Skeletal muscle protein metabolism in human heart failure.

PURPOSE OF REVIEW: This review considers evidence that the clinical condition of heart failure alters skeletal muscle protein synthesis and/or breakdown to promote skeletal muscle wasting and functional decrements that ultimately contribute to the symptomology of the disease. RECENT FINDINGS: Advanced HF is frequently accompanied by muscle atrophy and a cachectic phenotype. Protein metabolic derangements that promote this phenotype are understudied and poorly understood. Instead, most investigations have evaluated regulatory hormones/signaling pathways thought to be reflective of protein synt

Protein carbonylation and muscle function in COPD and other conditions.

Skeletal muscle, the most abundant tissue in mammals, is essential for any activity in life. Muscle dysfunction is a common systemic manifestation in highly prevalent conditions such as chronic obstructive pulmonary disease (COPD), cancer cachexia, and sepsis. It has a significant impact on exercise tolerance, thus worsening the patients' quality of life and survival. Among several factors, oxidative stress is a major player in the etiology of skeletal muscle dysfunction associated with those conditions. Whereas low levels of oxidants are absolutely required for normal cell adaptation, high le

Postnatal ontogeny of skeletal muscle protein synthesis in pigs.

The neonatal period is characterized by rapid growth and elevated rates of synthesis and accretion of skeletal muscle proteins. The fractional rate of muscle protein synthesis is very high at birth and declines rapidly with age. The elevated capacity for muscle protein synthesis in the neonatal pig is driven by the high ribosome content and, together with an increased efficiency of the translation process, promotes accelerated protein synthesis rates. Feeding profoundly stimulates muscle protein synthesis in neonatal pigs and the response decreases with age. The feeding-induced stimulation of …

Control of muscle protein kinetics by acid-base balance.

PURPOSE OF REVIEW: Abnormalities of acid-base balance accompany many pathological conditions. Acidosis is associated with several diseases such as chronic renal failure, diabetic ketosis, severe trauma and sepsis, and chronic obstructive respiratory disease, which are often associated with muscle wasting. There is evidence that acidosis can induce muscle protein catabolism and it could therefore be an important factor contributing to loss of muscle protein in these conditions. This review aims at outlining the effects of acid-base balance abnormalities on muscle protein metabolism, and will in

[Acute effects of peritoneal dialysis on muscle protein turnover].

BACKGROUND: Despite continuing glucose absorption and stimulation of insulin secretion, wasting is common in patients with chronic renal failure (CRF) treated with peritoneal dialysis. METHODS: To evaluate if peritoneal dialysis per se has any effect(s) on muscle protein turnover we employed the forearm perfusion method associated with the kinetics of 3H-phenylalanine in seventeen patients with CRF in the basal state and: a) during the systemic hyperinsulinemia associated with peritoneal dialysis (6 patients) (200-240 min); b) during locally-induced hyperinsulinemia, without systemic effects

Effects of diabetes and of injury on muscle protein in the mouse, and their interaction.

The effects of a 20% dorsal scald injury and of different severities of streptozotocin diabetes on hindquarter muscle protein have been studied in the mouse. Ten days after scald injury muscle protein contents were generally unaffected, whereas moderate diabetes (200 mg streptozotocin/kg body weight; plasma glucose concentrations 17--46 mmol/1; normal 11 mmol/l) led to net loss of muscle protein. Production of scald injury in the moderately diabetic mouse caused significant additional loss of muscle protein, especially from the extensor digitorum longus Milder diabetes (maximum plasma glucose

Protein Biomaterials with Muscle-like Water-Driven Actuation.

Unlike muscles, man-made shape-morphing biomaterials take much longer times to perform their actuation. Here we report a novel class of protein-based actuators that mimic muscle contraction through ethanol-induced fibril formation in bovine serum albumin (BSA) hydrogels, enabling reversible shape changes and fast, water-driven motion. These structural changes result in mechanical stiffening, enabling programmable and reversible shape changes, which take place over minutes to hours. At intermediate ethanol concentrations (40-80%), fibril formation dominates and contributes to shape retention, w

Calsequestrin, a key protein in striated muscle health and disease.

Calsequestrin (CASQ) is the most abundant Ca2+ binding protein localized in the sarcoplasmic reticulum (SR) of skeletal and cardiac muscle. The genome of vertebrates contains two genes, CASQ1 and CASQ2. CASQ1 and CASQ2 have a high level of homology, but show specific patterns of expression. Fast-twitch skeletal muscle fibers express only CASQ1, both CASQ1 and CASQ2 are present in slow-twitch skeletal muscle fibers, while CASQ2 is the only protein present in cardiomyocytes. Depending on the intraluminal SR Ca2+ levels, CASQ monomers assemble to form large polymers, which increase their Ca2+ bin

KBTBD13 is an actin-binding protein that modulates muscle kinetics.

The mechanisms that modulate the kinetics of muscle relaxation are critically important for muscle function. A prime example of the impact of impaired relaxation kinetics is nemaline myopathy caused by mutations in KBTBD13 (NEM6). In addition to weakness, NEM6 patients have slow muscle relaxation, compromising contractility and daily life activities. The role of KBTBD13 in muscle is unknown, and the pathomechanism underlying NEM6 is undetermined. A combination of transcranial magnetic stimulation-induced muscle relaxation, muscle fiber- and sarcomere-contractility assays, low-angle x-ray diffr

Mechanisms Underlying Muscle Protein Imbalance Induced by Alcohol.

Both acute intoxication and longer-term cumulative ingestion of alcohol negatively impact the metabolic phenotype of both skeletal and cardiac muscle, independent of overt protein calorie malnutrition, resulting in loss of skeletal muscle strength and cardiac contractility. In large part, these alcohol-induced changes are mediated by a decrease in protein synthesis that in turn is governed by impaired activity of a protein kinase, the mechanistic target of rapamycin (mTOR). Herein, we summarize recent advances in understanding mTOR signal transduction, similarities and differences between the

Regulation of muscle protein synthesis and the effects of catabolic states.

Protein synthesis and degradation are dynamically regulated processes that act in concert to control the accretion or loss of muscle mass. The present article focuses on the mechanisms involved in the impairment of protein synthesis that are associated with skeletal muscle atrophy. The vast majority of mechanisms known to regulate protein synthesis involve modulation of the initiation phase of mRNA translation, which comprises a series of reactions that result in the binding of initiator methionyl-tRNAi and mRNA to the 40S ribosomal subunit. The function of the proteins involved in both events

Lost in translation: regulation of skeletal muscle protein synthesis.

Skeletal muscle accounts for about 50% of the body's mass in higher vertebrates. Besides its obvious role in motor activity, it also can serve as a reservoir for amino acids during times of starvation, or even as a metabolic water supply for migratory birds' during long flights. An imbalance between anabolic and catabolic processes can lead to the loss of muscle mass and life-threatening cachexia or sarcopenia. This review summarizes the current state of knowledge about the regulation of protein translation in skeletal muscle; it also discusses the role of the mTOR pathway, as well as new find

Endotoxemia reduces skeletal muscle protein synthesis in neonates.

Protein synthesis in skeletal muscle is reduced by as much as 50% as early as 4 h after a septic challenge in adults. However, the effect of sepsis on muscle protein synthesis has not been determined in neonates, a highly anabolic population whose muscle protein synthesis rates are elevated and uniquely sensitive to insulin and amino acid stimulation. Neonatal piglets (n = 10/group) were infused for 8 h with endotoxin [lipopolysaccharide (LPS), 0 and 10 microg. kg(-1). h(-1)]. Plasma amino acid and glucose concentrations were kept at the fed level by infusion of dextrose and a balanced amino a

Regenerating human muscle fibres express GLUT3 protein.

The presence of the GLUT3 glucose transporter protein in human muscle cells is a matter of debate. The present study was designed to establish whether GLUT3 is expressed in mature human skeletal muscle fibres and, if so, whether its expression changes under different conditions, such as metabolic stress (obesity, obese non-insulin-dependent diabetes mellitus), hypertrophy (training), de- and reinnervation (amyotrophic lateral sclerosis) or regeneration (polymyositis). We used an immunohistochemical approach to detect and localise GLUT3. GLUT3 immunoreactivity was not detectable in adult skelet

A genetic approach to detect muscle protein interactions in vivo.

An organism is required to identify biologically relevant protein interactions. We propose Drosophila and its indirect flight muscles as a s … tative natural suppressors may help obscure the origin of FHC. The genetic procedure, used here for muscle proteins, could help diagnose FHC and other myopathies, and extend to proteins of clinical interest in other tissues, including the nervous and circulatory systems.

Patients with polymyositis show changes in muscle protein charges.

Polymyositis (PM) appears with indolent proximal muscle weakness and is an inflammatory disease with breakdown of muscle cells. In our study the protein charge concentrations of the contractile proteins in the A and I bands were determined, applying a microelectrode technique. Patients with PM show a lower protein charge concentration than healthy control subjects which may be caused by the breakdown and removal of the proteins in the contractile filaments. A tool to judge the state of the disease as well as an aid in diagnosis may have been found in this method.

Enzymatic capacity and protein synthesis in human muscle postoperatively.

The glycolytic and aerobic oxidative capacity in skeletal muscle was investigated to reveal if the decrease seen in muscle protein synthesis is accompanied by a fall in the enzymatic capacity to oxidize substrates. Six patients undergoing elective abdominal surgery were investigated by percutaneous muscle biopsies taken before surgery and on the first and third postoperative days. Protein synthesis as assessed by the polyribosome concentration was 40% lower on the third day postoperatively than before surgery (p less than 0.01). The glycolytic and oxidative capacity was evaluated by determinin

[Study of proteins of rat skeletal muscles in experimental rickets].

In dynamics of experimental rickets proteins of rat sceletal muscles were studied after and before physical exercises. Occurrence of rickets was estimated by following concentration of inorganic phosphorus and calcium in blood and by X-ray examination of knee joint. An increase in total amount of sarcoplasmic proteins was observed within the first week of development of the disease. After physical exercices the effect was noticed also within the second week of the development of rickets. Content of fractions of the sarcoplasmic proteins was similar in all animals and within all the periods stu

The double homeodomain protein DUX4c is associated with regenerating muscle fibers and RNA-binding proteins.

BACKGROUND: We have previously demonstrated that double homeobox 4 centromeric (DUX4C) encoded for a functional DUX4c protein upregulated in dystrophic skeletal muscles. Based on gain- and loss-of-function studies we have proposed DUX4c involvement in muscle regeneration. Here, we provide further evidence for such a role in skeletal muscles from patients affected with facioscapulohumeral muscular dystrophy (FSHD). METHODS: DUX4c was studied at RNA and protein levels in FSHD muscle cell cultures and biopsies. Its protein partners were co-purified and identified by mass spectrometry. Endogenous
