Histiocytic/dendritic cell tumors are rare in clinical practice. It is postulated that they originate from bone marrow stem cells. Accumulat … t in both patients. In one case, chronic lymphocytic leukemia/small lymphocytic lymphoma (CLL/SLL), Langerhans cell sarcoma, and histiocytic sarcoma coexisted in the lymph nodes. 11q22 deletion often present in CLL/SLL and expression of the BRAF V600E gene was detected in all the 3 components. In the other case, there diffuse large B-cell lymphoma and histiocytic sarcoma …

A hybrid histiocytic sarcoma-interdigitating dendritic cell sarcoma was found in a small perinephric lymph node of an asymptomatic 80-year-old man, who presented a year ago with two small foci of lung metastasis found during routine chest X-ray. Fine needle aspiration cytology demonstrated interconnecting long and thin cell processes radiating from dendrite-like neoplastic cells with oval, enlongated, reniform, and irregular nuclei with vesicular chromatin and distinct nucleoli. Histology showed spindled epitheliod and histiocytic cells with abundant, slightly eosinophilic cytoplasm with indis

Neoplasms and pseudotumors of the temporomandibular joint (TMJ) are very uncommon. Early recognition of such will prevent therapeutic delay and may have a dramatic impact on patient morbidity and mortality. Included in rare TMJ lesions are the following: 1) synovial chondromatosis, 2) osteochondroma, 3) osteoma, 4) osteoblastoma, 5) pigmented villonodular synovitis, 6) ganglion, 7) synovial cyst, 8) simple bone cyst, 9) aneurysmal bone cyst, 10) epidermal inclusion cyst, 11) hemangioma, 12) nonossifying fibroma, 13) Langerhans cell histiocytosis, 14) plasma cell myeloma, 15) sarcoma.

Langerhans cell sarcoma (LCS), malignant indeterminate dendritic cell tumor (IDCT), and interdigitating dendritic cell sarcoma (IDCS) are rare malignant neoplasms that originate from antigen-presenting conventional dendritic cells (cDCs). These tumors, collectively termed cDC sarcoma in this study, have been reported only rarely in association with myeloid malignancies. We collected five cases of cDC sarcoma coexisting with acute myeloid leukemia (AML) or myelodysplastic neoplasm with increased blasts (MDS-IB), conducted clinicopathologic and genetic analyses, and compared these with four repo…

Small round cell lesions of the bone encompass a heterogeneous group of tumors and tumor-like lesions, including Ewing sarcoma, small cell osteosarcoma, mesenchymal chondrosarcoma, neuroblastoma, non-Hodgkin's lymphoma (NHL), "Ewing-like" undifferentiated round cell sarcomas, metastasizing small cell carcinoma, along with plasma cell dyscrasia and Langerhan's cell histiocytosis. At the same time, there are tumor mimics, for example, chronic osteomyelitis, which has overlapping radiologic features with Ewing sarcoma and a primary intraosseous NHL. An exact diagnosis necessitates integration of

Histiocytic sarcoma (HS) is a rare, aggressive non-Langerhans histiocytic cell neoplasm of hematopoietic origin. Histiocytic sarcoma is prone to early systemic metastasis, rendering early diagnosis and treatment critical determinants for patient outcome. Primary HS originating from the central nervous system (CNS) is exceptionally rare and portends a poor prognosis. This grim clinical course is further complicated by the challenging diagnosis and the lack of standard treatment guidelines for the disease. This is due to the exceptionally rare nature of primary CNS histiocytic sarcoma and the li

Recent studies have demonstrated recurrent activating mutations involving the classical MAPK and PI3K signaling pathways in a large proportion of histiocytic neoplasms, such as Langerhans cell histiocytosis. However, very little is known about the molecular genetics of histiocytic sarcoma, a rare aggressive malignant neoplasm that shows pathologic characteristics of mature macrophages. Here we report the genomic characteristics of a large cohort of histiocytic sarcomas (n = 28) using a targeted next-generation sequencing approach to identify driver alterations. We identified recurrent mutation

PURPOSE OF REVIEW: Since the discovery of B-Raf proto-oncogene (BRAF) V600E mutations in histiocytic neoplasms, diverse kinase alterations have been uncovered in BRAF V600E-wildtype histiocytoses. The purpose of this review is to outline recent molecular advances in histiocytic neoplasms and discuss their impact on the pathogenesis and treatment of these disorders.
RECENT FINDINGS: Activating kinase alterations discovered in BRAF V600E-wildtype Langerhans (LCH) and non-Langerhans cell histiocytoses (non-LCH) result in constitutive activation of the mitogen-activated protein kinase and/or phos

In the Reticulo-Endothelial System (RES), named at present Accessory Immune System (AIS), two chief groups of cells can be recognized: 1--Antigen-presenting cells: Dendritic cells 2--Antigen-processing cells: Macrophages (or Histiocytic cell) True malignant Langerhans cell neoplasm has not been yet clearly demonstrated. Finally, it is important to note that marcrophages and dentritic cells are commonly observed as relative cells in numerous malignant tumors (carcinomas, sarcomas, lymphomas, etc) However, their precise role has yet to be determined.

The predominant types of dendritic cells (DC) in the skin and mucosa are Langerhans cells (LC) and interstitial dermal DC (iDDC). LC and iDDC process cutaneous antigens and migrate out of the skin and mucosa to the draining lymph nodes to present antigens to T and B cells. Because of the strategic location of LC and iDDC and the ability of these cells to capture and process pathogens, we hypothesized that they could be infected with human herpesvirus 8 (HHV-8) (Kaposi's sarcoma [KS]-associated herpesvirus) and have an important role in the development of KS. We have previously shown that HHV-8

The classification of histiocytic proliferations in the skin is confusing, largely due to faulty use of terms such as reticulum-cell sarcoma and reticulosis. Immunohistochemical staining procedures and ultrastructural studies have made it possible to differentiate the different cells of the mononuclear phagocyte system. The proliferations discussed here were those of the mononuclear phagocyte system that are associated with characteristic skin manifestations. They include proliferations of monocytes (acute monocytic leukemia), proliferations of tissue macrophages or histiocytes (histiocytic sa

An adult woman was followed up for the occurrence of multiple successive cutaneous and mucosal tumors of small size and classical histiocytosis X structure (histiocytoeosinophilic granuloma). In the second phase of the patient's clinical history there were successively appearing nodular or infiltrative cutaneous and subcutaneous tumors histologically resembling polymorphocellular sarcoma close either to malignant lymphogranuloma or to recently described "regressing atypical histiocytosis". Despite the marked cellular atypia and polymorphism the tumors exhibited a relatively favorable clinical

BACKGROUND: Secondary cancer is the most life-threatening late effect of childhood cancer. We investigated the clinical features of secondary bone/soft tissue sarcoma among childhood cancer survivors (CCSs).
METHODS: We conducted a retrospective case-series study of 10 069 CCSs newly diagnosed with cancer between 1980 and 2009 across 15 Japanese hospitals. Twenty-one cases of pathologically diagnosed secondary bone/soft tissue sarcoma were selected, and the respective clinical courses were determined using additional questionnaires.
RESULTS: The primary cancers included retinoblastoma (n = 7

In this study, we performed a comprehensive molecular analysis of paired skin and peripheral blood/bone marrow (BM) samples from 17 patients with cutaneous myeloid or cutaneous histiocytic-dendritic neoplasms. The cutaneous manifestations included 10 patients with cutaneous acute myeloid leukemia (c-AML), 2 patients with full or partial Langerhans cell differentiation, 2 patients with blastic plasmacytoid dendritic cell neoplasms (BPDCN), 1 patient with both Langerhans cell differentiation and BPDCN, and 2 patients with full or partial indeterminate dendritic cell differentiation. Seven of the

18F-fluorodeoxyglucose positron emission tomography-computed tomography (18F-FDG PET/CT) is an important but unevenly useful tool in pediatric oncology. Its clearest role is in lymphoma, especially Hodgkin lymphoma, where metabolic imaging informs staging and response-adapted therapy. In sarcoma, neuroblastoma, CNS tumors, Langerhans cell histiocytosis, germ cell tumors, and Wilms tumor, FDG PET is best used selectively as an adjunct to anatomic imaging, disease-specific tracers, and pathology. This review summarizes practical indications, limitations, pediatric protocol issues, and case-image

Histiocytic and dendritic cell neoplasms, especially histiocytic sarcoma, can show morphologic and phenotypic overlap with immature monocytic neoplasms. IRF8 immunohistochemical staining has been demonstrated to be useful in identifying monoblasts, but it has not been extensively studied in histiocytic and dendritic cell neoplasms. IRF8 immunohistochemistry was performed on cases of histiocytic sarcoma (HS, n=6), Langerhans cell histiocytosis (LCH, n=25), Rosai Dorfman disease (RDD, n=17), follicular dendritic cell sarcoma (FDCS, n=3), and Erdheim Chester disease (ECD, n=5), along with a contr

The aim was to evaluate the clinical and histopathological characteristics of hepatic Langerhans cell histiocytosis (LCH). Data from 10 patients with hepatic LCH were collected. The clinicopathological features and immunophenotypes of hepatic LCH were assessed. The study included 6 males and 4 females (median age: 20.5 years). Clinical manifestations and imaging findings were non-specific. Six cases had multisystem involvement, while 4 had isolated hepatic LCH. All lesions were located in the portal area. Histologically, Langerhans cells (LCs) with characteristic nuclear grooves infiltrated th
