[Elucidation of Phenomena Involving Cyanobacteria in Freshwater Ecosystem by Chemically Ecological Approach].

Lakes Sagami and Tsukui are reservoirs constructed by connecting to the Sagami River. Because of eutrophication of the lakes, cyanobacteria have appeared every year. This review deals with phenomena related to occurrence of cyanobacteria that have been observed for 40 years since 1974 at the lakes. These 40 years of observations raised three interesting issues including the retention of cyanobacteria on their surfaces. These phenomena have been attributed to the usual factors, such as illuminance, nutrition and water temperature, but our research results suggested that they cannot be resolved …

Fate of cyanobacteria and their metabolites during water treatment sludge management processes.

Cyanobacteria and their metabolites are an issue for water authorities; however, little is known as to the fate of coagulated cyanobacterial-laden sludge during waste management processes in water treatment plants (WTPs). This paper provides information on the cell integrity of Anabaena circinalis and Cylindrospermopsis raciborskii during: laboratory-scale coagulation/sedimentation processes; direct filtration and backwashing procedures; and cyanobacterial-laden sludge management practices. In addition, the metabolites produced by A. circinalis (geosmin and saxitoxins) and C. raciborskii (cyli…

Acclimation of chlorophyll a and carotenoid levels to different irradiances in four freshwater cyanobacteria.

This study investigated carotenoid and chlorophyll a (Chl-a) contents under two different growth irradiances in four freshwater cyanobacterial strains. We found an increased weight ratio of zeaxanthin to Chl-a after exposure to high irradiances over several days. Two out of four strains showed higher zeaxanthin amounts on a biomass basis as well. It appears that cyanobacteria enhance their carotenoid pool in response to high light conditions, as increased production of other carotenoids with photoprotective abilities has also been observed under high irradiance levels. Cyanobacteria do not pos…

Influence of blue-green algae (cyanobacteria) on survival of Legionella pneumophila in aerosols.

The fluid in which blue-green algae (cyanobacteria) (Fischerella sp.) had been grown (algal extract) was investigated for its effect on aerosols of Legionella pneumophila. The bacteria were significantly more stable when suspended in algal extract than in the tryptose-saline solution employed in previously reported experiments. The stabilizing property of the extract disappeared after dialysis, suggesting that a relatively small molecule was involved. The relationship of this observation to the epidemiology of Legionnaires disease is discussed.

Reduced mediators released by cyanobacteria during exoelectrogenesis detected using differential pulse voltammetry.

Cyanobacteria generate electrical current through exoelectrogenesis (extracellular electron transfer) downstream of photosynthesis, yet the identity of the endogenous redox mediator(s) responsible remains unresolved. Here, we apply differential pulse voltammetry (DPV) to detect and characterise redox-active species released by Synechocystis sp. PCC 6803 under illumination. To enable sensitive detection, we developed an electrolyte intermediate between BG11 (Blue-Green-11) growth medium and MOPS (3-(N-morpholino)propanesulfonic acid) buffer that minimises background electrochemical interference…

Multigenerational adaptation alters methylmercury uptake in cyanobacteria under warming and elevated CO2.

Climate-driven increases in atmospheric CO2 concentrations and temperature strongly regulate the physiology of phytoplankton, the primary entry point for neurotoxic methylmercury (MeHg) in aquatic food webs. How multigenerational adaptation to these stressors reshapes cellular traits governing MeHg uptake remains unclear. Here, we exposed the bloom-forming cyanobacterium Microcystis aeruginosa to long-term adaptation under elevated temperature (+5 °C) or CO2 (+580 ppm), then quantified MeHg uptake and cellular partitioning. Both stressors reduced cellular MeHg accumulation by 36% under warming

Polyphosphate Attenuates Oxidative Stress to Support Temperature Adaptability in Hot Spring Cyanobacteria.

Thermophilic cyanobacteria successfully colonize thermal gradients within hot springs, implying evolved mechanisms to cope with temperature-induced oxidative stress. Although polyphosphate (polyP) is known to contribute to oxidative stress resistance, its specific role in thermophilic cyanobacteria remains poorly understood. To address this, this study established a temperature gradient (30-70 °C) and used phloretin (polyP synthesis inhibitor) plus exogenous polyP to investigate polyP metabolism, redox homeostasis, photosynthetic function, and growth of Thermosynechococcus sp. FJSJ-1 from hot …

Carbon dioxide from geothermal gas converted to biomass by cultivating coccoid cyanobacteria.

The Blue Lagoon is a geothermal aquifer with a diverse ecosystem located within the Reykjanes UNESCO Global Geopark on Iceland's Reykjanes Peninsula. Blue Lagoon Ltd., which exploits the aquifer, isolated a strain of coccoid cyanobacteria Cyanobacterium aponinum (C. aponinum) from the geothermal fluid of the Blue Lagoon more than two decades ago. Since then Blue Lagoon Ltd. has cultivated it in a photobioreactor, for use as an active ingredient in its skin care products. Until recently, the cultivation of C. aponinum was achieved by feeding it on 99.99% (4N) bottled carbon dioxide (CO2). In th…

Signal peptides for engineered protein secretion by free and hydrogel-encapsulated cyanobacteria.

Cyanobacteria provide a promising platform for light-driven heterologous protein production. In heterologous production systems, cells are typically lysed for product purification or to facilitate interaction with external substrates, unless the product is secreted. Fusion to molecular "shipping labels" known as signal peptides can target chimeric proteins to native membrane translocation pathways and result in secretion. In this work, we identified signal peptides from the model cyanobacterium Synechococcus elongatus PCC 7942 (S. elongatus) and genetically fused to two heterologous enzymes: t…

Seasonal variation of cyanobacteria blooms and microcystin concentrations in an Amazonian river system.

Microcystins (MCs) are hepatotoxic cyanobacterial metabolites that pose risks to aquatic ecosystems and human health. Cyanobacterial blooms are promoted by high nutrient availability and elevated temperatures, negatively affecting water quality, supply, and recreational use. Despite these concerns, information on MCs occurrence in Amazonian waters remains limited. This study evaluated the seasonal variation of MCs concentrations in the Tapajós River and examined their relationship with environmental parameters throughout the hydrological cycle. We hypothesized that microcystin occurrence varie…

Interactive mechanism of elevated pCO2 and nutrient on 2-methylisoborneol production in cyanobacteria.

Cyanobacterial blooms, exacerbated by climate change and eutrophication, increasingly compromise water quality by producing taste and odor (T&O) compounds including 2-methylisoborneol (MIB). Cyanobacteria synthesize MIB by redirecting carbon metabolism intermediates into the methylerythritol phosphate pathway. This process appears to be influenced by rising atmospheric CO2 partial pressure (pCO2). This study examines how elevated pCO2 affects MIB production in Synechococcus sp. FACHB-1061 under varying nutrient conditions. The results indicate that elevated pCO2 boosts cyanobacterial biomass a…

The widespread capability of methylphosphonate utilization in filamentous cyanobacteria and its ecological significance.

Aquatic ecosystems comprise almost half of total global methane emissions. Recent evidence indicates that a few strains of cyanobacteria, the predominant primary producers in bodies of water, can produce methane under oxic conditions with methylphosphonate serving as substrate. In this work, we have screened the published 2 568 cyanobacterial genomes for genetic elements encoding phosphonate-metabolizing enzymes. We show that phosphonate degradation (phn) gene clusters are widely distributed in filamentous cyanobacteria, including several bloom-forming genera. Algal growth experiments revealed…
