Disulfidptosis modification patterns are involved in the immune microenvironment regulation of septic acute respiratory distress syndrome.

Disulfidptosis is a new form of programmed cell death. However, there is limited information available regarding the impact of disulfidptosis on septic acute respiratory distress syndrome (ARDS). The 16 disulfidptosis-related genes (DRGs) were collected from a previous study. Gene expression data of sepsis and septic ARDS samples were downloaded from the Gene Expression Omnibus database. The risk score model in septic ARDS was constructed based on the DRGs, followed by the investigation of immune microenvironment in septic ARDS patients. Furthermore, septic ARDS patients were divided into diff…

Metabolic cell death: ferroptosis, cuproptosis, and disulfidptosis as a unified framework for targeting metabolic vulnerabilities in disease.

Resistance to apoptosis-targeted cancer therapies remains a major clinical hurdle. Ferroptosis, cuproptosis, and disulfidptosis represent typical forms of metabolic cell death that bypass classical apoptotic pathways, offering unprecedented opportunities to overcome such resistance. These three death modalities converge on a shared metabolic hub-the SLC7A11/cystine/GSH/NADPH axis-where disruption at different nodes steers the outcome toward ferroptosis, cuproptosis, or disulfidptosis. This interconnection is the foundation of our unified framework. While recent seminal reviews (Mao et al.) hav…

Age-Related Lamin B1 Deficiency Activates SLC7A11-Dependent Disulfidptosis to Impair Osteogenesis via Intercepting Mitochondrial Fission.

Senile osteoporosis (SOP) is an aging-related disease characterized by decreased bone mass and susceptibility to fracture. The pathological … ancer (SE) formation to effectively drive SLC7A11 expression. Elevated SLC7A11 expression activated disulfidptosis in SOP-MSCs, thereby resulting in disulfide bond formation in actin and subsequent cytoskeleton collapse. The disulfide-crosslinked actin-induced cytoskeleton disorganization further disrupted their binding with DRP1, inducing mitophagy and oxidativ …

Emerging regulated cell death (cuproptosis, disulfidptosis, and PANoptosis) in ischemic stroke: research progress and translational prospects.

Ischemic stroke (IS) is one of the leading causes of disability and mortality worldwide. Its pathological mechanisms involve complex cascades including energy metabolism failure, excitotoxicity, oxidative stress, neuroinflammation, and multiple forms of regulated cell death (RCD). In recent years, cuproptosis, disulfidptosis, and PANoptosis, as three emerging RCD modalities, have attracted increasing attention in ischemic brain injury. Cuproptosis is triggered by copper overload and leads to proteotoxic stress via abnormal oligomerization of lipoylated mitochondrial proteins. Disulfidptosis oc…

NRF2 coordinates ferroptosis and disulfidptosis in dermal papilla cells via redox metabolic reprogramming in androgenetic alopecia.

Dysfunction of dermal papilla cells (DPCs) is a central feature of androgenetic alopecia (AGA), in which oxidative imbalance plays a critica … ity is associated with two distinct regulated cell death-related processes in DPCs: ferroptosis and disulfidptosis. Pharmacological activation of NRF2 by dimethyl fumarate (DMF), together with molecular and metabolic analyses, indicated that impaired NRF2 signaling was linked to suppression of the SLC7A11-GSH-GPX4 axis, increased lipid peroxidation, and ferropto …

A glucose metabolism-targeted nanoplatform amplifies disulfidptosis-associated immunogenic cell death in triple-negative breast cancer.

Disulfidptosis is a metabolism-dependent form of cell death driven by disulfide stress that induces actin cytoskeletal collapse in SLC7A11-high cells under glucose deprivation. However, strategies to therapeutically exploit this vulnerability remain underdeveloped. Given the high dependence of triple-negative breast cancer (TNBC) on glucose metabolism, we engineered a tumor microenvironment-responsive MnOx nanoplatform for sustained delivery of the GLUT1 inhibitor BAY-876 and concurrent glutathione (GSH) depletion. Within this integrated system, BAY-876 restricts GLUT1-mediated glucose uptake,…

Tumor Microenvironment-Responsive Nanoreactor Induces Disulfidptosis in Pancreatic Cancer via Metabolic Interference and Redox Catalysis.

Pancreatic cancer exhibits extensive metabolic reprogramming that supports rapid progression and therapeutic resistance, making metabolic vulnerabilities attractive targets for intervention. Here, a tumor microenvironment-responsive nanoreactor (Pht@HMnO2-HA) is designed to induce disulfidptosis in pancreatic cancer through coordinated metabolic interference and redox catalysis. Following CD44-mediated tumor targeting and cellular internalization, the nanoreactor's responsive self-optimization within the tumor microenvironment enables localized release of phloretin, suppressing glucose uptake …

Metabolic pathways and cell death modalities in diabetic complications: unraveling pyroptosis, ferroptosis, cuproptosis, and disulfidptosis.

The pathogenesis of diabetic complications involves a complex interplay of metabolic disturbances, in which diverse programmed cell death (PCD) pathways play a pivotal role. This article systematically reviews the molecular mechanisms of four novel types of metabolic cell death-pyroptosis, ferroptosis, cuproptosis and disulfidptosis-with a focus on their specific pathological roles in diabetic kidney disease, retinopathy, cardiovascular complications and neuropathy. It also analyzes the crosstalk and synergistic regulatory networks among these four cell death modalities. Studies have demonstra…

Alexidine dihydrochloride enhances the sensitivity of human hepatocellular carcinoma to disulfidptosis via ATF4-DDIT3 activation.

Disulfidptosis, an emerging regulated cell death modality characterized by pathological disulfide bond aggregation and subsequent actin cytoskeletal disintegration, presents promising therapeutic potential for apoptosis- and ferroptosis-resistant malignancies. However, the molecular initiators of disulfidptosis and their connection with mitochondrial dysregulation remain elusive. Herein, we identified alexidine dihydrochloride (AD) as a selective disulfidptosis enhancer in hepatocellular carcinoma (HCC), and the comprehensive multi-omics analysis and other functional validation indicated that …

Clarifying new molecular subtyping and precise treatment of melanoma based on disulfidptosis-related lncRNA signature.

Disulfidptosis, the newest form of programmed cell death, is closely associated with the immune microenvironment of cancer cells. Long non-coding RNA (lncRNA) has also been found to play a crucial role in melanoma. However, the role of disulfidptosis-related lncRNA in melanoma remains unclear. Through bioinformatic analysis of the transcriptional, clinical, and pathological data from the TCGA-SKCM (The Cancer Genome Atlas-Skin cutaneous melanoma) database, we established a 2-Disulfidptosis-related lncRNA (DRL) prognostic model and a novel molecular subtype for melanoma. The survival and ROC cu…

Cuproptosis and Disulfidptosis Converge to Empower PD-L1 Checkpoint Therapy via Cadict-Induced PD-L1 Translation.

Immune checkpoint blockade (ICB) has emerged as a cornerstone of cancer therapy, yet its effectiveness remains restricted in PD-L1-low malignancies due to insufficient target expression. We herein develop the cuproptosis and disulfidptosis co-delivery targeted (Cadict) nanodrug, an epidermal growth factor receptor (EGFR)-targeted nanoplatform designed to co-induce cuproptosis and disulfidptosis, thereby synergistically augmenting tumor cytotoxicity and sensitizing cancers to anti-PD-L1 therapy. BHLHE41-SLC7A11 transcriptional axis and chromatin remodeling signatures in osteogenic-lineage disulfidptosis-like stress in osteoporosis.

BACKGROUND: Osteoporosis (OP) is characterized by impaired bone homeostasis in which bone resorption exceeds bone formation. Disulfidptosis, a recently described disulfide stress-induced cell death program linked to cytoskeletal collapse, has been suggested to contribute to OP, yet its cell-type-specific relevance within the bone marrow mesenchymal stem cell (BM-MSC) osteogenic lineage-and the upstream upstream transcriptional and epigenetic programs shaping this stress response-remain unclear. METHODS: This study integrated a peripheral blood monocyte microarray dataset (GSE56815; 20 OP vs. …
