[Two cases of gram-positive sepsis successfully treated with vancomycin in combination with imipenem or cilastatin].

Against gram-positive sepsis, vancomycin (VCM) was administered in combination with imipenem or cilastatin (IPM/CS). Its excellent efficacy was confirmed in 2 cases, one affected with methicillin resistant Staphylococcus aureus and another with Gemella morbillorum. By calculating the FIC index according to a checkerboard technique, the in vitro synergistic effect was also demonstrated. At the present state multi-drug resistant gram-positive infections prevailed, the combination of VCM with IPM/CS can be expected as an effective measure for treating these diseases.

Gut content microbiota dysbiosis and dysregulated lipid metabolism in diarrhea caused by high-fat diet in a fatigued state.

Previous evidence has indicated that fatigue and a high-fat diet (HFD) cause the adaptive organism responses to be dysregulated, resulting in gastrointestinal (GI) disorders. Generally, gut microbiota plays a crucial role in GI disorders. However, the impact of fatigue and an HFD on the microbiome and GI disorders remains to be fully explored. Mice were randomly divided into the control group (CCN), standing group (CSD), lard group (CLD), and standing + lard group (CSLD). Mice in the CSD and CSLD groups stood on the multiple-platform apparatus for four h per day for 14 consecutive days. From t

Microbiota of preterm infant develops over time along with the first teeth eruption.

OBJECTIVE: The temporal growth of the infant microbiome in the early years of life influences short- and long-term infant health. The aim of this longitudinal study was to investigate bacterial dynamics in the microbiome of preterm infants during tooth eruption. METHODS: Saliva samples from normally delivered (n = 24) and preterm infants (n = 31) were collected 30 days after birth and after the eruption of two primary mandibular incisors. Based on Illumina MiSeq Sequencing of the 16S rRNA gene, the dynamic microbial changes of newborns at two-time points were investigated. Meanwhile, the Huma

Impact of Chemotherapy Alone and in Combination with Immunotherapy on Oral Microbiota in Cancer Patients-A Pilot Study.

The oral cavity harbors a highly intricate and dynamic microbial ecosystem of multiple microhabitats supporting diverse microbial populations. As the second most complex microbiome in the human body, surpassed only by the gut, the oral microbiome comprises over 1000 species. Disruptions in the microbial balance have been associated with an increased risk of both oral diseases (dental caries and periodontitis) and systemic conditions, including inflammatory diseases and certain types of cancers. In our pilot study, we purified bacterial DNA from pre-treated, saponin-based, host-depleted saliva

A comparison of oral microbiome composition between highly trained competitive athletes and untrained controls.

The oral microbiome has a crucial role in nitric oxide (NO) production and contributes to oral and systemic health. This study compared oral microbiome composition and markers of NO production between highlytrained competitive athletes and inactive controls. Competitive athletes and untrained controls (N = 10 per group) were recruited. Saliva, plasma, supragingival plaque and the tongue dorsum microbiome were sampled. The microbiome was examined using long-read 16S rRNA sequencing and ozone-based chemiluminescence used to measure nitrate (NO3-) and nitrite (NO2-) levels. Weekly training durati

Comparative Analysis of Intestinal Inflammation and Microbiota Dysbiosis of LPS-Challenged Piglets between Different Breeds.

Post-weaning diarrhea is common in piglets, causing huge economic losses worldwide. Associations between LPS challenge, intestinal inflammation, and microbiota have been reported in Duroc × Landrace × Yorkshire (DLY) crossbred pigs. However, the effects of LPS challenge in other breeds remain unclear. In the current study, we performed a comprehensive comparative analysis of the effects of LPS challenge on jejunal mucosal morphology, jejunal microbial composition, and serum indexes in two pig breeds: DLY and Heigai, an indigenous Chinese breed. The results showed that LPS caused considerable d

Dynamic changes of respiratory microbiota associated with treatment outcome in drug-sensitive and drug-resistant pulmonary tuberculosis.

BACKGROUND: Respiratory microbiota is closely related to tuberculosis (TB) initiation and progression. However, the dynamic changes of respiratory microbiota during treatment and its association with TB progression remains unclear. METHODS: A total of 16 healthy individuals and 16 TB patients (10 drug-sensitive TB (DS-TB) and 6 drug-resistant TB (DR-TB)) were recruited. Sputum samples were collected at baseline for all anticipants and after anti-TB treatment at Month-6 for TB patients. High throughput 16 S RNA sequencing was used to characterize the respiratory microbiota composition. RESULT

A microbiome case-control study of recurrent acute otitis media identified potentially protective bacterial genera.

BACKGROUND: Recurrent acute otitis media (rAOM, recurrent ear infection) is a common childhood disease caused by bacteria termed otopathogens, for which current treatments have limited effectiveness. Generic probiotic therapies have shown promise, but seem to lack specificity. We hypothesised that healthy children with no history of AOM carry protective commensal bacteria that could be translated into a specific probiotic therapy to break the cycle of re-infection. We characterised the nasopharyngeal microbiome of these children (controls) in comparison to children with rAOM (cases) to identif

Respiratory microbiome and metabolome features associate disease severity and the need for doxycycline treatment in children with macrolide-resistant Mycoplasma pneumoniae-mediated pneumonia.

INTRODUCTION: Commensal bacterial community along the upper respiratory tract functions against pathogens. The host determinants of Mycoplasma pneumoniae severity should be identified against the increasing threat of macrolide-resistant M. pneumoniae (MRMP) infection. We hypothesized that respiratory microbiome is involved in the clinical manifestations of M. pneumoniae infection. METHODS: From 2017 to 2020, 92 children with MRMP-mediated pneumonia were enrolled among 845 children with community-associated pneumonia. Oropharyngeal samplings were collected within 48 h after admission. We compa

Exploration of fecal microbiota in newly diagnosed patients with inflammatory bowel disease using shotgun metagenomics.

INTRODUCTION: Dysbiosis is a key mechanism in inflammatory bowel disease (IBD) pathophysiology. Previous microbiota studies in IBD generally have involved patients treated with immunosuppressive agents, which can affect the results. We aimed to elucidate the fecal microbiota composition in newly diagnosed treatment-naïve IBD patients. METHODS: Microbiota from stool samples were investigated using shotgun metagenomics sequencing and subsequent bioinformatics analysis. RESULTS: A total of 103 patients with Crohn's disease (CD), 144 with ulcerative colitis (UC), and 49 healthy controls (HC) wer

Comparative analysis of gut microbiota in hormone-sensitive and castration-resistant prostate cancer in Japanese men.

Gut microbiota plays a crucial role in the development and progression of prostate cancer, with previous studies indicating that certain bacterial taxa are more abundant in castration-resistant prostate cancer (CRPC) compared to hormone-sensitive prostate cancer (HSPC). Notably, the composition of gut microbiota can vary significantly by geographic region, and Japanese individuals have a distinct microbial profile. However, research exploring these differences within Japanese populations remains limited. This study investigated the gut microbiota differences between Japanese men with HSPC and

Oral microbiome and risk of lung cancer: results from a two-sample mendelian randomization analysis.

BACKGROUND: Numerous studies have suggested that the oral microbiome may function as a biomarker for lung cancer screening. However, the relationship between oral microbiome and lung cancer has not been thoroughly investigated. Consequently, investigating the causal relationship between oral microbiome and lung cancer was the primary goal of this study. METHODS: We conducted a two-sample Mendelian randomization (MR) analysis to investigate the causal relationship between the oral microbiome and lung cancer. Summary statistics for the oral microbiomes were obtained from large-scale metagenome-

Colostrum of healthy Slovenian mothers: microbiota composition and bacteriocin gene prevalence.

Microbial communities inhabiting the breast milk microenvironment are essential in supporting mammary gland health in lactating women and in providing gut-colonizing bacterial 'inoculum' for their infants' gastro-intestinal development. Bacterial DNA was extracted from colostrum samples of 45 healthy Slovenian mothers. Characteristics of the communities in the samples were assessed by polymerase chain reaction (PCR) coupled with denaturing gradient gel electrophoresis (DGGE) and by quantitative real-time PCR (qPCR). PCR screening for the prevalence of bacteriocin genes was performed on DNA of

Concomitant Assessment of Oral and Gastric Microbiota Composition in Autoimmune Gastritis Patients: A Case-Control Study.

Autoimmune gastritis (AIG) in its advanced atrophic stage is characterized by reduced acid secretion, dysbiosis, and gastric cancer (GC) risk. Swallowed oral bacteria surviving in increased intragastric pH may play a carcinogenic role. Oral microbiota was linked to increased GC risk. In AIG, the concomitant assessment of oral and gastric microbiota has not yet been performed. This study aimed to investigate the oral and gastric microbiota in AIG patients to clarify the role of oral bacteria in gastric dysbiosis. A case-control study on n = 20 histologically diagnosed AIG patients and n = 20 co
