Visual particulate was observed to form in parenteral hydroxyethylstarch over 24 months during stability studies. The particulate was identified as being amylose using FT-IR microscopy. Amylose values were measured in the bulk drug using the starch-iodine reaction. The amylose numbers measured in the bulk drug were found to correlate well with the measured particulate values in the parenteral formulation.

The 6% solution of hydroxyethyl starch (MS = 0.47-0.7, Mw = 120,000-220,000) prepared at the Department of Biochemistry, Institute of Haematology in Warsaw was administered in infusions to a group of surgical patients. It was found that the preparation complied with the conditions and requirements for plasma-replacing agents. In none of the treated patients any side effects were observed. Further investigations will be continued infusing greater volumes of HES to patients with hypovolaemia of different origin.

Male rats of the Wistar strain survived an isovolaemic blood volume replacement down to a haematocrit value of 20% using commercial dextran 60 or hydroxyethyl starch. No significant changes of the blood volume have been observed. 14 days after treatment, the blood values were found to be practically within the normal range. As early as 3 days after the blood exchange, a significant increase in reticulocytes has been noted which reached its maximum on the 7th day. In both series the blood regeneration was identical.

A quantitative gas chromatographic method for the determination of the sodium (caprylate) octanoate, antimicrobial in the plasma volume expanders hydroxyethyl starch and human serum albumin, has been developed. Based on the result of randomized controlled trials and meta-analysis recently, the infusion of hydroxyethyl starch (HES) was not shown to over match routine crystalline solution in exerting resuscitation effect against hypovolemia of patients with burn shock, severe systematic infection, or other critical conditions, on the other hand, it may induce renal toxicity and other toxic and side effects.
