The occurrence of a granulocytic sarcoma before any signs of leukemia is unusual and may pose confusing diagnostic problems. This report deals with such a case who presented with a mediastinal thymic tumor and superior vena cava syndrome. The authors emphasize the localization and the histological techniques that are utilized to ascertain the diagnosis.

Newer cell separators can collect more PMN, especially using macromolecular agents and steroid stimulation of donors. Proper indications for PMN transfusion are severe neutropenia and sepsis, or certain localized infections, when prolonged marrow aplasia is expected. Optimal dosage is at least 2 x 10(10) PMN per day, probably more. There is a need for more accurate definition of clinical indications for PMN support, for still greater PMN yields from leukapheresis, and for better PMN storage capability.

Granulocytes for transfusion can now be obtained from normal donors by one of four techniques that involve either centrifugation or reversible adhesion of granulocytes to nylon fibers. The leukapheresis process appears to be safe for donors and standards for the selection and care of donors are being formulated. It appears desirable to transfuse granulocytes that are compatible in a leukoagglutination crossmatching, however, better methods for histocompatibility testing must be developed. Granulocyte transfusions clearly are of benefit to patients with Gram-negative sepsis and granulocyte coun…

Clinical parameters and laboratory studies including granulocyte-associated IgG were documented in 15 patients with Felty's syndrome treated by splenectomy. Five patients did not benefit from splenectomy, six responded partially, and four completely. Response to splenectomy could not be predicted from age, sex, splenomegaly, preoperative granulocyte count, platelet count, lymphocyte count, bone marrow lymphocytosis, or granulocyte bound IgG. In contrast, marked elevation in preoperative serum granulocyte binding IgG predicted response to splenectomy. Furthermore, a postoperative fall in serum

BACKGROUND: Gastrointestinal involvement is common in eosinophilic granulomatosis with polyangiitis (EGPA) and has a poor prognosis.
OBJECTIVES: The aim of our study was to determine the density of eosinophils in the gastrointestinal tract that should be considered pathological in patients with EGPA, and how clinically and prognostically relevant intramucosal eosinophil accumulation is in patients with EGPA.
METHODS: Forty-nine subjects were included in the study: 18 with EGPA (9 women) and as a control group 31 with other rheumatological inflammatory diseases. Two hundred and forty-four bio

AIM: To determine whether the IG count (#) and IG percentage (%) are associated with disease activity in rheumatoid arthritis (RA).
METHODS: This retrospective study included 65 RA patients and 65 healthy controls. Clinical and demographic characteristics of controls and RA patients (at active period and when the patients achieved remission) were obtained from medical records. Disease activity was defined by disease activity score 28 (DAS28). Furthermore, the clinical disease activity index (CDAI), and simple disease activity index (SDAI) were calculated. For the differential diagnosis of RA

BACKGROUND: Treatment of brain tumors, epilepsy, or hemodynamic abnormalities requires a craniotomy to access the brain. Nearly 1 million cr … tence. In addition, the role of G-MDSC-derived IL-10 on neutrophil activity was examined.
RESULTS: Granulocytes (neutrophils and G-MDSCs) were the major producers of IL-10 during craniotomy infection. Bacterial burden was significantly reduced in IL-10 KO mice in the brain and galea at day 14 post-infection compared to WT animals, concomitant with increased C …

Sporotrichosis is a cutaneous mycosis that affects humans and animals and has a worldwide distribution. This infection is mainly caused by Sporothrix schenckii, Sporothrix brasiliensis, and Sporothrix globosa. Current research about anti-Sporothrix immunity has been mainly focused on S. schenckii and S. brasiliensis, using different types of human or animal immune cells. Granulocytes are a group of cells relevant for cytokine production, with the capacity for phagocytosis and the generation of neutrophil extracellular traps (NETs). Considering their importance, this study aimed to compare the …

For biomedical applications, it is important to know, which kinds of blood cells can capture quantum dots (QDs). The maximum accumulation of QDs was found for the monocyte fraction of leukocytes, the minimum binding of QDs was observed for lymphocytes. It was found that CdSe/ZnS-MPA QDs are actively absorbed by the cells and have more expressed toxicity. The classical mechanism of the phagocytosis of QDs was revealed for neutrophils, when the QDs are located in phagolysosomes. The capture of QDs by neutrophil granulocytes has resulted in a destruction of certain types of QDs. The interaction o…

The aetiology of paroxysmal nocturnal haemoglobinuria (PNH) is a somatic mutation in the X-linked phosphatidylinositol glycan class A gene (PIGA), resulting in global deficiency of glycosyl phosphatidylinositol-anchored proteins (GPI-APs). This study applied RNA-sequencing to examine functional effects of the PIGA mutation in human granulocytes. CXCR2 expression was increased in GPI-AP- compared to GPI-AP+ granulocytes. Macrophage migration inhibitory factor, a CXCR2 agonist, was significantly higher in plasma of PNH patients. Nuclear factor-κB phosphorylation was upregulated in GPI-AP- compar…

Eosinophil granulocytes have been implicated in anticancer immunity but recent data indicate that eosinophils can also promote cancer. Herein, we studied eosinophils in skin lesions from 43 patients with mycosis fungoides (MF). The presence of eosinophils correlated with disease stage: 78% of patients with advanced disease displayed eosinophil infiltration, whereas this was only seen in 11% of patients with patches (p<0.01), and in 48% of those with plaque disease. Importantly, 72% of patients with positive staining for phospho-signal-transducer-and-activator-of-transcription (pY-STAT3) in mal…

Rapid activation of immune responses is necessary for antibacterial defense, but excessive immune activation can result in life-threatening septic shock. Understanding how these processes are balanced may provide novel therapeutic potential in treating inflammatory disease. Fc receptors are crucial for innate immune activation. However, the role of the putative Fc receptor for IgM, known as Toso/Faim3, has to this point been unclear. In this study, we generated Toso-deficient mice and used them to uncover a critical regulatory function of Toso in innate immune activation. Development of innate

Dynamic cytochemical studies of neutrophilic granulocyte functional activities in erysipelas patients have revealed that reduced level of cationic proteins and unbalanced activities of the respiratory enzymes help assess the disease severity, and if these disorders persist during the convalescence period and between the recurrences, they indicate a reduction of the nonspecific resistance and help predict the recurrences of the condition.

Gamma-camera imaging after injection of radiolabelled autologous leucocytes can be very helpful in the diagnosis, localization and further clinical treatment of inflammatory diseases. We present a technique allowing sterile separation of white blood cells and labelling with 99mTc-phytate or -oxine and with 111In-oxine, -oxine sulphate or -tropolone. The method is non-invasive and the radiation dose amounts to less than 80 mrad using 100 microCi 111Indium. The use of radiolabelled granulocytes is of particular diagnostic value in patients with septicaemia of unknown origin. Whole body scanning …
