cJun overexpression in MCF-7 breast cancer cells produces a tumorigenic, invasive and hormone resistant phenotype.

We have previously demonstrated decreased Jun/AP-1 activity in the breast cancer cell line MCF-7 when compared to normal or immortalized mammary epithelial cells. In this paper, we overexpress Jun in MCF-7 cells (MCF7Jun) and demonstrate that it results in diverse biologic and biochemical changes, which mimic those seen clinically in breast cancer. Overexpression of Jun causes significant alterations in the composition of AP-1, decreased junB and increased fra-1 expression and results in an increased biologic aggressiveness. MCF7Jun cells exhibit increased cellular motility, increased expressi…

Coexistence of novel amylin-binding sites with calcitonin receptors in human breast carcinoma MCF-7 cells.

Amylin, calcitonin (CT) and calcitonin gene-related peptide (CGRP) share limited structural homology including amino-terminal ring structures linked by a disulfide bridge and amidated carboxy-termini. Here, we have compared [125I]Bolton-Hunter-[Lys1] rat amylin ([125I]amylin) binding and the stimulation of cyclic AMP accumulation by human (h) amylin, hCT and hCGRP-I in the human breast carcinoma cell lines MCF-7 and T47D, which predominantly express hCT1a and hCT1b receptor isoforms (hCTR1a, hCTR1b) at a similar total number of hCT-binding sites. In MCF-7 cells, half-maximal inhibition (IC50) …

Induction and inhibition of estradiol hydroxylase activities in MCF-7 human breast cancer cells in culture.

The effects of estradiol, progesterone, and tamoxifen on the activity of estradiol 2- and 16 alpha-hydroxylases were studied in human breast cancer cell cultures using a radiometric assay. After 5 days' exposure to these compounds, incubations in the presence of either [2-3H]estradiol or [16 alpha-3H]estradiol as substrate were carried out. In MCF-7 cells, estradiol (10(-8) M), progesterone (10(-6) M) and tamoxifen (10(-6) M) significantly increased 16 alpha-hydroxylase activity (estradiol; 21% progesterone 10% to 32%; tamoxifen 21% to 31%; P less than 0.01). Synergistic effects were observed …

Antimicrobials and in vitro systems: antibiotics and antimycotics alter the proteome of MCF-7 cells in culture.

Cell culture is widely used to study gene or protein changes in response to experimental conditions. The value of such experiments depends on stringent control and understanding of the in vitro environment. Despite well-documented evidence describing toxic effects in the clinical setting, antibiotics and antimycotics are routinely used in cell culture without regard for their potential toxicity. We cultured MCF-7 breast cancer cells in the presence/absence of antibiotics (penicillin/streptomycin) and/or the antimycotic amphotericin B. Differential protein expression was assessed using 2D-DIGE

Cloning of a gene expressed in human breast cancer and regulated by estrogen in MCF-7 cells.

Messenger RNAs (mRNAs) were prepared from MCF-7 breast cancer cells grown in the presence of estradiol. Complementary DNAs (cDNAs) were inserted into pBR322 plasmid and a library of 4400 recombinant bacterial clones was prepared. The clones were screened by in situ differential hybridization with cDNAs prepared from RNAs of MCF-7 cells grown either in the presence or absence of estradiol. Several estrogen-induced or estrogen-repressed clones were identified. One of them corresponded to a relatively frequent mRNA (0.8% of recombinant plasmids) of 650 nucleotides. The concentration of this mRNA …

Estimation of estrogenic and antiestrogenic activities of selected pesticides by MCF-7 cell proliferation assay.

Estrogenic activities of 20 selected pesticides-which are used for agricultural production as insecticides, fungicides and herbicides-were examined by estrogen receptor (ER)-dependent MCF-7 cell proliferation assay. Among them, chlordecone, dicofol, methoxychlor, gamma-HCH, fenarimol, EPN, triadimefon, and triadimenol had estrogenic activities, all of which were suppressed by the addition of pure antiestrogen ICI 182,780. The first 5 compounds exhibited binding capacities to ERalpha. The antiestrogenic activity of a compound was examined by estimating its suppressive effect on cell proliferati

Reduced expression of the BRCA1 gene and increased chromosomal instability in MCF-7 cell line.

Using clonal cell cultures, a significant increase in chromosomal aberrations (aneuplolidy, dicentrics and chromatid breaks) were observed in MCF-7 cells compared with HeLa. BRCA1 expression was lower in MCF-7 cells than in HeLa cells. Since BRCA1 is known to play a role in the maintenance of chromosomal integrity, the increase in chromosomal aberrations in MCF-7 clones suggests that downregulation of BRCA1 expression could be one of the possible mechanisms for increased chromosomal instability in this cell line.

Effects of Ferrocene and Ferrocenium on MCF-7 Breast Cancer Cells and Interconnection with Regulated Cell Death Pathways.

The effects of ferrocene (Fc) and ferrocenium (Fc+) induced in triple negative human breast cancer MCF-7 cells were explored by immunofluorescence, flow cytometry, and transmission electron microscopy analysis. The different abilities of Fc and Fc+ to produce reactive oxygen species and induce oxidative stress were clearly observed by activating apoptosis and morphological changes after treatment, but also after tests performed on the model organism D. discoideum, particularly in the case of Fc+. The induction of ferroptosis, an iron-dependent form of regulated cell death driven by an overload…

Effect of specific silencing of EMMPRIN on the growth and cell cycle distribution of MCF-7 breast cancer cells.

The extracellular matrix metalloproteinase inducer (EMMPRIN, CD147) is a member of the immunoglobulin family and shows increased expression in tumor cells. We examined the effect of RNAi-mediated EMMPRIN gene silencing induced by lentiviral on the growth and cycle distribution of MCF-7 breast cancer cells. Lentiviral expressing EMMPRIN-short hairpin RNA were packaged to infect MCF-7 cells. The inhibition efficiency of EMMPRIN was validated by real-time fluorescent quantitation polymerase chain reaction and western blotting. The effect of EMMPRIN on cell proliferation ability was detected using…

Potential-resolved single-nanoparticle collision electrochemistry for simultaneous quantification of MCF-7 and SK-BR-3 cells.

Compared to detecting individual target analytes by single immunoassay, multiplex immunoassays (MIAs) can simultaneously detect multiple analytes in a single experiment, saving more time and reducing sample consumption. Single-nanoparticle collision electrochemistry (SNCE), characterized by its high throughput, sensitivity, and rapid response, has been extensively employed in the field of biosensing. Herein, an innovative potential-resolved SNCE biosensor was proposed for the simultaneous and rapid detection of two types of breast cancer cells (MCF-7 and SK-BR-3 cells) based on platinum nanopa

Integrated data-independent acquisition and thermal proteome profiling for proteomic characterization of lamotrigine-treated MCF-7 cells.

Lamotrigine, a clinically approved anticonvulsant, has recently gained interest for its potential anticancer effects. However, the molecular mechanisms underlying its activity in breast cancer remain poorly understood. Here, an integrated mass spectrometry-based proteomic strategy combining data-independent acquisition (DIA) and thermal proteome profiling (TPP) was applied to investigate the cellular effects and potential targets of lamotrigine in the human breast cancer cell line MCF-7. The DIA-based quantitative approach identified 6622 proteins, of which 142 were significantly modulated fol

An acidic pH-activatable fluorescent probe for MUC1 detection and the precise imaging of MCF-7 cells.

Mucin 1 (MUC1), a pivotal cancer biomarker, is overexpressed on MCF-7 cells, which has stimulated the construction of numerous aptamer-based fluorescent probes for breast cancer diagnosis. However, MUC1 is also present in the serum of cancer patients and expressed at low levels on the surface of normal cells, which may result in false-positive signals upon recognition by aptamers. In this work, a gold nanoparticle (AuNPs)-based fluorescent probe termed b-NPs was proposed by utilizing the acidic tumor microenvironment and pH-responsive single-stranded DNA (f-DNA) with conformational switching c…

NSUN7 Suppression Reduces Metastatic Potential and Restores Sensitivity to 4-OH Tamoxifen in Resistant MCF-7 Cells.

BACKGROUND/AIM: Tamoxifen remains a first-line treatment for estrogen receptor-positive breast cancer. Emerging evidence indicates that aberrant 5-methylcytosine (m5C) modification of RNAs contributes to chemotherapeutic resistance in various types of cancer, however their role in determining tamoxifen resistance in breast cancer remains elusive. MATERIALS AND METHODS: We measured global m5C RNA methylation and expression of its regulating enzymes in tamoxifen-sensitive (MCF-7) versus tamoxifen-resistant cells (MCF-7 Tam1). Expression of the most significantly dysregulated enzyme NOP2/Sun RNA

Propofol suppresses migration and vascular mimicry of breast cancer MCF-7 cells by down regulating interleukin-6.

Propofol is widely used in anesthesia, but its role in breast cancer progression remains controversial. This study investigated the molecular mechanisms of propofol in breast cancer, focusing on IL-6 and tumor microenvironment modulation. Bioinformatics analysis identified IL-6 as a potential target of propofol. MCF-7 cells were treated with varying propofol concentrations (0-100 μg/mL), and cell viability was assessed via CCK-8 assay. Propofol at 50 μg/mL significantly reduced viability, while 25 μg/mL (a non-cytotoxic dose) was selected for further experiments. Western blot confirmed propofo…
