Enzyme immunoassay for Q fever: comparison with complement fixation and immunofluorescence tests and dot immunoblotting.

Enzyme-linked immunosorbent assays (ELISA) for the detection of specific immunoglobulin G (IgG) and IgM antibodies were developed by using purified Coxiella burnetii cells. Variables, including type of microtiter plate, blocking agent, incubation conditions, antigen stability, and substrate type, were examined to achieve optimal ELISA performance. The reliabilities of the assay systems were compared with those of complement fixation (CF) and enhanced immunofluorescence (EIF) tests with 600 human serum samples from defined clinical cases of Q fever, routine samples, and serum specimens from far

Difference in antibody reactivity between complement fixation and immune adherence hemagglutination tests with virus antigens.

Complement-fixing (CF) immunoglobulin M antibody to infantile gastroenteritis virus (a rotavirus) did not show the reactivity of immune adherence hemagglutination (IAHA). Early immunoglobulin G CF antibody produced both in patients and in guinea pigs experimentally infected with Japanese encephalitis virus (a flavivirus) had weak reactivity in IAHA test. However, late antibody showed higher titers by IAHA than by CF. These results suggested that early antibodies with lower affinity are inefficient in the IAHA reaction. The implications of this study are: (i) a low ratio of IAHA/CF antibody tit

False positive complement fixation tests with respiratory virus preparations in bird fanciers with allergic alveolitis.

False positive raised complement fixation titres to egg-grown virus preparations were found in six pigeon fanciers suffering from extrinsic allergic alveolitis but not in six similar budgerigar fanciers. The raised titres in the pigeon fanciers fell fourfold over a short interval in the absence of avian exposure and were attributable to antibodies directed against antigens from hen's egg in which the test virus preparations were grown. Such antigens were shown to be present in the virus preparations. In view of the clinical similarities between influenza and acute extrinsic allergic alveolitis

Evaluation of a modified complement fixation test and an indirect hemagglutination test for the serodiagnosis of melioidosis in pigs.

A complement fixation test modified by the addition of porcine serum and an indirect hemagglutination test were used to detect antibodies to Pseudomonas pseudomallei in pigs. These tests together with cultural examinations were carried out with 250 pigs. The sensitivity and specificity values were 79.3 and 99.5% and 82.8 and 93.2% for the modified complement fixation and hemagglutination tests, respectively. When results from the combination of both tests were considered, the values were 86.2 and 92.8%, respectively.

The relevance of the anti-human globulin (Coombs) test and the complement-fixation test in the diagnosis of brucellosis.

The relationship between the serological findings for brucellosis and the epidemiological factors has been studied in veterinary surgeons in Northern Ireland. The anti-human globulin (Coombs) test and the complement-fixation test for brucella were used in two groups of veterinary surgeons, those self-employed and those employed by the Ministry of Agriculture.Significant serological differences were found to exist between the two groups. Those in private practice showed changes related to age, cattle skin rash, reactions to S. 19 vaccine accidents and symptoms suggestive of brucellosis in the p

A new complement fixation test for toxoplasmosis. Comparison with other serological methods.

A new complement fixation test for toxoplasmosis has been compared with the classical test indirect immunofluorescence test (IFAT) and indirect haemoagglutination test (IHAT). Neither false positive or negative results were obtained with LBCF-H100-TTE. Notably these values correlate well with IFA titers. Furthermore LBCF-H100-TTE shows the highest titers in acute cases and its time course is practically superimposable to IFAT. The differences obtained performing these different tests on the preferential detection of different antigen antibodies systems by these 3 tests are discussed.

Value of the concomitant use of complement fixation and immunodiffusion tests in the diagnosis of coccidioidomycosis.

Complement fixation titers of 1:2 and 1:4 with coccidioidin are not always of diagnostic significance. The concurrent use of the complement fixation and immunodiffusion tests is an effective means for specific serologic diagnosis of coccidioidomycosis in patients with low levels of complement-fixing antibodies. In routine testing, it is recommended that only sera that are negative for complement-fixing antibodies at 1:8 but positive in the immunodiffusion test with coccidioidin be selected for titration at the 1:2 and 1:4 levels.

Filarial complement-fixation test for pulmonary tropical eosinophilia with Ascaris antigen.

A trial of antigen prepared from Ascaris lumbricoides in the complement-fixation test for pulmonary tropical eosinophilia and for filariasis is described. No significant difference was observed between this antigen and that prepared from Dirofilaria immitis.

Complement Fixation Test: An Update of an Old Method for Diagnosis of Bullous Pemphigoid.

The complement fixation test (CFT) is a method traditionally used for diagnosis of gestational pemphigoid. Its performance in diagnosis of bullous pemphigoid (BP) has not been investigated in a large patient cohort. The aim of this single-centre, retrospective, serological case-control study of 300 patients with BP and 136 control patients was to analyse its operating characteristics. CFT was found to have a sensitivity of 71.7% and a specificity of 100%. Furthermore, CFT diagnosed 20 of 46 patients with BP (43.5%) who were negative for both BP180 and BP230 enzyme-linked immunoassays (ELISAs),
