Two complementary experimental methods have been used to examine mitogen-induced transmembrane conductances in human B cells using the Daudi cell line as a model for human B cell activation. Spectrofluorometry was used to investigate mitogen-induced changes in [Ca++]i and transmembrane potential. We investigated the mechanisms that lead to polyclonal hypergammaglobulinemia in persons exposed to or infected with lymph-dwelling filarial parasites. Extracts of microfilariae of Brugia malayi induce the in vitro proliferation of lymphocytes from nonsensitized donors. The mitogenic factor(s) responsible for this effect is of low molecular weight (Mr) and acts on T4+ cells but not on T8+ T lymphocytes or on B cells. T4+ lymphocytes stimulated with filarial mitogen augment the in vitro production of immunoglobulins by autologous B lymphocytes.

From the crude snake venom of Agkistrodon rhodostoma two mitogenic principles could be detected: A potent T-lymphocyte stimulating anti-beta-D-galactose specific lectin with a molecular mass of about 18 kD and a very strong B-lymphocyte mitogenic factor. Whereas the lectin could be purified by affinity chromatography and characterized as a cell agglutinating, but not precipitating lectin, the highly active B-lymphocyte mitogen could be separated but not characterized. Both factors are much stronger than the mitogens known so far.

Changes in the activities of several lysosomal enzymes were studied during transformation of mouse spleen cells in vitro.
