Intracellular acidosis in human and experimental brain injury.

Brain tissue acidosis is considered to play a role in the complex sequence of events following traumatic brain injury. This report reviews the experimental and clinical research conducted at the Medical College of Virginia to help clarify the extent of metabolic derangement that occurs and to evaluate the effect of treatment. Experimental injury models in ventilated animals showed that trauma produces a mild brain tissue acidosis that recovers within hours of injury. Hypoxia combined with trauma produces a relative ischemia and exacerbates the acidosis, which eventually resolves with resuscita

Vascular abnormalities in experimental and human lymphatic filariasis.

Whereas clinical descriptions of grotesque lymphedema and standard light microscopy in human filariasis have elucidated the natural progression of this disease, the link between the nematode and vascular abnormalities including elephantiasis remains poorly understood. Accordingly, we examined the nature and distribution of lymphatic and blood vascular derangements in a variety of tissues and organs from 37 ferrets acutely and chronically infected with Brugia malayi and in 15 patients with Wuchereria bancrofti or Brugia malayi infestation (resected skin, subcutaneous tissue, and lymph nodes) us

Atrial natriuretic factor in experimental and human hypertension.

Plasma levels of immunoreactive atrial natriuretic factor (IR-ANF) were evaluated by radioimmunoassay in several models of experimental hypertension and in human hypertension. Plasma levels of IR-ANF are consistently increased in all models of experimental hypertension studied so far. This is accompanied by a decrease of IR-ANF levels in the left atrium at certain times. Plasma levels of IR-ANF are not increased in human essential hypertension, except in the severe form (diastolic blood pressure above 110 mmHg). Peripheral levels of IR-ANF in renovascular hypertension do not differ from normal

Superoxide in ocular inflammation: human and experimental uveitis.

A possible involvement of superoxide in the pathogenesis of uveal inflammation in man and experimental animals was investigated. Superoxide production by the leukocytes of Behcet patients was significantly higher in the attack phase than in the remission phase. Leukocyte superoxide generation was also enhanced in guinea pigs with S-antigen-induced experimental autoimmune uveoretinitis (EAU). If the animals were treated with superoxide dismutase (SOD) at the onset of EAU, aqueous humor cell count was significantly lower than that of control (i.e., without SOD treatment). Infiltration of the inf

A brief history of experimentation on condemned and executed humans.

Experimentation on condemned men is assumed to have been a common practice in ancient Alexandria, but disappeared in Rome and during the Middle Ages. Sporadic cases were documented in the Renaissance and afterward, involving experiments both before and immediately after execution. The advent of the guillotine raised the question of possible persistence of consciousness after execution and that spurred much electrophysiological study of freshly decapitated heads and bodies. In 19th-century Europe, interest focused on cardiac function immediately after beheading. In the early 20th century, many

19-Nor-corticosteroids in experimental and human hypertension.

Recent reports demonstrate that the 19-nor-corticosteroids (19-nor-DOC) are naturally-occurring substances in hypertensive animal models as well as man. Since some 19-nor-corticosteroids are potent mineralocorticoids, they may have a role in regulating systemic arterial pressure and be involved in the pathogenesis of hypertension. This paper reports the probable biosynthetic pathway, factors regulating the secretion or production, and measurement of 19-nor-DOC in man and the spontaneously hypertensive rat (SHR). These studies demonstrate (1) 19-nor-DOC is greatly influenced by ACTH and dexamet

Experimental studies of the deposition of particles in the human lungs.

Measurements of deposition, De, during steady breathing through the mouth of particles between 0.5 and 2.5 micons diameter and density 0.92 g/cm3 show that it is related to the tidal volume, VT, expiratory reserve volume, Vr, and a "normal" volume, Vro, by the formula 100 De= Vt/B+C/FD- E- Vr-Vro/A where B, C, D and E are constants which depend on particle size but do not vary from subject to subject enough to have much influence on corresponding values of De. A and and Vro are independent of particle size but relate to the individual subject, especially Vro. A new method is described of corre

Experimental dissection of tuberculosis protective immunity: a human perspective.

Mycobacterium tuberculosis (Mtb), the causative agent of tuberculosis (TB), has plagued humankind for millennia. Claiming 1.25 million lives in 2023, TB remains the worldwide leading cause of death from a single-infectious agent. Improved vaccines, diagnostics and treatment regimens for drug-susceptible and drug-resistant cases are paramount to attain the goals of the WHO's End TB Strategy. Our knowledge gap in protective immunity in TB impedes the development of such new vaccines and host-directed interventions. Mtb is a pathogen highly adapted to humans and primarily infects the lungs. Acces

Experimental Infection in Mice with Cryptosporidium Isolated from Humans.

Cryptosporidium is a genus of protozoa that infects the gastrointestinal and respiratory epithelium of various host species. The aim of this study was to perform experimental infection in conventional mice with three Cryptosporidium species isolated from humans. The three Cryptosporidium species, namely Cryptosporidium canis, Cryptosporidium parvum, and Cryptosporidium ryanae, were obtained from fecal samples collected from patients hospitalized in an infectious disease hospital. The mice, from 10-day-old litters kept with their mother, were divided into three groups and orally infected with o

Overexpression of TBX3 suppresses tumorigenesis in experimental and human cholangiocarcinoma.

TBX3 behaves as a tumor suppressor or oncoprotein across cancer. However, TBX3 function remains undetermined in intrahepatic cholangiocarcinoma (iCCA), a deadly primary liver malignancy with few systemic treatment options. This study sought to investigate the impact of TBX3 on iCCA. We found that overexpression of TBX3 strongly inhibited human iCCA cell growth. In the Akt/FBXW7ΔF mouse iCCA model, overexpression of Tbx3 reduced cholangiocarcinogenesis in vivo, while inducible genetic knockout of Tbx3 accelerated iCCA growth. RNA-seq identified MAD2L1 as a downregulated gene in TBX3-overexpress

CGRP release in an experimental human trigeminal pain model.

BACKGROUND: Migraine and trigemino-autonomic cephalalgia attacks are associated with an increase of α-calcitonin-gene related peptide levels in the ipsilateral jugular vein. It is however unknown whether trigeminal pain stimulation in healthy subjects without headache disorders also induces increase of calcitonin-gene related peptide levels. FINDINGS: We measured α-calcitonin-gene related peptide levels in eight healthy subjects after subcutaneous injection of capsaicin in the forehead and in the mandibular region and after injection of sodium chloride in the forehead. We observed a significa

The effects of human lactoferrin in experimentally induced systemic candidiasis.

Introduction. Candida albicans is responsible for several types of oral and systemic infections. In light of emerging resistance to antifungals, studies have demonstrated the antifungal effect of lactoferrin (LF), which is part of the innate immune system, has anticandidal activities.Methodology. C. albicans (2×106 c.f.u. ml-1) were incubated either with PBS or human LF (hLF) (100 µg ml-1) at 37 °C for 24 h and then RNA was isolated and virulence factors analysed. C. albicans (1×105 c.f.u.) was injected into the tail vein of immunocompromised wild-type and Ltf -/-. Then, 24 h later, the Ltf -/
