Localization of enterobacterial common antigen: Proteus mirabilis and its various L-forms.

An investigation of Proteus mirabilis wild-type strains and their various derived L-forms shows that the enterobacterial common antigen (ECA) is localized in the outer membrane of the cell envelope of these strains. In strains where the outer membrane is lacking (stable protoplast L-forms) or where its amount is reduced (spheroplast UL19) no ECA or only reduced amounts of it are detected by serological tests or by ferritin-labeling techniques.

Antibody reactivity to bacterial lipopolysaccharide in rheumatoid arthritis: insights from Proteus mirabilis O3.

Lipopolysaccharides and their O-polysaccharide are structurally diverse bacterial carbohydrates polymers that strongly influence immune recognition. Here, we investigated human affinity-purified IgG antibodies recognizing the structurally defined lysine containing Proteus mirabilis O3 LPS, focusing on the contribution of glycan-amino acid motifs to antibody binding specificity. Antibodies from rheumatoid arthritis patients and healthy donors displayed comparable affinity and avidity toward Proteus mirabilis O3 LPS, although significantly higher amounts of LPS-binding IgG were isolated from pat…

Phylogenomic structure and resistance profiles of clinical Proteus mirabilis from a Thai hospital.

Proteus mirabilis is an opportunistic pathogen associated with urinary tract infections, wound infections, and bacteremia. We analyzed 17 clinical isolates collected from hospitals in Narathiwat Province, southern Thailand, to characterize their antimicrobial resistance (AMR), virulence potential, plasmid replicon content, and genomic relatedness. Most isolates were phenotypically susceptible to all tested antibiotics. Resistance was limited to two strains: KS160 (ceftriaxone, ciprofloxacin, levofloxacin) and KS044 (ciprofloxacin, levofloxacin). Whole-genome analysis, however, identified 50 un…

Distribution of antimicrobial resistance and virulence markers in chicken originated Proteus mirabilis isolates.

Proteus mirabilis is a common enteric bacterium in livestock and humans. The increase and spread of the antimicrobial resistant P. mirabilis is considered alarming worldwide. Transmission mainly occurs through consumption of contaminated poultry products. We investigated antimicrobial resistance (AMR) and virulence markers in broiler chicken-originated P. mirabilis isolates from 380 fecal samples. Phenotypic AMR test was performed against seventeen different antimicrobials. Genotypic AMR test was performed to detect sixteen different AMR genes. The samples were also tested for the presence of …

Computational study to investigate Proteus mirabilis proteomes for multi-epitope vaccine construct design.

Proteus mirabilis is a gram-negative bacterium particularly known for its unique swarming ability. The swarming gives the bacteria ability to enhance adherence to the catheter surface and epithelium cells of the urethra to cause catheter associated urinary tract infections. P. mirabilis has evolved resistant to antibiotics. Additionally, there is an approved vaccine against P. mirabilis, thus demanding for identification of new vaccine targets. This gram-negative bacterium consists of 19,502 core proteins, out of which 19,063 are redundant proteins and remaining 439 are non-redundant proteins.…

Amikacin and bacteriophage treatment modulates outer membrane proteins composition in Proteus mirabilis biofilm.

Modification of outer membrane proteins (OMPs) is the first line of Gram-negative bacteria defence against antimicrobials. Here we point to Proteus mirabilis OMPs and their role in antibiotic and phage resistance. Protein profiles of amikacin (AMKrsv), phage (Brsv) and amikacin/phage (AMK/Brsv) resistant variants of P. mirabilis were compared to that obtained for a wild strain. In resistant variants there were identified 14, 1, 5 overexpressed and 13, 5, 1 downregulated proteins for AMKrsv, Brsv and AMK/Brsv, respectively. Application of phages with amikacin led to reducing the number of up- a…

Proteus mirabilis Targets Atherosclerosis Plaques in Human Coronary Arteries via DC-SIGN (CD209).

Bacterial DNAs are constantly detected in atherosclerotic plaques (APs), suggesting that a combination of chronic infection and inflammation may have roles in AP formation. A series of studies suggested that certain Gram-negative bacteria were able to interact with dendritic cell-specific intercellular adhesion molecule-3-grabbing non-integrin [DC-SIGN; cluster of differentiation (CD) 209] or langerin (CD207), thereby resulting in deposition of CD209s at infection sites. We wondered if Proteus mirabilis (a member of Proteobacteria family) could interact with APs through CD209/CD207. In this st…

Clinical significance of Proteus mirabilis bacteriuria in dogs, risk factors and antimicrobial susceptibility.

The objectives of this study were to describe the in vitro antimicrobial susceptibility and clinical significance of Proteus mirabilis in canine bacteriuria and to identify the risk factors associated with P. mirabilis urinary tract infections. This is a retrospective observational study of 48 P. mirabilis-positive canine urinary cultures. Only 22 of the 48 P. mirabilis isolates (45.8%) were non-susceptible to at least one tested antimicrobial. Most P. mirabilis isolates (98%) were susceptible to enrofloxacin, 93.7% to amoxicillin/clavulanic acid, and 85.4% to ampicillin, cephalothin, and trim…

Oral administration of Proteus mirabilis damages dopaminergic neurons and motor functions in mice.

Recently, studies on the relationship between gut dysbiosis and Parkinson's disease (PD) have increased, but whether a specific gut bacterium may cause PD remains unexplored. Here, we report, for the first time, that a specific gut bacterium directly induces PD symptoms and dopaminergic neuronal damage in the mouse brain. We found that the number of Enterobacteriaceae, particularly Proteus mirabilis, markedly and commonly increased in PD mouse models. Administration of P. mirabilis isolated from PD mice significantly induced motor deficits, selectively caused dopaminergic neuronal damage and i…

Regional outbreak of CTX-M-2 β-lactamase-producing Proteus mirabilis in Japan.

Proteus mirabilis is a common cause of urinary tract infection. Wild-type P. mirabilis strains are usually susceptible to penicillins and cephalosporins, but occurrences of P. mirabilis producing extended-spectrum β-lactamases (ESBLs) have been recently reported. Here, we surveyed the prevalence of cefotaxime resistance among P. mirabilis strains at seven different hospitals in Kanagawa Prefecture, Japan, and investigated their molecular epidemiology to explain the mechanism of their spread. The prevalence of cefotaxime resistance among P. mirabilis increased annually, from 10.1 % in 1998 to 2…

Complicated catheter-associated urinary tract infections due to Escherichia coli and Proteus mirabilis.

Catheter-associated urinary tract infections (CAUTIs) represent the most common type of nosocomial infection and are a major health concern due to the complications and frequent recurrence. These infections are often caused by Escherichia coli and Proteus mirabilis. Gram-negative bacterial species that cause CAUTIs express a number of virulence factors associated with adhesion, motility, biofilm formation, immunoavoidance, and nutrient acquisition as well as factors that cause damage to the host. These infections can be reduced by limiting catheter usage and ensuring that health care professio…

[WITHDRAWN: [Clinical strain of Proteus mirabilis able to produce a plasmid mediated ceftazidimase.]].

Resistance to third generation cephalosporins due to the acquisition and expression of extended spectrum beta-lactamases (ESBLs) enzymes among Gram negative bacteria continue to pose a challenge to infection management worldwide. The aim of this study was to determine the implication of the enzymatic mechanism and to describe the properties of ESBLs from a clinical strain of Proteus mirabilis NC4150 isolated in the intensive care unit at the Military Hospital in Tunisia during the period 2004. The isolate was identified, tested for antimicrobial susceptibility and analysed for presence of ESBL…

Encrustation of indwelling urethral catheters by Proteus mirabilis biofilms growing in human urine.

The encrustation and blockage of four types of urinary catheters was studied in a simple laboratory model of the catheterized bladder. Pooled human urine was supplied to the bladder chamber at 0.5 mL/min. The bladder urine was inoculated with a clinical strain of Proteus mirabilis that had been isolated from an encrusted catheter. The models were operated until the catheters blocked and atomic absorption spectrometry was used to assess the amounts of calcium and magnesium deposited on the catheters. Scanning electron microscopy was also used to locate and assess the degree of encrustation. All…

[Alkaline encrusted cystitis secondary to Proteus mirabilis infection in a HIV infected patient].

We described for the first time an alkaline encrusting cystitis in a patient with the acquired immune deficiency. This is an entity characterized by severe dysuria and long standing urinary frequency, alkaline urine and radiographically visible calcification of the urinary bladder. It has been observed in patients with permanent Foley catheter, persistent infections due to microorganisms with urolytic activity, urinary bilharziosis or tuberculosis, or malignant bladder tumors. None of these entities were described and the patient had all conditions for this syndrome.

Genomic and Biological Characterization of a Novel Proteus mirabilis Phage with Anti-Biofilm Activity.

The emergence of multidrug-resistant (MDR) Proteus mirabilis poses a significant threat in porcine farming and public health, highlighting the need for alternative biocontrol agents. This study aimed to isolate and characterize a lytic bacteriophage with therapeutic potential against MDR P. mirabilis. Using the clinical MDR P. mirabilis strain Pm 07 as host, a bacteriophage, vB_Pmc_P-07 (P-07), was successfully isolated from fecal and sewage samples via an enrichment protocol. Phage P-07 forms plaques surrounded by a distinct translucent "halo," suggesting the production of depolymerase. It ac…
