Lymphocyte activation gene 3 in autoimmune disease: from molecular mechanisms to clinical applications.

Lymphocyte activation gene 3 (LAG3) has become an important immune checkpoint receptor, which plays a crucial role in regulating the immune response. The immunoregulatory function of LAG3 is achieved through complex receptor-ligand interactions, structure-function relationships, and unique signaling pathways. The role of LAG3 in cancers has been widely explored, and drugs targeting LAG3 are constantly emerging, demonstrating promising application prospects. In autoimmune diseases, downregulation of LAG3 leads to the breakdown of immune tolerance and excessive immune activation. In this review,…

Aerobic Exercise Affected Lymphocyte Apoptosis by Modulating ROS Release and NLRP3 Inflammasome Activation.

Moderate sports training help to enhance spleen-mediated immune function, while prolonged and high-intensity physical exercises could produce an opposite effect. The specific mechanisms underlying the effects of physical load on apoptosis of splenic lymphocytes remain unclear. To address this gap, we developed a model of physical exercises in rats in order to explore the effect of intensity of these exercises on splenic lymphocyte apoptosis and its potential mechanism. We found that high-intensity exercises reduced the CD4+/CD8+ ratio, increased oxidative stress and mitochondrial membrane pote

Paroxetine alleviates T lymphocyte activation and infiltration to joints of collagen-induced arthritis.

T cell infiltration to synovial tissue is an early pathogenic mechanism of rheumatoid arthritis (RA). In the present work, we reveal that G protein coupled receptor kinase 2 (GRK2) is abundantly expressed in T cells of collagen-induced arthritis (CIA). A GRK2 inhibitor, paroxetine protects the joints from inflammation and destruction, primarily through inhibition of both CD4+ helper T (Th) cell and CD8+ cytotoxic T (Tc) cell migration to synovial tissue. Meanwhile, paroxetine restores the balance of Th/Tc, effector Th (Theff)/ naïve Th (Thnaive) and effector Tc (Tceff)/ naïve Tc (Tcnaive) to e

Effects of Yersinia enterocolitica O:3 derivatives on B lymphocyte activation in vivo.

The potential sequelae of intestinal infection with Yersinia enterocolitica include reactive arthritis, erythema nodosum, Reiter's syndrome and other autoimmune diseases. The role of the immune response in the pathogenesis of these diseases has not been fully defined, but autoimmune manifestations may be a consequence of the increase in autoantibodies as a result of polyclonal B-cell activation induced by Yersinia. We investigated the effects of Y. enterocolitica O:3 derivatives on B lymphocyte activation in vivo. Groups of five specific pathogen free (SPF) Swiss mice were inoculated with bact…

Regulation of telomerase RNA template expression in human T lymphocyte development and activation.

Telomeres are unique DNA-protein complexes at the terminals of chromosomes that appear to play a critical role in protecting chromosomal integrity and in maintaining cellular replicative potential. Telomerase is a ribonuclear protein that is capable of elongating telomeres by the addition of telomeric hexanucleotide repeats and therefore contributing to the capacity for cell replication. Telomerase activity is expressed in human germline cells and malignant cells, and it has recently been demonstrated that telomerase activity is highly regulated in normal lymphocytes at specific stages of deve

The pattern of activation antigen expression on T-lymphocyte subpopulation in infectious mononucleosis.

The peripheral blood mononuclear cells of patients with infectious mononucleosis (IM) have been characterized by the determination of activation antigens using a panel of 11 monoclonal antibodies (MoAbs) belonging to 9 clusters. The activation of CD8+ peripheral blood mononuclear cells of IM patients was found to be determined by HLA-DR and CD45RO MoAbs. The antibodies of the CD30, CD40 and CD70 termed antigens also showed an increased expression as compared to the controls. In contrast, the CD25, CD69, CD71, and CD45 antigens were expressed at a low rate on the surface of the CD8+ cells. We f

Analysis of human T lymphocyte activation in a T cell tumor model system.

The human T leukemia line Jurkat maintains functional characteristics of normal T cells in responding to inducing stimuli by the release of interleukin 2 (IL 2). Presence of a phorbol ester during stimulation eliminated the requirement for specialized accessory cells in the response to cell mitogenic agents such as the lectin concanavalin A or treatment with neuraminidase and galactose oxidase. Antibodies directed against the T cell receptor-associated antigen T3 served as efficient stimuli, especially if aided by agents that cross-link immunoglobulin, indicating that a triggering signal is re

Molecular and immune correlates of lymphocyte activation gene-3 expression in renal cell carcinoma.

BACKGROUND: Lymphocyte activation gene-3 (LAG-3) is an immune checkpoint receptor that has emerged as a biomarker of interest but its relationship with outcomes in renal cell carcinoma (RCC) is poorly characterized. This study evaluates molecular and immune correlates of LAG-3 expression and its association with outcomes. METHODS: De-identified DNA-NGS data (xT) and RNA-NGS (xR) from patients (pts) with RCC (n = 566) within the Tempus multi-modal database were analyzed using Lens. Eligible pts had first-line (1 L) immunotherapy (IO) and biopsies collected within 1 year of IO. Samples were str…

Super-resolution imaging of T lymphocyte activation reveals chromatin decondensation and disrupted nuclear envelope.

T lymphocyte activation plays a pivotal role in adaptive immune response and alters the spatial organization of nuclear architecture that subsequently impacts transcription activities. Here, using stochastic optical reconstruction microscopy (STORM), we observe dramatic de-condensation of chromatin and the disruption of nuclear envelope at a nanoscale resolution upon T lymphocyte activation. Super-resolution imaging reveals that such alterations in nuclear architecture are accompanied by the release of nuclear DNA into the cytoplasm, correlating with the degree of chromatin decompaction within…

Lymphocyte Activation in the Development of Immune Tolerance in Women with Recurrent Pregnancy Loss.

Association between lymphocyte activation and formation of immune tolerance, as well as pregnancy outcome, in the case of immunocytotherapy (ICT) was studied in women with idiopathic recurrent pregnancy loss (IRPL). The content and phenotypic characteristics of activated T lymphocytes and NK cells were investigated in the peripheral blood of IRPL patients with different pregnancy outcomes (pregnancy prolongation to the full term and habitual miscarriage). The fraction of activated cells in the subpopulation of cytotoxic T lymphocytes (CD3+CD8+/CD3+CD8+CD69+) before ICT was significantly lower …

T Lymphocyte Activation Markers as Predictors of Responsiveness to Rituximab among Patients with FSGS.

BACKGROUND AND OBJECTIVES: Rituximab is used with variable success in difficult FSGS. Because B cell depletion significantly affects T cell function, we characterized T cell subsets in patients with FSGS to determine if an immunologic signature predictive of favorable response to rituximab could be identified. DESIGN, SETTING, PARTICIPANTS, & MEASUREMENTS: Twenty-two consecutive patients with FSGS (median age =14.4 years old; range =6.2-25.0 years old) and age of onset of nephrotic syndrome 1-18 years old receiving rituximab for clinical indications between October of 2009 and February of 201

Chemokine triggered integrin activation and actin remodeling events guiding lymphocyte migration across vascular barriers.

Chemokine signals activate leukocyte integrins and actin remodeling machineries critical for leukocyte adhesion and motility across vascular barriers. The arrest of leukocytes at target blood vessel sites depends on rapid conformational activation of their α4 and β2 integrins by the binding of endothelial-displayed chemokines to leukocyte Gi-protein coupled receptors (GPCRs). A universal regulator of this event is the integrin-actin adaptor, talin1. Chemokine-stimulated GPCRs can transmit within fractions of seconds signals via multiple Rho GTPases, which locally raise plasma membrane levels o

Lymphocyte activation in type 1 diabetes mellitus: the increased significance of Kv1.3 potassium channels.

Kv1.3 and IKCa1 potassium channels participate in the maintenance of calcium-influx during lymphocyte activation. Kv1.3 channels have a prominent role in specific T cell subsets, presenting a possible target for selective immunomodulation. We investigated the impact of Kv1.3 and IKCa1 channel inhibitors on calcium-influx characteristics in human T cells in type 1 diabetes mellitus. We isolated lymphocytes from 9 healthy and 9 type 1 diabetic individuals and measured the alteration of calcium-influx with flow cytometry in the Th1, Th2, CD4 and CD8 subsets after treatment of samples with specifi…

Assessment of lymphocyte proliferation: CFSE kills dividing cells and modulates expression of activation markers.

The measurement of cell proliferation after mitogenic stimulation is an important parameter used in diagnosis of immunodeficiencies in clinical laboratory as well as in various fields of lymphocyte research. Recent methods try to overcome the radioactive assay using tritiated thymidine ((3)H) by flow-cytometric methods using different fluorochromes such as CFSE or even to substitute these direct methods by tracing the expression of cell-membrane activation markers associated with various steps of proliferation cycle. In our study we compared the (3)H assay with CFSE-staining method and express

The relationship between HA-1 compatibility and the activation of helper T lymphocyte precursors.

The relationship between the compatibility in minor histocompatibility HA-1 antigen and the activation of helper (IL-2 producing) T lymphocyte precursors in vitro was studied in the group of 17 HLA-A2 positive HLA identical siblings. Although the number of pairs studied is still small, no correlation has been found between HA-1 compatibility and helper T lymphocyte precursors activation. The results presented here could suggest the possibility that the HTLp assay does not have to be a relevant parameter for the detection of HA-1 mismatches in HLA identical siblings.

CD8 is critically involved in lymphocyte activation by a T. brucei brucei-released molecule.

T. brucei brucei released a lymphocyte triggering factor (TLTF), which triggered purified CD8+, but not CD4+, T cells to interferon gamma (IFN-gamma) mRNA expression and secretion and to [3H]thymidine incorporation. TLTF also induced mRNA for transforming growth factor beta, but not for interleukin-4. The action of this TLTF on mononuclear cell (MNC) cultures was blocked by anti-CD8 antibodies and by soluble CD8. MNCs from a mutant mouse strain lacking CD8 expression were not triggered by TLTF. IFN-gamma provides a growth stimulus for T. brucei brucei, and infected CD8- mice had much lower par

T lymphocyte activation state in the minor salivary glands of patients with Sjögren's syndrome.

Local lymphoplasmacytoid infiltration of the diseased exocrine glands is a cardinal sign of Sjögren's syndrome (SS). The state of T lymphocyte activation present in these local infiltrations was studied by three different techniques: determination of interleukin 2 (IL2) receptor (Tac) on cell surface membrane; autoradiography combined with immunoperoxidase staining of T cell epitopes; and electron microscopic analysis of the lymphoblast subclasses. Although 64 (SEM 4)% of the local inflammatory cells expressed Ia antigen, only 4 (SEM 1)% of them displayed the T cell activation antigen Tac. Aut…
