The gasotransmitter hydrogen sulphide decreases Na⁺ transport across pulmonary epithelial cells.

BACKGROUND AND PURPOSE The transepithelial absorption of Na(+) in the lungs is crucial for the maintenance of the volume and composition of epithelial lining fluid. The regulation of Na(+) transport is essential, because hypo- or hyperabsorption of Na(+) is associated with lung diseases such as pulmonary oedema or cystic fibrosis. This study investigated the effects of the gaseous signalling molecule hydrogen sulphide (H(2) S) on Na(+) absorption across pulmonary epithelial cells. EXPERIMENTAL APPROACH Ion transport processes were electrophysiologically assessed in Ussing chambers on H441 cell

Novel neurotherapeutic targets for substance use disorders: Neuroplasticity, neuroinflammation, gasotransmitters and non-canonical organ systems.

Substance use disorders (SUDs) are chronic disorders marked by intense and compulsive drug-seeking behavior, coupled with a high risk of relapse. Different theories have emerged over time regarding mechanisms that induce and exacerbate SUDs. This review explores new and evolving evidence on the role of reward systems and processes and aligns those conceptual frameworks with neuroplastic and neuroinflammatory mechanisms and potential therapeutic targets. As the addiction cycle progresses, substance use shifts to long-term chronic use, marked by reduced hedonic effects and intensified compulsion

Bioenergetic relevance of hydrogen sulfide and the interplay between gasotransmitters at human cystathionine β-synthase.

Merely considered as a toxic gas in the past, hydrogen sulfide (H2S) is currently viewed as the third 'gasotransmitter' in addition to nitric oxide (NO) and carbon monoxide (CO), playing a key signalling role in human (patho)physiology. H2S can either act as a substrate or, similarly to CO and NO, an inhibitor of mitochondrial respiration, in the latter case by targeting cytochrome c oxidase (CcOX). The impact of H(2)S on mitochondrial energy metabolism crucially depends on the bioavailability of this gaseous molecule and its interplay with the other two gasotransmitters. The H(2)S-producing h…

Analytical Methods for Detection of Gasotransmitter Hydrogen Sulfide Released from Live Cells.

Hydrogen sulfide (H2S) plays an important role in mammals as a signaling molecule. Recently, abnormal H2S concentration has been associated with several pathophysiological states, such as diabetes mellitus, hypertension, Alzheimer's disease, and Parkinson's disease. As regulating H2S concentration can be a very prominent way of developing new drugs, many researchers have paid great attention to H2S research. To understand the role of H2S in pathophysiology and develop H2S-based therapies, it is necessary to measure the exact concentration of H2S within biological systems. But, H2S is volatile

Gasotransmitter delivery via self-assembling peptides: Treating diseases with natural signaling gases.

Nitric oxide (NO), carbon monoxide (CO), and hydrogen sulfide (H2S) are powerful signaling molecules that play a variety of roles in mammalian biology. Collectively called gasotransmitters, these gases have wide-ranging therapeutic potential, but their clinical use is limited by their gaseous nature, extensive reactivity, short half-life, and systemic toxicity. Strategies for gasotransmitter delivery with control over the duration and location of release are therefore vital for developing effective therapies. An attractive strategy for gasotransmitter delivery is though injectable or implantab…

The role of gasotransmitters in Parkinson's disease: Interplay of nitric oxide, carbon monoxide, and hydrogen sulfide.

Parkinson's disease (PD) is a progressive neurodegenerative condition marked by dopaminergic neuron loss and α-synuclein accumulation. In recent years, a growing body of work has explored the roles of endogenous gasotransmitters-namely nitric oxide (NO), carbon monoxide (CO), and hydrogen sulfide (H2S)-in pathways relevant to PD pathology. Though traditionally viewed as toxic, these molecules may, under regulated conditions, influence oxidative stress, neuroinflammation, and mitochondrial function in ways that could be therapeutically significant. NO illustrates this complexity. At physiologic…

Interplay between gasotransmitters and potassium is a K+ey factor during plant response to abiotic stress.

Carbon monoxide (CO), nitric oxide (NO) and hydrogen sulfide (H2S) are gasotransmitters known for their roles in plant response to (a)biotic stresses. The crosstalk between these gasotransmitters and potassium ions (K+) has received considerable attention in recent years, particularly due to the dual role of K+ as an essential mineral nutrient and a promoter of plant tolerance to abiotic stress. This review brings together what it is known about the interplay among NO, CO, H2S and K+ in plants with focus on the response to high salinity. Some findings obtained for plants under water deficit an…

Modulation of inflammasome biology in age-associated neurodegenerative diseases: Therapeutic potential of endogenous gasotransmitters and synthetic molecules.

Inflammasomes, particularly the NLRP3 complex, play a central role in coordinating innate immune activation and neuroinflammatory responses within the cytosol. Persistent or dysregulated nucleotide-binding domain, leucine-rich-containing family, pyrin domain-containing-3 (NLRP3) activation promotes caspase-1-dependent maturation of interleukin (IL)-1β and IL-18 and triggers gasdermin D (GSDMD)-mediated pyroptosis, thereby contributing to the pathogenic cascades underlying Alzheimer's disease (AD) and Parkinson's disease (PD). Endogenous gasotransmitters, including hydrogen sulfide (H2S) and ni…
