Comment on "New insights into the biomineralization of mercury selenide nanoparticles through stable isotope analysis in giant petrel tissues".

Some birds and cetaceans can demethylate the toxic methylmercury cysteinate (MeHgCys) complex into inert mercury sulfide (HgSe) through the formation of an intermediate tetrahedral selenolate complex with selenocysteine (Sec) residues (Hg(Sec)4). The nucleation of the HgSe biominerals involves the substitution of the Se ligand for the Sec residues, which is considered to occur in the form of multinuclear Hgx(Se,Sec)y clusters mediated by proteins. Queipo-Abad et al. (2022) isolated HgSe nanoparticles from the biological tissues of giant petrels and measured the mass-dependent fractionation of

Application of speciated isotope dilution mass spectrometry to evaluate extraction methods for determining mercury speciation in soils and sediments.

Extraction techniques commonly used to extract methylmercury or mercury species from various matrixes have been evaluated regarding their potential to transform inorganic mercury to methylmercury, or vice versa, during sample preparation steps by applying speciated isotope dilution mass spectrometry. Two of the five tested methods were highly prone to form inorganic mercury from methylmercury. Some published methods converted methylmercury to inorganic mercury approximately 100% (including the spiked CH(3)(201)Hg(+)). In other methods, as much as 45% of methylmercury was converted to inorganic

Mercury isotope fractionation during methylmercury transport and transformation: A review focusing on analytical method, fractionation characteristics, and its application.

Methylmercury (MeHg), a potent neurotoxin, can be formed, migrated and transformed in environmental compartments, accompanying with unique mass-dependent and mass-independent fractionation of mercury (Hg). These Hg isotope fractionation signals have great potential to probe the transformation and transport of MeHg in aquatic environments. However, the majority of studies to date have focused on total Hg isotopic composition, with less attention to the isotopic fractionation of MeHg due to technical difficulties in analysis, which severely hinders the understanding of MeHg isotopic fractionatio

Measurement of mercury species in human blood using triple spike isotope dilution with SPME-GC-ICP-DRC-MS.

The measurement of different mercury compounds in human blood can provide valuable information about the type of mercury exposure. To this end, our laboratory developed a biomonitoring method for the quantification of inorganic (iHg), methyl (MeHg), and ethyl (EtHg) mercury in whole blood using a triple-spike isotope dilution (TSID) quantification method employing capillary gas chromatography (GC) and inductively coupled dynamic reaction cell mass spectrometry (ICP-DRC-MS). We used a robotic CombiPAL(®) sample handling station featuring twin fiber-based solid-phase microextraction (SPME) injec

Application of ultrafiltration and stable isotopic amendments to field studies of mercury partitioning to filterable carbon in lake water and overland runoff.

Results from pilot studies on colloidal phase transport of newly deposited mercury in lake water and overland runoff demonstrate that the combination of ultrafiltration, and stable isotope amendment techniques is a viable tool for the study of mercury partitioning to filterable carbon. Ultrafiltration mass balance calculations were generally excellent, averaging 97.3, 96.1 and 99.8% for dissolved organic carbon (DOC), total mercury (Hg(T)), and methylmercury (MeHg), respectively. Sub nanogram per liter quantities of isotope were measurable, and the observed phase distribution from replicate ul

Mercury exposure and isotopic signatures in overwintering Black-headed Gulls (Chroicocephalus ridibundus): Age-related variation and origin tracing.

Black-headed Gulls (Chroicocephalus ridibundus), a long-distance migratory bird species, serve as unique spatial integrators of mercury (Hg) pollution across their various geographical range. This study investigated the Hg exposure, Hg isotopic signatures, and trophic dynamics in the feathers of wintering Black-headed Gulls (70 adults, 28 juveniles) at Dianchi Lake in Southwest China. Results showed that adults had significantly higher concentrations of total Hg (THg: 4.4 ± 2.4 mg/kg) and methylmercury (MeHg: 3.3 ± 1.9 mg/kg) than juveniles (THg: 3.3 ± 1.6 mg/kg; MeHg: 2.3 ± 1.2 mg/kg). Species-specific isotopic analysis of mercury in marine biota: Systematic evaluation of extraction and oxidation methods for a straightforward analytical approach.

Mercury (Hg) is a globally distributed heavy metal with significant environmental and health implications. Along its biogeochemical cycle, Hg can transform into various chemical species, including methylmercury (MeHg), a potent neurotoxin capable of crossing biological barriers, thereby posing serious risks to the human nervous system, especially in fetuses and infants. While determination of the Hg concentration and its speciation analysis already offer valuable insights, isotopic analysis, particularly species-specific isotopic analysis, can reveal detailed information on the environmental p

Probing methylmercury photodegradation by different fractions of natural organic matter in water: Degradation kinetics and mercury isotope fractionation characteristics.

Recent advancements in mercury (Hg) isotopic fractionation research have evolved from conceptual demonstrations to practical applications. However, few studies have focused on revealing fractionation fingerprinting for aqueous methylmercury (MeHg) photodegradation due to its sensitivity to natural organic matter (NOM). Here, the impact of NOM fractions with varying chemical properties on MeHg photodegradation kinetics and Hg isotope fractionation characteristics was investigated. Findings reveal that reduced NOM, containing alcohol/phenol groups, slows the degradation rate compared to the oxid

Mercury Methylation Potentials in Sediments of an Ancient Cypress Wetland Using Species-Specific Isotope Dilution GC-ICP-MS.

Wetlands are of a considerable environmental value as they provide food and habitat for plants and animals. Several important chemical transformations take place in wetland media, including the conversion of inorganic mercury (Hg) to monomethylmercury (MeHg), a toxic compound with a strong tendency for bioconcentration. Considering the fact that wetlands are hotspots for Hg methylation, we investigated, for the first time, Hg methylation and demethylation rates in an old growth cypress wetland at Sky Lake in the Mississippi Delta. The Sky Lake ecosystem undergoes large-scale water level fluctu

High-precision isotopic analysis sheds new light on mercury metabolism in long-finned pilot whales (Globicephala melas).

Whales accumulate mercury (Hg), but do not seem to show immediate evidence of toxic effects. Analysis of different tissues (liver, kidney, muscle) and biofluids (blood, milk) from a pod of stranded long-finned pilot whales (Globicephala melas) showed accumulation of Hg as a function of age, with a significant decrease in the MeHg fraction. Isotopic analysis revealed remarkable differences between juvenile and adult whales. During the first period of life, Hg in the liver became isotopically lighter (δ202Hg decreased) with a strongly decreasing methylmercury (MeHg) fraction. We suggest this is

Simultaneous determination of mercury methylation and demethylation capacities of various sulfate-reducing bacteria using species-specific isotopic tracers.

The use of species-specific isotopic tracers for inorganic and methyl mercury has allowed the simultaneous determination of the methylation and demethylation potentials of pure culture of isolated sulfate-reducing (SR) bacterial strains using low Hg species concentration levels (7 µg/L (199)Hg(II), 1 µg/L Me(201)Hg). A major advantage of the method reported here is that it can be used to follow simultaneously both the degradation of the species added but also the formation of their degradation products and thus the determination during the same incubation of the specific methylation/demethylat

Levels of mercury and organochlorine compounds and stable isotope ratios in three tuna species taken from different regions of Japan.

Levels of mercury (Hg) and organochlorine compounds (OCs), such as PCBs and p,p'-DDE, as well as the stable isotope ratios of carbon (delta 13C) and nitrogen (delta 15N) were compared in Pacific bluefin, yellowfin, and albacore tuna taken from the southern, central, and northern regions of Japan according to tuna species and the region of origin. Levels of Hg and OCs as well as the delta 15N value were the highest in the bluefin tuna, reflecting their higher trophic position and longer life span. The average Hg concentrations tended to be higher in specimens taken in the southern region than i

Mercury, selenium, and stable carbon and nitrogen isotopes in the striped marlin Kajikia audax and blue marlin Makaira nigricans food web from the Gulf of California.

Mercury (Hg) concentrations based on dietary sources have shown to predict differences in fish; however, they are usually applied at an individual scale and are rarely directed at a known trophic transfer. We combined gut content analysis and stable isotope analysis (δ15N and δ13C) to provide a quantitative estimate of Hg and selenium (Se) biomagnification in the striped marlin (Kajikia audax) and blue marlin (Makaira nigricans) food web from the southwestern Gulf of California. Hg and Se concentrations (mean ± SD; μg g-1, dw) were different among K. audax (Hg = 3.6 ± 2.1, Se = 5.5 ± 5.4) and

Compound-specific stable isotopes of amino acids reveal influences of trophic level and primary production sources on mercury concentrations in fishes from the Aleutian Islands, Alaska.

Total mercury concentrations ([THg]) exceed thresholds of concern in some Steller sea lion (Eumetopias jubatus) tissues from certain portions of the Aleutian Islands, Alaska. We applied compound-specific stable isotope analyses of both carbon and nitrogen in amino acids from fish muscle tissue to quantify the proportional contributions of primary production sources and trophic positions of eight prey species (n = 474 total) that are part of Steller sea lion diets. Previous THg analyses of fish muscle, coupled with monomethylmercury analyses of a subset of samples, substantiated previous findin

Mercury bioaccumulation in tilefish from the northeastern Gulf of Mexico 2 years after the Deepwater Horizon oil spill: Insights from Hg, C, N and S stable isotopes.

Mercury (Hg) concentration in fish of the Gulf of the Mexico (GoM) is a major concern due to the importance of the GoM for U.S. fisheries. The Deepwater Horizon (DWH) oil spill in April 2010 in the northern GoM resulted in large amounts of oil and dispersant released to the water column, which potentially modified Hg bioaccumulation patterns in affected areas. We measured Hg species (methylmercury (MMHg) and inorganic Hg (IHg)) concentrations, and light (C, N and S) and Hg stable isotopes in muscle and liver tissues from tilefish (Lopholatilus chamaleonticeps) sampled in 2012 and 2013 along th

Mercury isotope signatures of seawater discharged from a coal-fired power plant equipped with a seawater flue gas desulfurization system.

Seawater flue gas desulfurization (SFGD) systems are commonly used to remove acidic SO2 from the flue gas with alkaline seawater in many coastal coal-fired power plants in China. However, large amount of mercury (Hg) originated from coal is also transferred into seawater during the desulfurization (De-SO2) process. This research investigated Hg isotopes in seawater discharged from a coastal plant equipped with a SFGD system for the first time. Suspended particles of inorganic minerals, carbon residuals and sulfides are enriched in heavy Hg isotopes during the De-SO2 process. δ(202)Hg of partic

On-line species-unspecific isotope dilution analysis in the picomolar range reveals the time- and species-depending mercury uptake in human astrocytes.

In order to reveal the time-depending mercury species uptake by human astrocytes, a novel approach for total mercury analysis is presented, which uses an accelerated sample introduction system combined on-line with an inductively coupled plasma mass spectrometer equipped with a collision/reaction cell. Human astrocyte samples were incubated with inorganic mercury (HgCl2), methylmercury chloride (MeHgCl), and thimerosal. After 1-h incubation with Hg(2+), cellular concentrations of 3 μM were obtained, whereas for organic species, concentrations of 14-18 μM could be found. After 24 h, a cellular

In situ experiments for element species-specific environmental reactivity of tin and mercury compounds using isotopic tracers and multiple linear regression.

The fate of mercury (Hg) and tin (Sn) compounds in ecosystems is strongly determined by their alkylation/dealkylation pathways. However, the experimental determination of those transformations is still not straightforward and methodologies need to be refined. The purpose of this work is the development of a comprehensive and adaptable tool for an accurate experimental assessment of specific formation/degradation yields and half-lives of elemental species in different aquatic environments. The methodology combines field incubations of coastal waters and surface sediments with the addition of sp

Simultaneous speciation of mercury and butyltin compounds in natural waters and snow by propylation and species-specific isotope dilution mass spectrometry analysis.

A robust method has been developed for simultaneous determination of mercury and butyltin compounds in aqueous samples. This method is capable of providing accurate results for analyte concentrations in the picogram per liter to nanogram per liter range. The simultaneous determination of the mercury and tin compounds is achieved by species-specific isotope dilution, derivatization, and gas chromatography-inductively coupled plasma mass spectrometer (GC-ICP-MS). In derivatization by ethylation and propylation, reaction conditions such as pH and the effect of chloride were carefully studied. Eth
