α-Methyl-α-phenylsuccinimide ameliorates neurodegeneration in a C. elegans model of TDP-43 proteinopathy.

The antiepileptic drug ethosuximide has recently been shown to be neuroprotective in various Caenorhabditis elegans and rodent neurodegeneration models. It is therefore a promising repurposing candidate for the treatment of multiple neurodegenerative diseases. However, high concentrations of the drug are required for its protective effects in animal models, which may impact on its translational potential and impede the identification of its molecular mechanism of action. Therefore, we set out to develop more potent neuroprotective lead compounds based on ethosuximide as a starting scaffold. Ch…

Aza-Michael approach for the rapid synthesis of 3,5-dimethylpyrazole derivatives with potential biological significance.

A promising strategy for preventing epileptogenic processes involves the development of modified aza-Michael adducts, which demonstrate a range of biological activities, including antioxidant, anti-inflammatory, and antiviral effects. In the present study, compound 3, administered at a dose of 50 mg/kg, prevented pentylenetetrazole-induced clonic seizures in 60% of mice (ED50 = 37.5 mg/kg), while compound 4 achieved seizure prevention in 40% of mice. With an ED50 value of 155.5 mg/kg, compound 3 was approximately four times more potent than ethosuximide. Importantly, neither the tested compoun…

Flash-evoked afterdischarge in rat as a model of the absence seizure: dose-response studies with therapeutic drugs.

The flash-evoked afterdischarge (FEAD) is a self-sustained burst of wave-and-spike complexes recorded from occipital cortex in the rat and o … EAD by six antiepileptic drugs with established clinical profiles. It was found that phenobarbital, ethosuximide, and trimethadione suppressed FEAD in a dose-related manner, and that ethosuximide was approximately three times as potent as trimethadione. Mephenytoin produced a maximal reduction of FEAD of only 30 to 40%, which was not dose-related. Neither phen …

Anticonvulsant potency of common antiepileptic drugs in the gerbil.

In gerbils, 'minor' (myoclonic) and 'major' (clonic-tonic) seizures were induced by blowing at the animals with compressed air. The anticonvulsant ED50 of the following drugs was determined after oral administration against both types of seizures: phenytoin, phenobarbital, carbamazepine, sodium valproate, ethosuximide, and diazepam. Valproate, ethosuximide, and diazepam were most potent against 'minor' seizures which could not or only partially be suppressed by phenytoin or carbamazepine, respectively. The 'grand mal' drugs phenytoin, phenobarbital, and carbamazepine were, on the other hand, m…

The effects of cobalt chloride, SKF-525A, and N-(3,5-dichlorophenyl)succinimide on in vivo hepatic mixed function oxidase activity as determined by single-sample plasma clearances.

1. Four drugs--antipyrine, theophylline, quinidine, and ethosuximide--were used as probes of in vivo hepatic mixed function oxidase (MFO) activity. Functional MFO activity was evaluated by estimating probe clearances subsequent to pretreatment of rats with either cobalt chloride, SKF-525A, or N-(3,5-dichlorophenyl) succiminide (NDPS). 2. Clearances of each probe were estimated from single plasma concentration measurements. Each pretreatment altered the clearances of this panel of probes in a different way. NDPS pretreatment increased theophylline clearance while slowing quinidine and ethosuxim…

Effects of classical antiepileptics on thresholds for phenomena induced by cortical stimulation in rats.

Our aim was to study the effects of phenobarbital, phenytoin and ethosuximide on epileptic afterdischarges induced by cortical stimulation in rats. Fifteen-second series of low-frequency (8 Hz) rhythmic stimulation of the sensorimotor cortex were applied in rats with chronically implanted electrodes. Intervals between the stimulation series were at least 10 min and intensity was increased in a step-wise manner. Threshold current intensities were estimated for movements directly induced by stimulation, epileptic afterdischarges of the spike-and-wave type, clonic seizures accompanying this type …

Simultaneous rapid HPLC determination of anticonvulsant drugs in plasma and correlation with EMIT.

A method is presented for measuring simultaneously five anti-convulsants (primidone, ethosuximide, phenobarbital, carbamazepine and diphenylhydantoin) in serum and plasma by high-performance liquid chromatography (HPLC) using alphenal (5 - allyl - 5 - phenyl - barbituric acid) as the internal standard. All five drugs are separated from each other and, in the case of primidone, from its metabolie, 2 - phenyl - 2 - ethyl - malondiamide. Total HPLC time for the separation is seven minutes. The chromatography is performed on a C-8 reverse phase column with a mobile phase consisting of acetonitrile…

[Plasma determination of 7 common drugs by high performance liquid chromatography].

A high performance liquid-chromatographic procedure is reported for measuring the plasma concentration of 5 anticonvulsants: phenobarbital, phenytoin, carbamazepine, ethosuximide, valproic acid and 2 bronchodilators: theophylline and caffeine. Limits of detection of ethosuximide and valproic acid are 28 mumol/l, 5 mumol/l for the other drugs. Linearity limits are always very upper than the higher therapeutic dose concentration, the inter assay relative standard deviation are less than 8 p. cent for the 7 drugs at the 3 levels of concentration and the analytical recoveries ranged from 93 to 104…

Anticonvulsant drugs: mechanisms of action.

A variety of the anticonvulsant drugs, including carbamazepine, phenytoin, primidone, phenobarbital, clonazepam, valproic acid, and ethosuximide, are available for use in the treatment of patients with seizure disorders. These anticonvulsants vary in their efficacy against experimental seizures in animals and against seizures in humans. The mechanistic basis for this variability in anticonvulsant drug action remains uncertain, but numerous mechanisms of action have been proposed. We have used mouse neurons in primary dissociated cell culture to study the action of these anticonvulsant drugs on…

Diphenylhydantoin substrate induced difference spectra: inhibition by other anticonvulsant drugs.

The ability of five commonly used anticonvulsants to produce substrate induced difference specta was assessed. Only diphenylhydantoin (DPH) and phenobarbitone were observed to produce such spectra and were of Type I. Mean Ks values for DPH and phenobarbitone were 6.8 x 10(-5) and 4.0 x 10(-4)M, respectively. The anticonvulsants sulthiame, ethosuximide, sodium valproate and phenobarbitone exhibited inhibition of DPH substrate induced difference spectra to varying degrees. Analysis of data as in the classical enzyme/inhibitor models resulted in mean Ksi values for the four anticonvulsants of 1.3…

A Practical Guide to Treatment of Childhood Absence Epilepsy.

Childhood absence epilepsy (CAE) is a common pediatric epilepsy syndrome with distinct seizure semiology, electroencephalography (EEG) features, and treatment. A diagnosis of CAE can be obtained during an office visit with a careful history, physical exam including prolonged hyperventilation, and a routine EEG. The treatment of choice for CAE with absence seizures only is ethosuximide. Valproic acid and lamotrigine are also effective treatments for many patients, but when compared to ethosuximide, valproic acid has more adverse effects and lamotrigine is less effective. Attention to predictors…

Synthesis and anticonvulsant evaluation of some new 2-benzylsuccinimides.

A series of 2-benzylsuccinimides (4a-f) were prepared for evaluation as potential anticonvulsants. Primary (Phase I) screening of these comp … in mice, 4d and 4e both demonstrated anticonvulsant potency superior to that of the prototype drug (ethosuximide) by the MES and scMet assays. However, they also exhibited greater neurotoxicity than ethosuximide in the rotorod test.

Early treatment suppresses the development of spike-wave epilepsy in a rat model.

PURPOSE: Current treatments for epilepsy may control seizures, but have no known effects on the underlying disease. We sought to determine whether early treatment in a model of genetic epilepsy would reduce the severity of the epilepsy phenotype in adulthood. METHODS: We used Wistar albino Glaxo rats of Rijswijk (WAG/Rij) rats, an established model of human absence epilepsy. Oral ethosuximide was given from age p21 to 5 months, covering the usual period in which seizures develop in this model (age approximately 3 months). Two experiments were performed: (1) cortical expression of ion channels…

Seizure control following administration of anticonvulsant drugs in the quaking mouse.

The effectiveness of a variety of clinical anticonvulsant drugs was evaluated in the quaking mutant mouse model of epilepsy. In this model, tonic-clonic seizures are easily elicited by handling and the effects of administration of carbamazepine (CBZ), phenytoin (DPH), phenobarbital (PB), diazepam, valproic acid (VPA) and ethosuximide were quantitatively evaluated. Chronic oral administration of CBZ, DPH and PB reduced the frequency of seizures and this was positively correlated with plasma levels of the drugs. The plasma levels of the 10,11-epoxide metabolite of CBZ were found to be approximat…

Evaluation of longevity enhancing compounds against transactive response DNA-binding protein-43 neuronal toxicity.

In simple systems, lifespan can be extended by various methods including dietary restriction, mutations in the insulin/insulin-like growth f … ctive properties. We report here that resveratrol, rolipram, reserpine, trolox, propyl gallate, and ethosuximide protect against mutant TAR DNA-binding protein-43 neuronal toxicity. Finally, of all the compounds tested, only resveratrol required daf-16 and sir-2.1 for protection, and ethosuximide showed dependence on daf-16 for its activity.
