Gliotoxin Suppresses Macrophage Immune Function by Subverting Phosphatidylinositol 3,4,5-Trisphosphate Homeostasis.

UNLABELLED: Aspergillus fumigatus, an opportunistic fungal pathogen, spreads in the environment by releasing numerous conidia that are capable of reaching the small alveolar airways of mammalian hosts. In otherwise healthy individuals, macrophages are responsible for rapidly phagocytosing and eliminating these conidia, effectively curbing their germination and consequent invasion of pulmonary tissue. However, under some circumstances, the fungus evades phagocyte-mediated immunity and persists in the respiratory tree. Here, we report thatA. fumigatusescapes macrophage recognition by strategical

Genome-wide analysis of the HDAC gene family in Pyrus betulifolia and functional validation of PcHDAC5 and PcHDT1.2 in Pyrus calleryana seed dormancy release.

Histone deacetylation, catalyzed by histone deacetylases (HDACs), is a core epigenetic regulatory mechanism in plants. However, no systematic characterization of the HDAC gene family has been reported in pear (Pyrus spp.), a globally important temperate fruit crop. In this study, we performed genome-wide identification and functional characterization of HDAC genes in pear, with a specific focus on their regulatory roles in seed dormancy release. We identified 22 PbHDAC genes from the genome of Pyrus betulifolia Bunge, which were classified into three conserved subfamilies. Evolutionary analysi

Siderophore-producing Aspergillus species as multifunctional bioinoculants for sustainable cultivation of cowpea (Vigna unguiculata (L.) Walp.).

The excessive reliance on synthetic fertilizers and pesticides has undeniably enhanced crop productivity; however, it has also led to long-term environmental degradation, highlighting the urgent need for sustainable alternatives. Bioinoculants, particularly siderophore-producing fungi, represent a potential eco-friendly approach to improving nutrient acquisition, plant health, and resistance to pathogens. In the present study, 23 fungal species were isolated from agricultural and non-agricultural soils, among which four exhibited plant growth-promoting fungal (PGPF) activity. Of these, Aspergi

Probiotic Levilactobacillus brevis GH1 from baijiu Daqu enhanced γ-aminobutyric acid and antioxidant ability in Cordyceps militaris fruiting body extract and brown rice beverage.

γ-Aminobutyric acid (GABA) is a functional amino acid with various physiological activities. Increasing the concentration of GABA in foods and enhancing dietary supplementation are important for health benefits. Baijiu, a traditional Chinese liquor, is one of the oldest and most consumed fermented beverages. Daqu, the fermentation starter of Baijiu, contains functional lactic acid bacteria. However, whether these lactic acid bacteria have the potential to prepare functional beverages enriched with GABA is not clear. In this study, Daqu was used as a screening resource, and fruiting body extrac

Microemulsion formulation of Carbendazim and its in vitro antifungal activities evaluation.

The fungus Rhizoctonia solani Kuhn is a widespread and destructive plant pathogen with a very broad host range. Although various pathogens, including R. solani, have been traditionally controlled using chemical pesticides, their use faces drawbacks such as environmental pollution, development of pesticide resistance, and other negative effects. Carbendazim is a well-known antifungal agent capable of controlling a broad range of plant diseases, but its use is hampered by its poor aqueous solubility. In this study, we describe an environmentally friendly pharmaceutical microemulsion system using

Activated M-phase-promoting factor (MPF) is exported from the nucleus of starfish oocytes to increase the sensitivity of the Ins(1,4,5)P3 receptors.

Starfish oocytes that are extracted from the ovaries are arrested at the prophase of the first meiotic division. At this stage of maturation, they are characterized by a large nucleus called the germinal vesicle. Meiosis resumption (maturation) can be induced in vitro by adding the hormone 1-methyladenine (1-MA) to the seawater in which the oocytes are suspended. Earlier work in our laboratory had detected Ca(2+) increases in both the cytoplasm and the nucleus of the oocytes approx. 2 min after the 1-MA challenge. The nuclear Ca(2+) increase was found to be essential for the continuation of th

Glycolysis-Dependent Fatty Acid Metabolism Supports Human Tfh Cell Function.

Follicular helper T (Tfh) cells orchestrate germinal center (GC) derived humoral immunity by providing essential help to B cells. Despite their key role in humoral immunity, the metabolic cues that shape Tfh cell differentiation and functions in humans remain poorly understood. Here, using a human ex vivo Tfh cell differentiation model, we investigated the metabolic requirements of Tfh cell development and function through targeted metabolic perturbation of glycolysis and fatty acid metabolism, transcriptomic analyses, and metabolic rescue experiments. Unexpectedly, early glycolytic inhibition

Exacerbation of autoimmune arthritis by copolymer-I through promoting type 1 immune response and autoantibody production.

Copolymer-I (COP-I) is an unique immune regulatory polymer that has been shown to suppress experimental autoimmune encephalomyelitis (EAE) and is a treatment option for multiple sclerosis (MS). To investigate whether its immune suppressive effects can be extended to other autoimmune diseases, we treated mice with COP-I during the induction of collagen-induced arthritis (CIA). Our results show that COP-I treatment exacerbated CIA, leading to faster onset, more severe and longer-lasting disease. The mechanisms underlying the exacerbation of CIA by COP-I treatment include enhanced activation and
