Trained Immunity in Host Defense: Mechanisms, Pathogen Interactions, and Therapeutic Potential.

Trained immunity refers to the functional reprogramming of innate immune cells-such as monocytes, macrophages, and natural killer cells-those results in a modulated, and often heightened, response to secondary stimuli. This phenomenon challenges the traditional view that immunological memory is restricted to the adaptive immune system and has emerged as a concept linking host defense, vaccination, and inflammatory disease. This review synthesizes current evidence on the molecular basis of trained immunity, including chromatin remodeling, histone modifications, and metabolic rewiring, and exami

Drosophila host defense mechanisms against filamentous fungal pathogens with diverse lifestyles.

Entomopathogenic fungi serve as powerful regulators of insect populations in nature. However, how immune effectors combat fungal pathogens remains incompletely understood. We employ Drosophila melanogaster as a genetically tractable model to dissect immune defense mechanisms against diverse fungal pathogens. We show that the Toll pathway is the key determinant of immunity against all species tested regardless of their ecological strategy, primarily through resistance mechanisms that limit fungal proliferation. In addition, melanization, but not phagocytosis or the Imd pathway, also has a role …

β-Glucan in antiviral defense: mechanisms, immune modulation, and therapeutic prospects.

β-Glucans, naturally occurring β-D-glucose polysaccharides from fungi, yeast, bacteria, algae, and cereals, have emerged as promising immunomodulatory agents in antiviral defense. Their structural diversity-encompassing β-1,3, β-1,6, and β-1,4 linkages-underpins varied solubility, bioavailability, and biological activity, driving their therapeutic potential. Unlike conventional antivirals that target viral replication, β-glucans enhance host immunity by activating innate and adaptive responses through receptors such as dectin-1, toll-like receptors, and complement receptor 3, thereby stimulati

Metallothioneins: an unraveling insight into remediation strategies of plant defense mechanisms.

Phytoremediation is an eco-friendly, sustainable way to clean up the environment using green plants that effectively remove and degrade pollutants from soil, water, or air. Certain hyperaccumulator plants can effectively mitigate heavy metals, organic compounds, and radioactive substances through absorption, adsorption, and transformation. This method offers a cost-effective and esthetically pleasing alternative to traditional remediation techniques, contributing to the restoration of contaminated ecosystems. Nanophytoremediation entails combining nanotechnology with phytoremediation technique

Defense Mechanisms and Parenting Styles in Patients With Anorexia and Bulimia.

Dysfunctional parenting styles are risk factors for eating disorders (EDs). In this observational study, we examined 57 women with ED, a psy … ture factor (p = 0.016). Patients with anorexia and bulimia overutilize more neurotic and primitive defense mechanisms compared with nonclinical subjects, and this use is associated with greater severity of eating symptomatology. This can be related to diversified ego forces, can inform about the nature and severity of disease, and can characterize prognostic and ps …

Impulsivity, Consciousness and Defense Mechanisms of the Ego Among Pathological Gamblers.

Pathological gambling is a growing health problem in Romania, recognized in DSM V (2013) as a form of psychopathology included in the category of addictions. This study aims to find out whether there are differences between gamblers and the control group among the dependent variables (impulsivity, conscientiousness, and defense mechanisms). According to the literature, impulsivity and low conscientiousness are specific features to pathological gamblers. A second goal is to check if there are one or more defense mechanisms defining gamblers. A recent study (Waqas et al., Waqas et al., Psychiatr…

Non-Reflex Defense Mechanisms of Upper Airway Mucosa: Possible Clinical Application.

The sinonasal mucosa has an essential role in defense mechanisms of the upper respiratory tract. The innate immune system presents the primary defense against noxious microorganisms followed by induction of the adaptive immune mechanisms as a consequence of the presence of pathogens. This well-known activation of adaptive immune system in response to presence of the antigen on mucosal surfaces is now broadly applicated in vaccinology research. Prevention of infectious diseases belongs to substantial challenges in maintaining the population health. Non-invasive, easily applicable mucosal vaccin…

Thromboinflammation: challenges of therapeutically targeting coagulation and other host defense mechanisms.

Thrombosis with associated inflammation (thromboinflammation) occurs commonly in a broad range of human disorders. It is well recognized clinically in the context of superficial thrombophlebitis (thrombosis and inflammation of superficial veins); however, it is more dangerous when it develops in the microvasculature of injured tissues and organs. Microvascular thrombosis with associated inflammation is well recognized in the context of sepsis and ischemia-reperfusion injury; however, it also occurs in organ transplant rejection, major trauma, severe burns, the antiphospholipid syndrome, preecl

Tuneable poration: host defense peptides as sequence probes for antimicrobial mechanisms.

The spread of antimicrobial resistance stimulates discovery strategies that place emphasis on mechanisms circumventing the drawbacks of traditional antibiotics and on agents that hit multiple targets. Host defense peptides (HDPs) are promising candidates in this regard. Here we demonstrate that a given HDP sequence intrinsically encodes for tuneable mechanisms of membrane disruption. Using an archetypal HDP (cecropin B) we show that subtle structural alterations convert antimicrobial mechanisms from native carpet-like scenarios to poration and non-porating membrane exfoliation. Such distinct m

Overlapping and complementary oxidative stress defense mechanisms in nontypeable Haemophilus influenzae.

The Gram-negative commensal bacterium nontypeable Haemophilus influenzae (NTHI) can cause respiratory tract diseases that include otitis media, sinusitis, exacerbations of chronic obstructive pulmonary disease, and bronchitis. During colonization and infection, NTHI withstands oxidative stress generated by reactive oxygen species produced endogenously, by the host, and by other copathogens and flora. These reactive oxygen species include superoxide, hydrogen peroxide (H2O2), and hydroxyl radicals, whose killing is amplified by iron via the Fenton reaction. We previously identified genes that e

Attachment style and defense mechanisms in parents who abuse their children.

Adult attachment style, defense mechanisms, and personal history of abuse was studied in a group of abusive parents. As a group, these parents made unusually high use of the defense of denial; this was especially true of those with a Fearful attachment style. However, the use of Identification was characteristic of those with a Preoccupied attachment style. Further, personal abuse history was related to adult attachment style. Those who reported having been abused as a child were less likely to have a Secure attachment style, and a history of physical or sexual abuse was associated with a Preo…

Oral defense mechanisms are impaired early in HIV-1 infected patients.

We have examined the hypothesis that individuals infected with human immune deficiency virus type 1 (HIV-1) experience significant, specific … infected patients compared to controls (41% vs. 8%, p less than 0.05). We conclude that normal oral defense mechanisms show signs of compromise in HIV-1 infected individuals. We suggest that (a) effects of HIV-1 infection are seen early in the oral cavity, (b) impairment of oral defense mechanisms may facilitate entry of microorganisms with an attendant increased ri …

The antiphage mechanism of a widespread trypsin-MBL defense module.

Protease-mediated activation of immune effectors is an evolutionarily conserved mechanism. This study identifies a widespread trypsin-MBL (metallo-β-lactamase) module as a core effector in diverse antiviral bacterial immune systems, such as Hachiman, AVAST and Argonaute. Focusing on the Hachiman-associated trypsin-MBL system, we show that trypsin•HamAB protease activity is inhibited by ATP, while MBL is an autoinhibited DNase with two insertion loops obstructing its catalytic site. Upon infection, trypsin•HamAB senses foreign DNA and hydrolyzes ATP, activating trypsin-like activity, which spec

Role of brain-gut axis in mechanism of gastrointestinal defense.

The digestive system, comprising gastrointestinal (GI)-tract and connecting organs, such as liver or pancreas, has a wide autonomy in the human body due to the presence of specific nervous and endocrine systems of its own and can act voluntarily without stimulation from the rest of the body. This wide physiological autonomy of a digestive tract does not exclude connection with other organs and central as well as peripheral mechanisms controlling nervous and hormonal systems. The vagal nerve is a structure connecting GI-tract with brain centers. Both, the afferent and efferent components of the

Emerging Evasion Mechanisms of Macrophage Defenses by Pathogenic Bacteria.

Macrophages participate to the first line of defense against infectious agents. Microbial pathogens evolved sophisticated mechanisms to escape macrophage killing. Here, we review recent discoveries and emerging concepts on bacterial molecular strategies to subvert macrophage immune responses. We focus on the expanding number of fascinating subversive tools developed by Listeria monocytogenes, Staphylococcus aureus, and pathogenic Yersinia spp., illustrating diversity and commonality in mechanisms used by microorganisms with different pathogenic lifestyles.

A potential defense mechanism against amyloid deposition in cerebellum.

Amyloid-β (Aβ) is the major component of senile plaques in Alzheimer's disease (AD) brains. Senile plaques are generally observed in cerebral cortex (CTX) rather than cerebellum (CBL) in AD patients. However, it is not clear why CBL has less Aβ deposition than CTX. It is very important to elucidate the mechanism of suppressing Aβ deposition in CBL, because it contributes to understanding of not only AD pathogenesis but also prevention and cure of AD. In this study, we explored to figure out the potential mechanism of reducing Aβ deposition in CBL. We observed higher age-dependent elevation of

Graves' disease: a host defense mechanism gone awry.

In this report we summarize evidence to support a model for the development of Graves' disease. The model suggests that Graves' disease is initiated by an insult to the thyrocyte in an individual with a normal immune system. The insult, infectious or otherwise, causes double strand DNA or RNA to enter the cytoplasm of the cell. This causes abnormal expression of major histocompatibility (MHC) class I as a dominant feature, but also aberrant expression of MHC class II, as well as changes in genes or gene products needed for the thyrocyte to become an antigen presenting cell (APC). These include
