Toxicity of free radicals to mesothelial cells and peritoneal membrane.

We studied the toxicity of free radicals to human mesothelial cells in vitro and to the peritoneal membrane of rats during peritoneal dialysis. Free radicals cause damage to mesothelial cells as measured by release of cytosolic markers such as 86Rb and lactate dehydrogenase. Vitamin E neutralized the toxic effect of free radicals in vitro. Human mesothelial cells exposed over 6 h to a mixture of essential and nonessential amino acids in medium are more vulnerable to the cytotoxic effect of free radicals than control cells exposed to medium alone. Cells exposed previously to glucose or glycerol

Role of oxygen-derived free radicals in digestive tract diseases.

Evidence is presented that supports a role of oxygen free radicals in the pathogenesis of various disorders of the digestive system. In the intestine, there is evidence that oxygen radicals play an important role in the endothelial and epithelial damage associated with certain models of ischemia. The mechanism for superoxide production in this condition differs from that described for other pathologic states (i.e., oxygen toxicity and neutrophil-mediated inflammation). This mechanism involves the reaction of xanthine oxidase, hypoxanthine, and molecular oxygen to produce a burst of oxygen radi

Free radicals as potential mediators of metal-allergy: Ni2+- and Co2+-mediated free radical generation.

The generation of free radicals by Ni(2+) and Co(2+) was studied at physiological pH in H(2)O(2)-containing solutions in the absence and presence of various radical-mediating ligands and in human peripheral blood mononuclear cell (PBMC) cultures. With ESR spectroscopy, free radical species were identified and quantitated by spin trapping with 5,5-dimethyl-1-pyrroline-N-oxide (DMPO). Co(2+) generated hydroxyl radicals from H(2)O(2) in PBS solutions containing glutathione (GSH) or histidine (His). Omission of GSH or His from the reaction mixture significantly reduced the ESR-signal, indicating t

Free-Radical Carbocyanation of Olefins.

The free-radical three-component carbocyanation of electron-rich olefins was investigated with p-tosyl cyanide as cyanide source. The scope and limitations of the process were established by varying the nature of the alkene and radical precursor. Carbocyanation of chiral allylsilanes was shown to occur with high diastereocontrol, leading to syn β-silyl nitriles. The origin of the stereocontrol was rationalized by a Felkin-Anh-type transition-state model. Finally, a tin-free carbocyanation process was also devised, based on the use of a new alkylsulfonyl cyanide incorporating both carbon fragme

Free radical reactions in the gallbladder.

The changes in the composition of bile can lead to the process of it's crystallization in the gallbladder. In bile model it was shown that inflammation with the generation of reactive oxygen metabolites may induce and influence the cholesterol monohydrate crystal formation within supersaturated bile. The aim of this study was to investigate the ability to detect traces of reactive oxygen metabolites, thiobarbituric acid reactive compounds and dien, in order to compare cholesterol and bilirubin contents in bile and serum during different conditions of inflammation in the gallbladder's wall. Novel Methods for the Formation of Free Carbyne Radicals in Solution.

Carbyne free radicals (RC) are usually associated with high-energy processes, thus research on their preparation, chemical reactivity, and prevalence under mild conditions is scarce. Recently, it was reported that metallo-complexes containing CR ligands may undergo spontaneous degradation in aqueous solutions to produce free RC radicals. These highly reactive species may react with each other to form the corresponding alkyne and other products. The reaction of 1,1,1 halo-carbo-hydrates with Cr(II) ions also forms RC radicals under mild conditions. Herein, we report two additional synthetic rou

Metal-Catalyzed Formation of Organic Pollutants Intermediated by Organic Free Radicals.

Metal compounds play important roles in the formation of organic pollutants during thermal-related processes. However, the metal-catalyzed predominant organic pollutants have not previously been characterized nor have any detailed catalytic mechanisms been clarified. Here, we preciously distinguished the multiple organic free radical intermediates on metal catalyst surfaces during the organic pollutant formation through laboratory and theoretical studies. Differences between the organic free radical intermediate species, concentrations, and formation mechanisms under the catalysis of different

Free Radicals and ROS Induce Protein Denaturation by UV Photostability Assay.

Oxidative stress, photo-oxidation, and photosensitizers are activated by UV irradiation and are affecting the photo-stability of proteins. Understanding the mechanisms that govern protein photo-stability is essential for its control enabling enhancement or reduction. Currently, two major mechanisms for protein denaturation induced by UV irradiation are available: one generated by the local heating of water molecules bound to the proteins and the other by the formation of reactive free radicals. To discriminate which is the likely or dominant mechanism we have studied the effects of thermal and

Formation of Environmentally Persistent Free Radicals on Microplastics under Light Irradiation.

Microplastics (MPs) are presumed to be inert during aging under ambient conditions. In this study, four types of virgin MPs, including polystyrene (PS), phenol-formaldehyde resin (PF), polyethylene (PE), and polyvinyl chloride (PVC), were aged under simulated solar light irradiation. Surprisingly, several environmentally persistent free radicals (EPFRs), which are considered to be a type of emerging contaminant, were detected on the irradiated PS and PF, rather than PE and PVC, by electron paramagnetic resonance (EPR) spectroscopy. Depending on the photoaging duration time, the characteristic

Detecting free radicals in sunscreens exposed to UVA radiation using chemiluminescence.

One of the current concerns with the application of nanoparticles in sunscreens, and in particular nano-TiO2 and ZnO, is their potential to photogenerate free radicals and reactive oxygen species (ROS) when they absorb ultraviolet wavelengths from sunlight. Free radicals and ROS are known to be associated with UV-induced skin damage and oxidative stress, from which sunscreens are expected to offer significant protection. Here we describe a simple method, based on chemiluminescence emission, for detecting free radicals generated in commercial sunscreens alone, and when applied to various substr

[Phosphorus-containing iminoxy and nitroxy free radicals as perspective spin labels].

A structure and magnetic parameters of a series of nitroxy and iminoxy radicals with different position of the phosphorus atom with respect to the radical center were established by means of EPR. Free radicals were obtained in the course of chemical and electrochemical reactions and also through UV-radiation with the use of "spin traps". The studied phosphorylated nitroxy and iminoxy radicals are revealed to be highly sensitive to the medium and temperature that makes them perspective "spin labels" in biological objects.

Female infertility and free radicals: potential role in adhesions and endometriosis.

Free radicals are highly reactive molecules produced in the cell either as part of, or as end-products of, biochemical reactions that have crucial roles in the homeostasis of the organism. Thus, excess production or impaired elimination of free radicals leads to increased oxidative stress, which has been implicated in the development of several different disease states, including hypoxia-reperfusion injury, cancer, and aging. Peritoneal adhesions and endometriosis are relatively commonly identified in women, and are known to be associated with infertility without clearly understood pathophysio

The role of free radicals in the pathogenesis of rheumatoid arthritis.

Free radicals are reactive chemical species that differ from other compounds in that they have unpaired electrons in their outer orbitals. They are capable of damaging cellular components, and accumulating evidence suggests that they may contribute to various disease entities including inflammatory joint disease. Reactive oxygen species (ROS) as well as reactive nitrogen species (RNS) can directly or indirectly damage basic articular constituents and lead to the clinical expression of the inflammatory arthritis. Hydroxyl radicals degrade isolated proteoglycans, and HOCl fragments collagen. Hyd

Free radicals in alcoholic myopathy: indices of damage and preventive studies.

Chronic alcoholic myopathy affects up to two-thirds of all alcohol misusers and is characterized by selective atrophy of Type II (glycolytic, fast-twitch, anaerobic) fibers. In contrast, the Type I fibers (oxidative, slow-twitch, aerobic) are relatively protected. Alcohol increases the concentration of cholesterol hydroperoxides and malondialdehyde-protein adducts, though protein-carbonyl concentration levels do not appear to be overtly increased and may actually decrease in some studies. In alcoholics, plasma concentrations of alpha-tocopherol may be reduced in myopathic patients. However, al

Alcohol-induced free radicals in mice: direct toxicants or signaling molecules?

Tumor necrosis factor alpha (TNF-alpha) and free radicals are produced in early alcohol-induced liver injury. Recently, pathology caused by alcohol was blocked nearly completely in tumor necrosis factor alpha receptor 1 (TNF-R1) knockout mice. With this model, it is now possible to evaluate whether free radicals are directly toxic or act as redox regulators of TNF-alpha production. Specifically, if free radicals were directly toxic, a parallel decrease in free radicals and pathology in TNF-R1 knockout mice would be predicted. If they only affect TNF-alpha production, radicals would be expected

[Free radicals and brain damage in newborns with hypoxic-ischemic lesion].

Free radicals (FR) are highly reactive chemical molecules containing one or more unpaired electrons. Oxygen-derived free radicals, collectively termed reactive oxygen species (ROS), are normally produced in living organisms. When over produced, they are major mediators of cell and tissue injury. There is a critical balance between free radical generation and antioxidant defenses. Oxidative stress in vivo is a degenerative process due to the over production and propagation of FR reactions. FR reactions lead to oxidation of lipids, proteins, polysaccharides and to DNA damage. Newborns and partic

[Acute hypoxia induce free radicals generation in rat's cortex and hippocampus].

Anesthetized rats were ventilated artificially to 10.5%-11.0% O2 in N2 (equivalent to altitude about 5 000 m). PBN (alpha-phenyl-ter-butylnitrone) was injected intravenously 5 min before hypoxia. The experiment results showed that the ESR (electron spin resonance) signal intensity of PBN-spin adduct in cortex and hippocampus both increased significantly during hypoxia (15 min) as compared with normoxia group (n = 5, P < 0.01). In the third group of rats injected with MPEG-SOD (monomethoxy-polyethylene glycol-superoxide dismutase) intravenously 20 min before hypoxia, the ESR signal intensity of

Sperm-damaging effects of electric current: possible role of free radicals.

The generation of reactive oxygen species (ROS) and sperm damage was evaluated in (a) samples obtained during electroejaculation (EE) of men with spinal cord injury and (b) in electrolyzed HAM's F-10 medium subjected to electric current in vitro. Chemiluminescence data showed a significant increase in ROS in the ejaculates (6 x 10(7) photons/ml) collected immediately after EE and in the electrolyzed medium (3 to 7 x 10(6) photons/ml) when compared to the control (4 to 7 x 10(4) photons/ml). Incubation of normal human sperm with the electrolyzed medium resulted in a significant threefold decrea

Are free radicals involved in the pathogenesis of idiopathic Parkinson's disease?

Since the original description of Parkinson's disease (PD) more than 170 years ago, there have been major advances in the understanding and treatment of PD. This article reviews some current knowledge concerning the role of free radicals, especially superoxide, in the etiology of PD. Hypotheses regarding inhibition of mitochondrial respiration and free radicals generation in PD are discussed in relation to the mechanisms of action of the N-methyl-4-phenyl-1,2,3,6-tetrahydropyridine, a neurotoxin known to produce parkinsonian syndromes.

Pulsed UV laser generated short-lived free radicals from biological samples.

EPR characterization of the short-lived free radicals generated by pulsed UV laser ablation of biological samples has been investigated using a spin trap method. The obtained EPR spectra suggest that the trapped short-lived free radicals generated by excimer laser ablation of collagen and myocardium are identical. The obtained results are discussed in association with the production scheme of free radicals and an empirical mechanism of laser generated short-lived free radicals.
