Vicia ervilia lectin (VEA) has an antibiofilm effect on both Gram-positive and Gram-negative pathogenic bacteria.

Bacterial growing resistance to antibiotics poses a critical threat to global health. This study investigates, for the first time, the antib … entration ranging from 100 to 500 µg/ml, the VEA antibiofilm activity was more relevant against the Gram-positive bacteria Streptococcus aureus and Staphylococcus epidermidis, whose biofilm was reduced up to 50% by VEA purified from accessions #5 and #36. VEA antibiofilm variability between accessions was observed, likely due to co-purified small molecules rather than d …

The Etiological Bacterial Spectrum of Neck Abscesses of Lymph Node Origin - Gram-Positive and Gram-Negative Bacteria.

According to our medical practice as maxillofacial and oral surgeons, operated patients with purulent infections of the cervical lymph nodes are not many in number. On the other hand, the presence of a purulent infection requires not only the surgical evacuation of the pus but also the application of antimicrobial preparations. This necessitates good knowledge of the spectrum of the bacterial causative agents of the disease, the determination and analysis of which is the purpose of this original article. The bacteria studied in 181 patients with a mean age of 26.25 years, ranging between 29 da

Light-controlled inhibition of Gram-positive bacteria by photoswitchable amphiphilic lipids (PALs).

Amphiphilic lipids are potent membrane-disrupting antibacterials, but their activity cannot normally be modulated after administration. Here we report photoswitchable amphiphilic lipids (PALs) that enable reversible, light-controlled modulation of antibacterial potency through photoinduced changes in molecular conformation. The PALs incorporate azobenzene photoswitches linking quaternary ammonium headgroups to variable alkyl chains, allowing their geometry and membrane affinity to be tuned by trans-cis isomerisation. Systematic studies against multidrug-resistant Gram-positive bacteria reveale…

Visualizing extracellular vesicle biogenesis in gram-positive bacteria using super-resolution microscopy.

BACKGROUND: Recently, bacterial extracellular vesicles (EVs) have been considered to play crucial roles in various biological processes and have great potential for developing cancer therapeutics and biomedicine. However, studies on bacterial EVs have mainly focused on outer membrane vesicles released from gram-negative bacteria since the outermost peptidoglycan layer in gram-positive bacteria is thought to preclude the release of EVs as a physical barrier. RESULTS: Here, we examined the ultrastructural organization of the EV produced by gram-positive bacteria using super-resolution stochasti…

Detection of toxins involved in foodborne diseases caused by Gram-positive bacteria.

Bacterial toxins are food safety hazards causing about 10% of all reported foodborne outbreaks in Europe. Pertinent to Gram-positive pathogens, the most relevant toxins are emetic toxin and diarrheal enterotoxins of Bacillus cereus, neurotoxins of Clostridium botulinum, enterotoxin of Clostridium perfringens, and a family of enterotoxins produced by Staphylococcus aureus and some other staphylococci. These toxins are the most important virulence factors of respective foodborne pathogens and a primary cause of the related foodborne diseases. They are proteins or peptides that differ from each o

Predicting and preventing antimicrobial resistance utilizing pharmacodynamics: Part I gram positive bacteria.

INTRODUCTION: Antimicrobial resistance is a potentially inevitable consequence of widespread use of antibiotics in the healthcare system. An enhanced understanding of pharmacodynamic (PD) targets that prevent antimicrobial resistance development will improve currently availably therapies and help to guide future drug development strategies. Current in vitro methods to predict bacterial resistance to antimicrobials consist of serial dilution experiments, determination of the mutant prevention concentration (MPC), mutant selection window (MSW), and human simulated pharmacodynamics studies. Distribution of mef(A) in gram-positive bacteria from healthy Portuguese children.

We screened 615 gram-positive isolates from 150 healthy children for the presence of the erm(A), erm(B), erm(C), erm(F), and mef(A) genes. The mef(A) genes were found in 20 (9%) of the macrolide-resistant isolates, including Enterococcus spp., Staphylococcus spp., and Streptococcus spp. Sixteen of the 19 gram-positive isolates tested carried the other seven open reading frames (ORFs) described in Tn1207.1, a genetic element carrying mef(A) recently described in Streptococcus pneumoniae. The three Staphylococcus spp. did not carry orf1 to orf3. A gram-negative Acinetobacter junii isolate also c

Horizontal spread of mer operons among gram-positive bacteria in natural environments.

Horizontal dissemination of the genes responsible for resistance to toxic pollutants may play a key role in the adaptation of bacterial populations to environmental contaminants. However, the frequency and extent of gene dissemination in natural environments is not known. A natural horizontal spread of two distinct mercury resistance (mer) operon variants, which occurred amongst diverse Bacillus and related species over wide geographical areas, is reported. One mer variant encodes a mercuric reductase with a single N-terminal domain, whilst the other encodes a reductase with a duplicated N-ter

Immunoglobulin G N-glycosylation predicts outcome in sepsis caused by pathogenic Gram-negative bacteria and Gram-positive bacteria: a nested case-control study.

BACKGROUND: Sepsis remains a critical global health challenge with high mortality. Rapid distinction between Gram-positive and Gram-negative pathogens is critical for empiric antibiotic selection, yet reliable biomarkers for such pathogen stratification are lacking. Immunoglobulin G (IgG) N-glycosylation modulates inflammatory responses in various diseases, suggesting its potential role in sepsis pathogenesis and prognosis. METHODS: In this nested case-control study, 180 septic patients (100 Gram-negative, 80 Gram-positive) and 100 healthy controls were enrolled. IgG N-glycosylation was analy

Lipoteichoic acid as a key wound healing mediator in Gram-positive bacteria.

Lactobacillus species are probiotic Gram-positive bacteria known to exert regenerative effects on cutaneous wounds. However, the key components and underlying mechanisms responsible for these effects remain poorly defined. To identify the bioactive components responsible for wound healing, lipoteichoic acid (LTA), lipoprotein, and peptidoglycan were purified from Lactiplantibacillus plantarum and the purified molecules were administered intradermally in a murine excisional wound splinting model. Among them, only LTA significantly accelerated dermal wound healing. Histological analysis confirme…

Nigericin is effective against multidrug resistant gram-positive bacteria, persisters, and biofilms.

Multidrug-resistant (MDR) bacteria pose a significant clinical threat to human health, but the development of antibiotics cannot meet the urgent need for effective agents, especially those that can kill persisters and biofilms. Here, we reported that nigericin showed potent bactericidal activity against various clinical MDR Gram-positive bacteria, persisters and biofilms, with low frequencies of resistance development. Moreover, nigericin exhibited favorable in vivo efficacy in deep-seated mouse biofilm, murine skin and bloodstream infection models. With Staphylococcus aureus, nigericin disrup…

Genetic determinants of antimicrobial resistance in Gram positive bacteria from organic foods.

Bacterial biocide resistance is becoming a matter of concern. In the present study, a collection of biocide-resistant, Gram-positive bacteria from organic foods (including 11 isolates from genus Bacillus, 25 from Enterococcus and 10 from Staphylococcus) were analyzed for genes associated to biocide resistance efflux pumps and antibiotic resistance. The only qac-genes detected were qacA/B (one Bacillus cereus isolate) and smr (one B. cereus and two Staphylococcus saprophyticus isolates). Efflux pump genes efrA and efrB genes were detected in Staphylococcus (60% of isolates), Bacillus (54.54%) a…
