Fine structure of soft and hard tissues involved in eye migration in metamorphosing Japanese flounder (Paralichthys olivaceus).

The body of a Japanese flounder (Paralichthys olivaceus) changes from a symmetrical to an asymmetrical form during metamorphosis. To obtain detailed information on the mechanisms of the migration of the right eye to the left side, soft and hard tissues in the head of larval flounders were examined using transmission electron microscopy (TEM). Retrorbital vesicles (Rvs) are pairs of sac-like structures under the eyes. It has been suggested that the asymmetrical development of Rvs, with the right (blind) one being bigger than the left, is the driving force behind eye migration. The present study…

Decontamination of winter flounder (Pleuronectes americanus) following chronic exposure to effluent from a pulp and paper mill.

A study was conducted to determine the influence of decontamination on winter flounder living downstream from a pulp and paper mill. The fish and controls were held 8-26 weeks under contaminant-free conditions in aquaria supplied with ambient sea water. Groups of flounder, for comparison, were also sampled near the paper mill at the time of or following autopsy. No differences were apparent in length/organ-weight relationships between depurated and control groups after 26 weeks at 0-6 degrees C, but condition factor, gonadal and hepatosomatic indices, coincident with elevated levels of detoxif…

Size-, sex- and site-dependent morphological variation in the stone flounder (Kareius bicoloratus) from Korean coastal waters.

Kareius bicoloratus is a commercially important flatfish in Korean coastal fisheries and an ecologically relevant benthic species in nearshore ecosystems. To evaluate how multiple biological and environmental factors jointly shape flatfish morphology, we analysed 81 morphometric indices and two-dimensional geometric morphometrics (GMM) in wild K. bicoloratus collected from two Korean coastal regions, incorporating size, sex and sampling site. Classical morphometrics based on a three-way ANOVA framework identified body size and sex as the primary drivers of shape variation, particularly influen

Intraperitoneal extracellular trap-derived proteins mediate the amplification of vaccine-induced immune responses in flounder (Paralichthys olivaceus).

Myeloid cells are capable of releasing web-like extracellular structures composed of DNA and granular proteins, known as extracellular traps (ETs), which function to eliminate invading pathogens and modulate immune responses. Intraperitoneal vaccine administration has been demonstrated to induce ETs, but their roles in vaccine-mediated immune responses remain largely unknown. In this study, we show that a protein subunit vaccine triggers the release of ETs in the peritoneal cavity of teleost fish. Pharmacological inhibition of ET formation by 4-aminobenzoic acid hydrazide (ABAH) or enzymatic d

Characterization and potential pathogenicity of Ceratomyxa koreana n. sp. (Myxozoa: Ceratomyxidae) in cultured olive flounder (Paralichthys olivaceus).

Myxosporean parasites have been understudied in Korea, with only a few species described in marine environments. This study aimed to identify and characterize a new Ceratomyxa species associated with emaciation in olive flounder (Paralichthys olivaceus) farmed in Jeju Island, Korea. Morphological analysis revealed that the isolate had spores with an average length of 10.8 ± 1.0 μm and thickness of 41.9 ± 7.4 μm, characterized by two subspherical polar capsules (4.0 μm). The specimens morphologically resembled C. protopsettae Fujita, 1923, but were differentiated from it by their smaller spore …

mRNA-miRNA sequencing reveals mechanisms of 2,2'-dipyridyl disulfide-induced thyroid disruption in Japanese flounder (Paralichthys olivaceus).

This study was conducted to evaluate the thyroid-disrupting effects of 2,2'-dipyridyl disulfide using Japanese flounder (Paralichthys olivaceus) as an animal model and to reveal the underlying mechanisms from the perspective of miRNA-mRNA interactions. The results indicated that 2,2'-dipyridyl disulfide exposure decelerated the metamorphic progress of P. olivaceus, suggesting its thyroid-disrupting property as an antagonist. Furthermore, radioimmunoassays, thyroid histological observation, real-time polymerase chain reaction, and mRNA sequencing showed that 2,2'-dipyridyl disulfide exposure ex…

The potential role of the Sox9a protein in regulating cyp19a transcription and ovarian development in the olive flounder (Paralichthys olivaceus).

Understanding the gonadal differentiation and development of fish is critical for both basic biology and aquaculture. Sox9 has been associated with the determination and subsequent development of fish testes. In teleosts, genome duplication events produced two co-orthologs of the sox9 gene, known as sox9a and sox9b. However, direct evidence of their exact roles at the protein level remains limited. In this study, the bioactive recombinant Sox9a protein from the olive flounder Paralichthys olivaceus, an important mariculture fish in Asia, was obtained. Expression analysis showed that the flound…

Isotopes and otolith chemistry provide insight into the biogeochemical history of mercury in southern flounder across a salinity gradient.

Methylmercury (MeHg) continues to pose a significant global health risk to wildlife and humans through fish consumption. Despite numerous advancements in understanding the mercury (Hg) cycle, questions remain about MeHg sources that accumulate in fish, particularly across transitional coastal areas, where harvest is prominent and Hg sources are numerous. Here we used a unique combination of Hg and nutrient isotopes, and otolith chemistry to trace the biogeochemical history of Hg and identify Hg sources that accumulated in an economically important fish species across Mobile Bay, Alabama (USA).

Effect of miR-155 as a molecular adjuvant of DNA vaccine against VHSV in olive flounder (Paralichthys olivaceus).

Rhabdoviral G protein-based DNA vaccines have been recognized as a useful way to protect cultured fish from rhabdoviral diseases. In Korea, viral hemorrhagic septicemia virus (VHSV) genotype IVa has been the primary culprit of high mortalities of cultured olive flounder (Paralichthys olivaceus). In this study, we inserted a miR-155-expressing cassette into the VHSV's G protein-based DNA vaccine, and analyzed the effects of miR-155 on the antiviral activity and on the vaccine efficacy in olive flounder. Olive flounder fingerlings were intramuscularly (i.m.) immunized with 10 μg/fish (1st experi…

Effects of polychlorinated biphenyls on metamorphosis of a marine fish Japanese flounder (Paralichthys olivaceus) in relation to thyroid disruption.

This study examined the influence of environmental concentrations of Aroclor 1254 (10, 100, and 1000ng/L) on metamorphosis of Paralichthys olivaceus, and analyzed the mechanisms in relation to thyroid disruption. Results showed that 100 and 1000ng/L Aroclor 1254 delayed metamorphosis and that 1000ng/L Aroclor 1254 caused abnormal morphology. Thyroxine and triiodothyronine levels in the control group were significantly elevated at metamorphic climax, but treatment with 100 and 1000ng/L delayed the increase in thyroid hormones (THs) and retarded metamorphic processes. In larvae exposed to 1000ng

Early development of circadian rhythmicity in the suprachiamatic nuclei and pineal gland of teleost, flounder (Paralichthys olivaeus), embryos.

Circadian rhythms enable organisms to coordinate multiple physiological processes and behaviors with the earth's rotation. In mammals, the suprachiasmatic nuclei (SCN), the sole master circadian pacemaker, has entrainment mechanisms that set the circadian rhythm to a 24-h cycle with photic signals from retina. In contrast, the zebrafish SCN is not a circadian pacemaker, instead the pineal gland (PG) houses the major circadian oscillator. The SCN of flounder larvae, unlike that of zebrafish, however, expresses per2 with a rhythmicity of daytime/ON and nighttime/OFF. Here, we examined whether th…

Ontogeny of the digestive tract during larval development of yellowtail flounder: a light microscopic and mucous histochemical study.

The histological development and mucous histochemistry of the alimentary tract in larval yellowtail flounder were studied using light microscopy. Samples were taken when the larvae were first offered food at 3 days post-hatch, then at 7, 10, 29, 36, and 46 days post-hatch, at which time they were metamorphosing. Regional partitioning of the digestive tract into the buccal cavity, pharynx, oesophagus, post-oesophageal swelling (PES), intestine, and rectum was complete by day 10. Goblet cells were present only in the buccal cavity, pharynx and intestine by day 7, but increased in number and dist…

Sodium chloride absorption by the urinary bladder of the winter flounder. A thiazide-sensitive, electrically neutral transport system.

The urinary bladder of the winter flounder absorbs NaCl by a process independent of the transepithelial voltage. In contrast to most other epithelia which have a neutral NaCl-absorptive system, the flounder bladder has a high transepithelial resistance. This feature simplifies analysis of the cellular transport system because the rate of ion transfer through the paracellular pathway is rather low. Experiments were designed to distinguish among three possible mechanisms of neutral NaCl absorption: (a) Na/K/2Cl cotransport; (b) parallel Na/H and Cl/OH exchange; (c) and simple NaCl cotransport. A…

Comparative Genomics of Edwardsiella piscicida in the Japanese Flounder (Paralichthys olivaceus): Discovery and Implications of a Novel Genomic Island.

Edwardsiella piscicida is a significant pathogen that poses a particular threat to Japanese flounder (Paralichthys olivaceus) aquaculture in Japan and other countries. The damage is caused by the pathogen's ability to evade host immune defences and establish intracellular infections, intensified by its genomic plasticity and capacity for horizontal gene transfer. To investigate evolutionary adaptations between one older (2019) and four recent (2023) E. piscicida strains from the same geographical locations, we performed comparative genomic analysis of five isolates using high-quality hybrid ge…

Regulations of Gata-3 on CD4+ T cell differentiation via its zinc finger domains in flounder (Paralichthys olivaceus).

CD4+ T cells are central to adaptive immunity, with GATA binding protein 3 (Gata-3) acting as a key transcription factor for Th subset differentiation. This study aimed to characterize Gata-3 in flounder (Paralichthys olivaceus) and investigate its role in CD4+ T cell differentiation via its zinc finger domains (ZnFs). Flounder Gata-3 contains two conserved C4-type ZnFs and showed peak expression in gills. Immunofluorescence assays suggested that Gata-3 located in gill leukocytes, co-localized with CD4-1+/CD4-2+ cells but not CD8+ or IgM+ cells. Under in vitro Th2 differentiation, the proporti…

Chemical Attenuation of Miamiensis avidus Using Oxytetracycline Reduces Virulence and Confers Partial Protection in Olive Flounder (Paralichthys olivaceus).

Scuticociliatosis caused by the ciliate Miamiensis avidus remains a serious constraint in the intensive culture of olive flounder (Paralichthys olivaceus), yet effective vaccines are still unavailable. In this study, we evaluated chemical attenuation as a novel approach to generate a safe whole-cell vaccine candidate against M. avidus. Ciliates were attenuated by oxytetracycline (OTC) treatment, and the safety, residual virulence and protective potential of the chemically attenuated preparation were assessed in vitro and in vivo. Treatment with 5 mM OTC for 3 h suppressed ciliate proliferation…

Antibody-dependent immune response of olive flounder (Paralichthys olivaceus) induced by inactivated viral hemorrhagic septicemia virus (VHSV) vaccine.

In teleosts, vaccine-induced protection is commonly assessed using serum antibody titers; however, these measurements often fail to fully explain protective outcomes due to limited understanding of B cell differentiation and antibody effector functions. In olive flounder (Paralichthys olivaceus), the cellular basis of antibody-mediated immunity remains poorly characterized. In this study, we investigated B cell differentiation, antibody production, and antibody-dependent immune responses post-immunization with an inactivated viral hemorrhagic septicemia virus (VHSV) vaccine. Plasma exosomes from viral hemorrhagic septicemia virus-infected olive flounder (Paralichthys olivaceus): Proteomic insights and wound healing effects.

Extracellular vesicles (EVs) or their subtypes, such as exosomes, are valuable nano-biomolecules for immunotherapeutic, drug delivery, and diagnostic purposes. Freshwater and marine fish, including olive flounder (Paralichthys olivaceus), are highly susceptible to the contagious Viral hemorrhagic septicemia virus (VHSV). In this study, we aimed to determine how infection alters the biological responses by analyzing the proteomic profiles of plasma-derived exosomes from phosphate-buffered saline (PBS) injected (PBS-Exo) and VHSV challenged (VHSV-Exo) olive flounders at the initial stages of inf…
