Effects of alverine citrate on cat intestinal mechanoreceptor responses to chemical and mechanical stimuli.

BACKGROUND: Alverine citrate is commonly used in the treatment of painful affections of the colon. AIM: To determine whether alverine citrate acts on the vagal sensory endings. METHODS: Unitary recordings were performed at the level of the vagal fibres in the nodose ganglion of anaesthetized cats using extracellular glass microelectrodes, and the patterns of response to chemical and mechanical stimuli applied to identified vagal intestinal mechanoreceptors were studied. RESULTS: The intestinal mechanoreceptors located at the endings of type C vagal fibres responded mainly to mechanical stim…

Neural response of mechanoreceptors to acute inflammation in the rotator cuff of the shoulder joint in rabbits.

We examined with electrophysiological techniques the effects of experimentally induced inflammation on the mechanosensitive afferent units in the rotator cuff of the shoulder joint of 21 rabbits. We identified 21 mechanosensitive units belonging to group III. 12 units had mechanical thresholds of > 7.0 g and 9 units had thresholds of < 7.0 g. After injection of inflammatory agents, kaolin and carrageenan, into the joint space, ongoing afferent discharge rates increased in all units. The average discharge rate increased significantly from 7 imp/s to 15 imp/s after injection. 5 units had a decre

Vibration sensitivity of slowly and rapidly adapting cutaneous mechanoreceptors in the human foot and leg.

The activities of 30 rapidly adapting cutaneous receptors (FA) and 23 slowly adapting cutaneous receptors (SA) were recorded from the lateral peroneal nerve using the microneurographic method. Their sensitivity to mechanical vibrations with constant amplitude applied at various frequencies to the center of the receptive field was studied. These two populations of cutaneous receptors were found to be very sensitive to this stimulus: they could be driven in a one to one manner up to between 100 and 200 Hz. The difference lay in the response observed when the vibration frequency was increased to

Children with cerebral palsy have uncharacteristic somatosensory cortical oscillations after stimulation of the hand mechanoreceptors.

Numerous clinical investigations have reported that children with cerebral palsy (CP) have tactile discrimination deficits that likely limit their ability to plan and manipulate objects. Despite this clinical awareness, we still have a substantial knowledge gap in our understanding of the neurological basis for these tactile discrimination deficits. Previously, we have shown that children with CP have aberrant theta-alpha (4-14 Hz) oscillations in the somatosensory cortices following tactile stimulation of the foot. In this investigation, we evaluated if these aberrant theta-alpha oscillations

Organization of vagal afferents in pylorus: mechanoreceptors arrayed for high sensitivity and fine spatial resolution?

The pylorus is innervated by vagal mechanoreceptors that project to gastrointestinal smooth muscle, but the distributions and specializations of vagal endings in the sphincter have not been fully characterized. To evaluate their organization, the neural tracer dextran biotin was injected into the nodose ganglia of rats. Following tracer transport, animals were perfused, and their pylori and antra were prepared as whole mounts. Specimens were processed to permanently label the tracer, and subsets were counterstained with Cuprolinic blue or immunostained for c-Kit. Intramuscular arrays (IMAs) in…

Identification of functional intramuscular rectal mechanoreceptors in aganglionic rectal smooth muscle from piebald lethal mice.

The mechanosensitive endings of low-threshold, slowly adapting pelvic afferents that innervate the rectum have been previously identified as … ential stretch applied to the rectum activated graded increases in firing of slowly adapting rectal mechanoreceptors. In s(l)/s(l) mice, graded stretch of the aganglionic rectum activated similar graded increases in rectal afferent firing. Stretch-sensitive afferents responded at low mechanical thresholds and fired more intensely at noxious levels of stretch. They …

Responsiveness and ultrastructure of slowly adapting type I cutaneous mechanoreceptors in vitamin A deficient rats.

Single-unit recordings were made from afferent nerve fibres supplying slowly adapting type I (s.a. I) cutaneous mechanoreceptors in anaesthetized vitamin A deficient and control rats. Trains of thirty repetitive mechanical stimuli with 0.1 s rise time, 1.9 s plateau phase, and 0.7 s interstimulus interval were applied. A feed-back mechanism maintained the force of stimulation at 20 mN during the plateau phases and the contact force between stimuli at 0.5 mN. All displacement values in the group of vitamin A deficient rats were significantly larger than the corresponding control values. Residua…

Synchrony within Aβ mechanoreceptor subtypes governs signal propagation to primary somatosensory cortex.

Fast-conducting Aβ slowly adapting type I low threshold mechanoreceptors (SAI-LTMRs) and Aβ rapidly adapting type I (RAI-)LTMRs are critical for discriminative touch from glabrous skin. Recent studies used mouse genetic loss-of-function and optogenetic gain-of-function manipulations to determine that signals from these Aβ LTMRs are integrated subcortically to build the cortical representation of touch. However, the precise influence of each subtype on downstream responses is unclear, in part because previous manipulations lacked the capacity to reproduce physiological firing rates and patterns…

Anatomical investigation of cutaneous plantar mechanoreceptor pathways and their connections to hypothalamic structures in laboratory rats.

BACKGROUND: Cutaneous mechanoreceptors detect physical stimuli. Their pathways to the CNS are not fully mapped. Understanding these connections helps explain the sensory regulation of endocrine function. AIM: This study mapped the anatomical connections between plantar skin mechanoreceptors and hypothalamic nuclei. The structural, cellular, and neurochemical features were examined. METHODS: A total of 24 male Wistar rats were used. The plantar skin was stimulated with calibrated vibration. Tissue samples were collected from the skin, spinal cord, and hypothalamus. We used Fast Blue and BDA a…

Differential contributions of cardiac, coronary and pulmonary artery vagal mechanoreceptors to reflex control of the circulation.

Distinct populations of stretch-sensitive mechanoreceptors attached to myelinated vagal afferents are found in the heart and adjoining coronary and pulmonary circulations. Receptors at atrio-venous junctions appear to be involved in control of intravascular volume. These atrial receptors influence sympathetic control of the heart and kidney, but contribute little to reflex control of systemic vascular resistance. Baroreceptors at the origins of the coronary circulation elicit reflex vasodilatation, like feedback control from systemic arterial baroreceptors, as well as having characteristics th…

A morphologic and quantitative comparison of mechanoreceptors in the tibial remnants of the ruptured human anterior cruciate ligament.

Reconstruction of the ruptured anterior cruciate ligament (ACL) does not always result in expected successful outcome. A satisfactory outcome depends not only on the tightness or strength of the graft but also on the quality of proprioceptive restoration. Mechanoreceptors of ACL are supposed to play considerable roles in the proprioceptive feedback system of knee. This study aimed to observe the condition and number of the surviving mechanoreceptors in the tibial remnant of ruptured ACL in human knees. From April 2009 to January 2012, 60 patients with existing free tibial remnants who had under…

Abnormalities of mechanoreceptors in a rat model of neuropathic pain: possible involvement in mediating mechanical allodynia.

1. Peripheral nerve injury sometimes leads to the development of neuropathic pain. One of the symptoms of such neuropathic pain is mechanical allodynia, pain in response to normally innocuous mechanical stimuli. We hypothesized that sympathetically driven dysfunction of cutaneous mechanoreceptors is responsible for signaling mechanical allodynia. The present study was undertaken to identify the types of sensory receptors that potentially mediate mechanical allodynia, with the use of a rat neuropathic pain model we have developed. 2. One week to 10 days after tight ligations of the L5 and L6 sp…

Impact of antral mechanoreceptor activation on the vago-vagal reflex in the rat: functional zonation of responses.

1. Activation of gastric sensory afferents alters gastric motor and secretory function via the gastric vago-vagal reflex. In this report, we investigated in the rat the impact of gastric mechanoreceptor activation on the brain stem components of the reflex, which are located in the dorsal vagal complex (DVC), i.e. the nucleus of the solitary tract (NTS) and the subjacent dorsal motor nucleus (DMN). 2. In our extracellular recordings of single-cell activity in the DVC, we observed a relation between the response to antral distention and the location of the cell in the DVC. Specifically, cells t

Splanchnic slowly adapting mechanoreceptors with punctate receptive fields in the mesentery and gastrointestinal tract of the cat.

1. A class of slowly adapting mechanoreceptors with A-delta and C fibres running in the splanchnic nerves of cats is described.2. The mechanoreceptors have punctate regions of mechanical sensitivity at macroscopic vascular branching points and have been found in the lesser omentum, the mesentery of the gall-bladder, porta hepatis, portal vein, pancreas, spleen and the duodenum, jejunum, ileum, colon and their mesenteries.3. The receptive fields of these mechanoreceptors vary considerably in size in the different regions. The largest receptive fields were found in the small intestinal mesentery…

Low-threshold cutaneous mechanoreceptors on the foot sole have enhanced sensitivity when skin temperature is elevated.

Cutaneous mechanoreceptor input from the foot sole is critical for balance and gait, yet ageing and disease can compromise skin sensitivity, increasing risks for falls and related injuries. Heating improves perceptual sensitivity, but its direct effects on afferent firing remain unknown. This study examined how topical heating alters Aβ afferent responses to mechanical loads that closely emulate walking. Single tibial nerve afferents were recorded using microneurography and classified as fast-adapting (FA) or slowly adapting (SA), type I or II. Ramp-and-hold compressions, normalized to regiona
