Development and validation of a new algorithm for the reclassification of genetic variants identified in the BRCA1 and BRCA2 genes.

BRCA1 and BRCA2 sequencing analysis detects variants of uncertain clinical significance in approximately 2 % of patients undergoing clinical diagnostic testing in our laboratory. The reclassification of these variants into either a pathogenic or benign clinical interpretation is critical for improved patient management. We developed a statistical variant reclassification tool based on the premise that probands with disease-causing mutations are expected to have more severe personal and family histories than those having benign variants. The algorithm was validated using simulated variants base

4D-QSAR analysis and pharmacophore modeling: electron conformational-genetic algorithm approach for penicillins.

4D-QSAR studies were performed on a series of 87 penicillin analogues using the electron conformational-genetic algorithm (EC-GA) method. In this EC-based method, each conformation of the molecular system is described by a matrix (ECMC) with both electron structural parameters and interatomic distances as matrix elements. Multiple comparisons of these matrices within given tolerances for high active and low active penicillin compounds allow one to separate a smaller number of matrix elements (ECSA) which represent the pharmacophore groups. The effect of conformations was investigated building

Clinical features of dementia associated with apolipoprotein epsilon4: discrimination with a neural network genetic algorithm.

BACKGROUND: It is unclear whether the APOE epsilon4 allele is associated with distinct clinical features in dementia. METHOD: 100 cases meeting ICD criteria for dementia were interviewed using standardized instruments and genotyped for APOE. The presence of the epsilon4 allele was used by a genetic algorithm neural network (GANN) to discriminate symptoms and signs. RESULTS: The GANN selected six features: gender, systolic blood pressure, absence of ankle tendon reflexes, history of weight loss, history of falls, and interviewer observed lability of mood. Using these features, a neural networ

A Novel Load Balancing Scheme for Satellite IoT Networks Based on Spatial-Temporal Distribution of Users and Advanced Genetic Algorithms.

Satellite IoT networks (S-IoT-N), which have been a hot issue regarding the next generation of communication, are quite important for the coming era of digital twins and the metaverse because of their performance in sensing and monitoring anywhere, anytime, and anyway, in more dimensions. However, this will cause communication links to face greater traffic loads. Satellite internet networks (SIN) are considered the most possible evolution road, possessing characteristics of many satellites, such as low earth orbit (LEO), the Ku/Ka frequency, and a high data rate. Existing research on load bala

Comparative optimization of overcurrent relay coordination in DG-integrated distribution networks: water cycle algorithm versus genetic algorithm and big bang-big crunch.

The increasing penetration of distributed generation (DG) has significantly complicated protection coordination in modern distribution networks by introducing bidirectional power flows and variable fault current levels. These challenges become more pronounced under different operating modes, particularly in islanded operation, where reduced fault current levels place severe constraints on overcurrent relay (OCR) coordination. This paper presents a comparative assessment of metaheuristic optimization techniques for coordinating overcurrent protection in DG-integrated distribution networks. Two

A 100-Year Review: Methods and impact of genetic selection in dairy cattle-From daughter-dam comparisons to deep learning algorithms.

In the early 1900s, breed society herdbooks had been established and milk-recording programs were in their infancy. Farmers wanted to improve the productivity of their cattle, but the foundations of population genetics, quantitative genetics, and animal breeding had not been laid. Early animal breeders struggled to identify genetically superior families using performance records that were influenced by local environmental conditions and herd-specific management practices. Daughter-dam comparisons were used for more than 30 yr and, although genetic progress was minimal, the attention given to p

FedGA: Genetic Algorithm-Guided Federated Learning for Medical Image Segmentation with Non-IID Features.

Federated learning (FL) enhances data privacy and compliance with data regulations by enabling multiple decentralized parties to collaboratively train machine learning models without sharing their data. This makes it an ideal paradigm for the healthcare domain. Despite its inherent privacy-by-design, FL faces performance and convergence challenges when dealing with non-independent and identically distributed (non-IID) data. Although previous studies have primarily addressed the challenges of skewed label distribution across clients, which are more effective for classification, we focus in this

Genetic algorithm-based coverage path planning for autonomous aircraft cabin cleaning by reconfigurable robot.

Designing an optimal Coverage Path Planning (CPP) framework for autonomous aircraft cabin cleaning is a critical challenge due to the time-sensitive nature of aircraft turnaround operations. Conventional domestic cleaning robots struggle to adapt to the confined and irregular cabin layouts of commercial aircraft. To address this, the paper proposes a two-stage CPP approach utilizing the reconfigurable robot. In the first stage, the robot operates in its full-size configuration to efficiently clean open regions such as aisles and galleys, skipping hard-to-access seat rows to minimize total clea

Application of Improved Genetic Algorithm Based on Voronoi Partitioning in Pseudolite Deployment for Tunnel Positioning Systems.

Reliable high-precision positioning in railway tunnels is essential for intelligent train operation and safety monitoring, yet GNSS signals are severely degraded by blockage and multipath. This paper proposes a deployment-oriented numerical framework to optimize pseudolite layouts in tunnels by explicitly modeling visibility obstruction and controlling worst-case geometry along the train trajectory. A high-fidelity 3D tunnel-train model is established, in which line-of-sight (LoS) availability is screened under vehicle occlusion and trajectory-level geometric quality is evaluated accordingly.

A steady state micro genetic algorithm for hyper-heuristic generation in one-dimensional bin packing.

The one-dimensional bin packing problem (1DBPP) is a well-known NP-hard problem in computer science and operations research that involves many real-world applications. Its primary objective is to allocate items into bins while minimizing the number of bins used. Due to the complexity of the problem, exact algorithms are often impractical for large instances, which has led to a reliance on tailored heuristics that may perform well on some instances but poorly on others. In this study, we propose a method to automatically generate selection hyper-heuristics (HHs), which are then applied to solve

Genetic algorithm type 2 fuzzy logic controller of microgrid system with a fractional-order technique.

This paper presents a hybrid approach that combines a genetic algorithm (GA)-optimized type-2 fuzzy logic controller (T2FLC) with a fractional-order technique for enhanced control of a microgrid system. The T2FLC approach is employed to handle the inherent uncertainties in the microgrid due to fluctuating renewable energy inputs and varying loads. The GA optimizes the parameters of the designed FO-T2FLC approach, ensuring optimal performance under different operational conditions. This developed strategy is a modification and development of the traditional approach, as it is characterized by r

Exploring Imaging Genetic Markers of Alzheimer's Disease Based on a Novel Nonlinear Correlation Analysis Algorithm.

Alzheimer's disease (AD) is an irreversible neurological disorder characterized by insidious onset. Identifying potential markers in its emergence and progression is crucial for early diagnosis and treatment. Imaging genetics typically merges genetic variables with multiple imaging parameters, employing various association analysis algorithms to investigate the links between pathological phenotypes and genetic variations, and to unearth molecular-level insights from brain images. However, most existing imaging genetics algorithms based on sparse learning assume a linear relationship between ge

A multiparametric computational algorithm for comprehensive assessment of genetic mutations in mucopolysaccharidosis type IIIA (Sanfilippo syndrome).

Mucopolysaccharidosis type IIIA (MPS-IIIA, Sanfilippo syndrome) is a Lysosomal Storage Disease caused by cellular deficiency of N-sulfoglucosamine sulfohydrolase (SGSH). Given the large heterogeneity of genetic mutations responsible for the disease, a comprehensive understanding of the mechanisms by which these mutations affect enzyme function is needed to guide effective therapies. We developed a multiparametric computational algorithm to assess how patient genetic mutations in SGSH affect overall enzyme biogenesis, stability, and function. 107 patient mutations for the SGSH gene were obtaine

Transcriptome-guided amyloid imaging genetic analysis via a novel structured sparse learning algorithm.

MOTIVATION: Imaging genetics is an emerging field that studies the influence of genetic variation on brain structure and function. The major task is to examine the association between genetic markers such as single-nucleotide polymorphisms (SNPs) and quantitative traits (QTs) extracted from neuroimaging data. The complexity of these datasets has presented critical bioinformatics challenges that require new enabling tools. Sparse canonical correlation analysis (SCCA) is a bi-multivariate technique used in imaging genetics to identify complex multi-SNP-multi-QT associations. However, most of the
