasis by modulating cellular levels of S-nitrosoglutathione (GSNO), thereby controlling protein S-nitrosylation dynamics. However, the functional significance of GSNOR in fungal pathogenicity remains insufficiently characterized. In this study, we investigated the function of CgGSNOR in the phytopathogenic fungus Colletotrichum …
