Efficient access to all four stereoisomers of phenylalanine cyclopropane analogues by chiral HPLC.

Bonded polysaccharide-derived chiral stationary phases were found to be useful for the preparation of the four stereoisomers of the cyclopropane analogue of phenylalanine (c(3)Phe) as well as for the direct determination of the enantiomeric purity of c(3)Phe derivatives by HPLC. Three chiral stationary phases, consisting of cellulose and amylose derivatives chemically bonded on allylsilica gel, were tested. The mixed 10-undecenoate/3, 5-dimethylphenylcarbamate of cellulose, 10-undecenoate/3, 5-dimethylphenylcarbamate of amylose and 10-undecenoate/p-methylbenzoate of cellulose were the starting

A structure-activity relationship study of spirolactones. Contribution of the cyclopropane ring to antimineralocorticoid activity.

A series of spirolactones containing a cyclopropane ring in the molecule was examined for its effects on the mineralocorticoid receptor. The results were compared with those of a similar series of spirolactones in which the cyclopropane ring was replaced by a double bond. Insertion of a double bond or an alpha-cyclopropane ring into the 1,2 or the 6,7 position leads to a reduction in the binding affinity. The pi-bonding system of the beta-cyclopropane ring at C-6 and C-7 does not promote binding to the receptor. The presence of the 6 beta, 7 beta-cyclopropane ring may deter metabolic activatio

Anæsthesia in Chest Surgery, with Special Reference to Controlled Respiration and Cyclopropane: (Section of Anæsthetics).

Problems in chest surgery: Cases with prolonged toxaemia or amyloid disease require an anaesthetic agent of low toxicity. When sputum or blood are present in the tracheobronchial tree the anaesthesia should abolish reflex distrubances and excessive sputum be removed by suction. The technique should permit the use of a high oxygen atmosphere; controlled respiration with cyclopropane or ether fulfil these requirements. Open pneumothorax is present when a wound of the chest wall allows air to pass in and out of the pleural cavity. The lung on the affected side collapses and the mediastinum moves

Copper-Mediated Radical Cyclopropanation of Activated Alkenes under Continuous-Flow Conditions.

Polysubstituted cyclopropanes are valuable in complex molecule synthesis but challenging to access due to low atom economy and harsh conditions. Herein, we report a copper-mediated, peroxide-initiated cyclopropanation of activated alkenes with active methylene compounds via a radical pathway. This diazo-free, prefunctionalization-free method, integrated with continuous flow, enhances safety, scalability, and efficiency. Operable under mild conditions with broad substrate scope, it offers a practical and atom-economical route to diverse polysubstituted cyclopropanes.

Non-natural olefin cyclopropanation catalyzed by diverse cytochrome P450s and other hemoproteins.

Recent work has shown that engineered variants of cytochrome P450BM3 (CYP102A1) efficiently catalyze non-natural reactions, including carbene and nitrene transfer reactions. Given the broad substrate range of natural P450 enzymes, we set out to explore if this diversity could be leveraged to generate a broad panel of new catalysts for olefin cyclopropanation (i.e., carbene transfer). Here, we took a step towards this goal by characterizing the carbene transfer activities of four new wild-type P450s that have different native substrates. All four were active and exhibited a range of product sel

Synthesis and application of chiral cyclopropane-based ligands in palladium-catalyzed allylic alkylation.

A series of chiral, cyclopropane-based phosphorus/sulfur ligands have been synthesized and evaluated in the palladium-catalyzed allylic alkylation of 1,3-diphenylpropenyl acetate with dimethyl malonate. Variation of the ligand substituents at phosphorus, sulfur, and the carbon backbone revealed 24d to have the optimal configuration for this reaction, giving the product in high yield and with good enantioselectivity (93%). A model for the observed enantioselectivity is discussed within the context of existing models, using X-ray crystallographic data, solution-phase NMR studies, and the absolut

[Enantiomeric resolution of trans-1,2-disubstituted cyclopropane by high performance liquid chromatography].

The enantiomeric resolution of racemic mixtures of thirteen trans-1,2-disubstituted cyclopropane was achieved with HPLC by using Chiralcel OD and Chiralcel OJ as chiral stationary phase and hexane/2-propanol mixtures with different concentrations as eluent. The chromatographic parameters of these racemates on Chiralcel OD and Chiralcel OJ columns were examined. The results showed that enantiomeric resolution ability of the racemic mixtures of trans-1,2-disubstituted cyclopropane on Chiralcel OD and Chiralcel OJ was strongly dependent on the site and the property of substituents on the benzene

Gas chromatographic determination of Ostwald solubility coefficients for cyclopropane, halothane and trichloroethene (trichloroethylene).

Gas chromatographic methods using solvent extraction for the analysis of cyclopropane and and trichloroethene (trichloroethylene) are described and evaluated; cyclopropane was extracted into carbon tetrachloride and trichloroethene into carbon disulphide, using chloroform and toluene respectively as the internal standards. Ostwald solubility coefficients were measured for cyclopropane, halothane and trichloroethene in Krebs solution: at 310 K the respective values +/- SEM of the Ostwald coefficients are 0.181 +/- 0.009, 0.78 +/- 0.02 and 1.54 +/- 0.02; over the temperature range 295-310 K the

Photoredox-Neutral Platform for Direct Decarboxylative Cyclopropanation via Acridine-Iron Dual Catalysis.

A photoredox-neutral approach to cyclopropanes from in situ generated non-stabilized carbenes via a new acridine-iron dual catalytic system is reported. This strategy merges the oxidative decarboxylation of bench-stable carboxylic acids with the reduction of an iron porphyrin under mild photochemical conditions. Bespoke acridine photocatalysts efficiently shuttle electrons between carboxylic acids and iron complexes, obviating the need for a sacrificial reductant. Mechanistic studies reveal that, contrary to prevailing assumptions, decarboxylation does not proceed through a concerted PCET path…

Lipid-Inspired Low Melting Ionic Liquids via Synergistic Cyclopropanation and Branching of Terpenoids.

Bacteria employ cyclopropane motifs as bioisosteres for unsaturations to modulate lipid bilayer fluidity and protect cellular membranes under environmental stress. Drawing inspiration from this biological strategy, we investigated how cyclopropanation impacts the thermophysical properties of lipid-inspired ionic liquids. We synthesized a series of imidazolium-based ionic liquids incorporating cyclopropanated derivatives of three renewable terpenoids: phytol, farnesol, and geraniol. Through an integrated approach combining property-driven design, thermophysical analysis, X-ray crystallography,

Cyclopropanation of Alkenes with Dichloromethane and Chloroform via Halogen Atom Transfer Using Amine Carboxyborane.

Cyclopropanes are privileged motifs in medicinal chemistry due to their role as bioisosteres of arenes, alkenes, and small alkyl groups. Herein, we report a transition-metal-free, photoredox-catalyzed cyclopropanation of diverse alkenes via halogen atom transfer between dichloromethane (CH2Cl2) as a C1 synthon and amine-ligated boryl radicals generated from amine carboxyborane. This method proceeds under mild reaction conditions, exhibits a broad substrate scope, and is scalable. The synthetic utility is further highlighted by deuterium incorporation using CD2Cl2 and the formation of chlorocyc…

Vitamin B12 -Photocatalyzed Cyclopropanation of Electron-Deficient Alkenes Using Dichloromethane as the Methylene Source.

The cyclopropyl group is of great importance in medicinal chemistry, as it can be leveraged to influence a range of pharmaceutical properties in drug molecules. This report describes a Vitamin B12 -photocatalyzed approach for the cyclopropanation of electron-deficient alkenes using dichloromethane (CH2 Cl2 ) as the methylene source. The reaction proceeds in good to excellent yields under mild conditions, has excellent functional group compatibility, and is highly chemoselective. The scope could also be extended to the preparation of D2 -cyclopropyl and methyl-substituted cyclopropyl adducts st

A Diverse Library of Chiral Cyclopropane Scaffolds via Chemoenzymatic Assembly and Diversification of Cyclopropyl Ketones.

Chiral cyclopropane rings are key pharmacophores in pharmaceuticals and bioactive natural products, making libraries of these building blocks a valuable resource for drug discovery and development campaigns. Here, we report the development of a chemoenzymatic strategy for the stereoselective assembly and structural diversification of cyclopropyl ketones, a highly versatile yet underexploited class of functionalized cyclopropanes. An engineered variant of sperm whale myoglobin is shown to enable the highly diastereo- and enantioselective construction of these molecules via olefin cyclopropanati…

Synthesis and biological evaluation of bromophenol derivatives with cyclopropyl moiety: Ring opening of cyclopropane with monoester.

Trans-(1R*,2R*,3R*)-Ethyl 2-(3,4-dimethoxyphenyl)-3-methylcyclopropane-1-carboxylate (6) and its cis isomer 7 were obtained from the reaction of the methyl isoeugenol (5) with ethyl diazoacetate. The reduction and bromination reactions of the ester 6 and 7 together with the hydrolysis of all esters were carried out. Opening ring of cyclopropane was observed in the reaction of 7 with bromine. The opening of cyclopropane ring with COOR and synthesis of esters, alcohols and acids (6-26) are new. These obtained bromophenol derivatives (6-26) were effective inhibitors of the cytosolic carbonic anhy

Cooperative photoredox and N-heterocyclic carbene-catalyzed formal C-H acylation of cyclopropanes via a deconstruction-reconstruction strategy.

Cyclopropanes are ubiquitous and key structural motifs in commercially available drugs and bioactive molecules. Herein, we present regio-selective acylation of aryl cyclopropanes with cooperative photoredox and N-heterocyclic carbene catalysis. This approach involves a deconstruction-reconstruction strategy via γ-chloro-ketones as intermediates and fulfills the formal C(sp3)-H functionalization of cyclopropanes.
